The bioengineered radiofrequency technology utilized has the potential to counter deleterious effects of insomnia and sleep deprivation on brain toxin cleansing and to even reduce the amount of sleep normally required for brain toxin cleansing. Both blood and brain levels of the toxins beta-amyloid and tau were measured at several months and 1-2 years into treatment. Radiofrequency wave treatment resulted in a substantial increase in toxin drainage from the brain and into blood of the Alzheimer's patients. "It appears that radiofrequency wave treatment increases the natural flow of fluid out of the brain through meningeal lymphatic vessels" explained Dr. Arendash. "Acting as sinks, these lymphatic vessels normally carry around 50% of toxins and metabolic wastes out of the brain during sleep, but their flow is decreased in normal aging and Alzheimer's Disease". In the treated Alzheimer's patients, cognitive impairment was stopped and even reversed by 1-hour radiofrequency wave treatments to the entire brain during wakefulness. Novartis will be gaining global rights to Kyorin Pharmaceutical’s pre-clinical inflammatory disease drug in a deal worth over $830m. The agreement gives Novartis an exclusive worldwide licence to develop, manufacture and commercialise KRP-M223, an MRGPRX2 antagonist for chronic spontaneous urticaria (CSU) and other allergic and inflammatory diseases involving mast cells. Approximately 40 million people globally are affected by CSU, a common skin condition characterised by the sudden appearance of raised, itchy and sometimes painful hives (wheals) that last for at least six weeks. Alongside its physical symptoms, CSU can significantly impact well-being, with many patients experiencing sleep deprivation, as well as psychiatric disorders such as anxiety and depression. Under the terms of the agreement, Kyorin will receive $55m upfront and will be eligible for milestone payments of up to $777.5m, as well as tiered royalties on net sales. Kyorin will also have the option to commercialise KRP-M223 in Japan, with Novartis retaining its own option to co-promote the product in this market. Novartis has made a number of recent deals to expand its pipeline across multiple therapeutic areas. A few weeks ago, the company announced that it will be expanding its late-stage cardiovascular pipeline by acquiring Anthos Therapeutics for approximately $3.1bn. The acquisition will give Novartis access to abelacimab, which is currently in phase 3 development for the prevention of stroke and systemic embolism in patients with atrial fibrillation, the most common form of abnormal heart rhythm. Shreeram Aradhye, president, development and chief medical officer, Novartis, said at the time of the February announcement: “Welcoming Anthos Therapeutics strengthens our focus in the cardiovascular space and complements our portfolio of… treatments, comprehensive clinical programmes, and strategic collaborations that help thousands of patients with heart disease around the world.” Novartis also revealed in December that it would be gaining exclusive global rights to PTC Therapeutics’ mid-stage Huntington’s disease programme in a deal worth up to $2.9bn, and entered into a worldwide licence and collaboration agreement worth $745m with Ratio Therapeutic in November to advance a somatostatin receptor 2-targeting radiotherapeutic candidate for cancer.

Xolair binds to immunoglobulin E in the blood, preventing it from triggering allergic reactions. In 2024, the injectable anti-inflammatory antibody netted about $1.6 billion in sales. Sanofi and Regeneron have worked for years to challenge Xolair’s CSU dominance with Dupixent (dupilumab). With Xolair\'s compound patent already expired and its formulation patent set to expire in November 2025, Novartis has been hard at work seeking a successor to the blockbuster drug. In August 2023, Novartis announced that oral BTK inhibitor remibrutinib improved symptom severity in patients with CSU in a pair of phase 3 trials. Later, the company said it planned to submit the antibody for approval sometime in 2025 (PDF). Potential Xolair competitors are also looming in biotech. For example, Third Harmonic Bio is going all in on its CSU asset, with plans to bring the KIT inhibitor into a phase 2 trial by the middle of 2025.  Lately, Novartis has been looking externally for preclinical prospects. The Swiss giant signed a potential $1 billion deal last month with newly emerged Light Horse Therapeutics to pursue multiple hard-to-drug cancer targets. And, at the end of last year, BioAge Labs inked a pact with Novartis worth up to $550 million to discover multiple therapeutic targets in age-related diseases.
