Effects of a Bacillus-based direct-fed microbial on digestibility, ruminal in situ nutrient disappearance, microbiome, and fermentation parameters in forage-fed beef cattle.

This experiment evaluated dry matter intake, ruminal in situ nutrient disappearance, total-tract apparent nutrient digestibility, microbiome, and ruminal fermentation parameters of crossbred beef cows offered forage-based diets and a Bacillus-based direct-fed microbial (DFM). Fifteen rumen-cannulated Angus × Hereford cows were used in a crossover design. Treatments consisted of forage-based diets plus 500 g of protein-mineral supplement mixed with 1) 3 g of a Bacillus-based DFM containing Bacillus licheniformis and B. subtilis (BAC; n = 15; 2.2 × 109 CFU of the mixture/g; Bovacillus, Novonesis, Lyngby, Denmark), or 2) without BAC (CON; n = 15). Each experimental period lasted 33 days with 30 days of wash-out between periods. Cows were fed daily with chopped grass-mixed hay [Idaho fescue (Festuca idahoensis) and Bluebunch wheatgrass (Pseudoroegneria spicata)]. From day 17 to 26, 5 g of titanium dioxide (TiO2) was dosed twice daily, at 12-h intervals, as an external marker of fecal output. From day 22 to 26, fecal spot samples were collected at 12-h intervals. On day 27, ruminal fluid was collected prior to feeding (0 h) and at 4, 8, 12, 16, and 20 h post-feeding to evaluate ruminal pH, volatile fatty acids (VFA) and NH3-N. From day 28 to 32, 4 g of grass-mixed hay was placed in Dacron bags and introduced through the cannulas for 0, 12, 24, 48, 72, and 96 h. A treatment effect was observed (P = 0.01) for DMI, which was greater for cows supplemented with BAC vs. CON. Cows supplemented with BAC tended (P > 0.06) to have greater DM and NDF disappearance vs. CON. A treatment effect was observed (P ≤ 0.02) for total-tract apparent DM and CP digestibility, which were greater for BAC vs. CON cows. Also, cows supplemented with BAC tended (P ≥ 0.06) to have a greater total-tract NDF and ADF digestibility vs. CON cohorts. Therefore, cows supplemented with BAC had greater (P < 0.01) amounts of DM, CP, NDF, and ADF digested vs. CON cows. A treatment × time effect was observed (P = 0.02) for ruminal pH, which increased at 16 and 20 h of collection for BAC vs. CON cows. Treatment effects were not (P ≥ 0.18) observed for VFA, N-NH3, and plasma concentrations of glucose, whereas plasma concentration of urea tended (P = 0.06) to be reduced for cows supplemented with BAC vs. CON. Hence, supplementation with a Bacillus-based DFM increased forage intake, the digestibility of DM and CP, and the amount of nutrients digested by rumen-cannulated cows receiving a forage-based diet.

Bacillus licheniformis as a protective agent in broiler chicken concurrently exposed to mycotoxins and necrotic enteritis: Toxicopathological and hematobiochemical perspectives

Mycotoxins negatively impact intestinal cell viability, leading to the depletion of beneficial bacteria and rendering birds susceptible to intestinal infections such as necrotic enteritis (NE). Furthermore, they impair the effective digestion and absorption of nutrients. This study aimed to evaluate the effects of Bacillus licheniformis supplementation on broiler birds exposed to mycotoxins and subsequent necrotic enteritis infection. A total of 280 one-day-old broiler chicks were divided into eight groups and subjected to B. licheniformis supplementation (1 × 10⁶ CFU/kg of feed) and mycotoxin exposure (aflatoxin and ochratoxin A, each at 150 ppb). Clostridium perfringens (3 × 10¹⁰ CFU/ml) was later administered to induce necrotic enteritis. This study evaluated body weight, feed intake, relative organ weights, hematological and serum biochemical parameters and performed histopathological examinations of liver, kidney and intestine. All the obtained data was statistically analyzed (P ≤ 0.05). The results demonstrated that B. licheniformis supplementation reduced the susceptibility to necrotic enteritis in broilers initially exposed to mycotoxins. Body weight and feed intake were significantly decreased in groups challenged with mycotoxins and necrotic enteritis, both individually and concurrently, compared to the control group. Relative weights of the liver, kidney and intestine were significantly higher in treatment groups. Hematological analysis revealed significantly lower erythrogram parameters (TEC, Hb, and PCV) in birds fed mycotoxin-contaminated feed, with or without necrotic enteritis. Hepatic and renal biomarkers were significantly elevated, and serum protein levels (total protein, albumin) were significantly lower. In contrast, birds supplemented with B. licheniformis and challenged with either mycotoxins or NE showed no significant differences in body weight, feed intake, erythrogram and leucogram compared to the control group. However, B. licheniformis did not mitigate these effects when supplemented in group with concurrent challenge of mycotoxins and NE, however, intensity of changes was reduced. In conclusion, B. licheniformis supplementation effectively alleviates the pathological changes induced by mycotoxins and necrotic enteritis when presented individually but is not sufficiently effective against the combined challenge of mycotoxins and necrotic enteritis.

Bacillus licheniformis suppresses Clostridium perfringens infection via modulating inflammatory response, antioxidant status, inflammasome activation and microbial homeostasis in broilers

Pathogenic bacteria infection, especially Clostridium perfringens (C. perfringens), markedly threatened the health of animals, and further caused huge economic loss. In this study, Bacillus licheniformis HJ0135 (BL) was used. Oxford cup bacteriostatic test and inhibitory rate test were conducted to evaluate the antibacterial ability of BL. Results showed the strongest inhibitory role of BL on C. perfringens (P < 0.05). Afterwards, 540 one-day-old yellow-feather broilers (32.7 ± 0.2 g) were randomly allocated into 3 groups, including CON group (basal diet), CP group (basal diet + 1 × 10⁹ CFU C. perfringens in gavage), and BL + CP group (basal diet containing 7.5 × 10⁶ CFU/g BL + 1 × 10⁹ CFU C. perfringens in gavage). At d 70, broilers in the CP and BL + CP groups were treated with C. perfringens by continuously oral administration for 5 d. The experiment lasted for 75 d. The serum, immune organs, jejunal mucosa, and cecal contents were collected for analysis. In vivo experiment showed that BL supplementation markedly improved (P < 0.05) BW, ADG, thymus index, serum immunoglobins and antioxidases, reduced feed conversion ratio (FCR) and serum pro-inflammatory cytokines of C. perfringens-infected broilers. Furthermore, the increased jejunal injury and levels of pro-inflammatory cytokines, decreased gene expressions of tight junction proteins in the jejunal mucosa were significantly alleviated (P < 0.05) by BL. More importantly, the activation of NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) inflammasome was inhibited (P < 0.05) by BL to further attenuate jejunal damage. Besides, BL supplementation markedly increased (P < 0.05) the cecal isobutyric acid and isovaleric acid. Microbial analysis showed that BL changed the composition and relative abundances of microbiota in the cecal contents (P < 0.05), especially the short chain fatty acids (SCFAs)-producing bacteria including Eubacterium_coprostanoligenes_group, Megamonas, Faecalibacterium, and Lactobacillus, which further protected against C. perfringens-induced jejunal inflammation in broilers. Our study laid a theoretical basis for the application of probiotics in lessening C. perfringens-related diseases in poultry farming.
