The prevalence of obesity in Mexico is 35.4%, and it is considered a risk factor for the development of diabetes, systemic arterial hypertension and dyslipidemia. Obesity due to the increased distribution and growth of adipose tissue creates a pro-inflammatory state induced by molecules secreted by the adipocytes themselves. Netrin-1 is a cell migration protein, which directs the recruitment, migration and entrapment of macrophages in different tissues, within adipose tissue the entrapment of macrophages induces the release of pro-inflammatory cytokines, which increase the secretion of pro-inflammatory adipokines. It has been found in high concentration in patients with obesity, insulin resistance and type 2 diabetes. Thymol is a phytopharmaceutical derived from oregano oil that has shown powerful anti-inflammatory and antioxidant effects through the stimulation of PPAR-gamma, adiponectin and inhibition of the NF-κB pathway mediated by the JNK pathway, pathways in which netrin-1 is involved in macrophage entrapment and recruitment.

Antibacterial and biodegradable PLA-based nanofibers loaded with natural phenolic monoterpenes for sustainable biomedical or food application

Antibacterial biodegradable PLA-based nanofibers loaded with phenolic monoterpenes - thymol, eugenol, carvacrol, and cinnamaldehyde, were prepared by electrospinning. The effect of bioactive molecule on the surface, thermal, morphological, and biological properties has been investigated about the potential pharmaceutical and food processing applications. Fiber diameters ranged from 320 nm for PLA fibrous mat up to 480 nm for PLA membrane with 6 % thymol. All the prepared active nanofibers exhibited hydrophobic surfaces with a slightly decreasing contact angle after the incorporation of phenols. Antimicrobial testing proved a strong efficiency against Escherichia coli and Staphylococcus aureus, depending on the specific type and content of the bioactive compound. A significant biofilm formation reduction of bioactive PLA nanofibers was revealed against tested microorganisms. Modification of PLA fibers with active molecules did not significantly affect the biodegradation kinetics in comparison to PLA samples with their absence. This study demonstrates the high potential of newly developed PLA-based/phenol nanofibrous membranes for use as antibacterial and antifouling systems applicable in wound dressings and food packaging.

Characterization of γ-CD-MOF-stabilized thymol Pickering emulsion films with enhanced preservation properties for Basa (Pangasius) fish

Maintaining the freshness of perishable foods, especially seafood, is a major challenge due to rapid spoilage. This study developed active packaging films by incorporating thymol-loaded Pickering emulsions stabilized by γ-cyclodextrin metal-organic framework (γ-CD-MOF) into polyvinyl alcohol (PVA). The films were characterized for their physical, mechanical, and barrier properties. FTIR, XRD, and thermal analysis confirmed successful incorporation and good stability. The films exhibited antimicrobial activity against foodborne bacteria. When applied to Basa fish fillets at 4 °C, the films reduced bacterial counts from 9.87 to 6.1 log CFU/g over 18 days and extended shelf life by 6 days. Chemical stability improvements were observed through changes in pH, TBARS, and TVB-N levels. This research offers a promising approach to addressing food spoilage challenges and provides a potential solution for extending the shelf life of highly perishable seafood products through advanced active packaging technology.

Biological evaluations and computational studies of newly synthesized thymol-based Schiff bases as anticancer, antimicrobial and antioxidant agents

Three new thymol-based molecules were synthesized and evaluated as anticancer, antimicrobial and antioxidant agents. Liver, colon, lung and prostate cancer cell lines were utilized in cytotoxicity tests. The results demonstrated that synthesized molecules had a cytotoxic effect against the screened cell lines. One of the molecules (4a) was found to have a higher efficacy towards the colon cancer cell line (DLD-1) with an IC₅₀ value of 12.39 µM and the other (4c) towards the prostate cancer cell line (PC3) with an IC₅₀ value of 7.67 µM than the positive control drug cisplatin. To assess the antimicrobial activity of molecules (4a-c), Gram-positive bacteria, Gram-negative bacteria and yeast were subjected to agar disc diffusion and broth microdilution assays. The investigation of antioxidant potential was conducted using the DPPH radical scavenging activity assay. While all compounds displayed strong cytotoxic and antioxidant properties, they exhibited only moderate antimicrobial activity. Molecular docking studies were performed on epidermal growth factor receptor (EGFR), vascular endothelial growth factor receptor 2 (VEGFR-2), focal adhesion kinase (FAK), B-Raf and phosphoinositide 3-kinase (PI3K). The binding energies and interactions obtained from the docking results of compounds (4a-c) supported the experimental results. Drug similarity rates and pharmacokinetic properties were analyzed with the absorption, distribution, metabolism and excretion (ADME) method. Geometric parameters such as chemical potential (µ), electrophilicity index (ω) and chemical softness (σ) of compounds (4a-c) were calculated using the 6-31*G basis set B3LYP method.Communicated by Ramaswamy H.
