Endophytic Fungus of Achyrocline satureioides: Molecular Identification, Chemical Characterization, and Cytotoxic Evaluation of its Metabolites in Human Melanoma cell line

Endophytic fungi are important sources of anticancer compounds. An endophytic fungus was isolated from the medicinal plant Achyrocline satureioides, and molecularly identified as Biscogniauxia sp. (family Xylariaceae) based on partial nucleotide sequences of the internal transcribed spacer genomic region (GenBank Accession No. ON257911). The chemical characterization and cytotoxic properties of secondary metabolites produced by Biscogniauxia sp. were evaluated in a human melanoma cell line (A375). The fungus was grown in potato-dextrose liquid medium for 25 days, and the extracted compounds were subjected to solid-phase fractionation to obtain the purified F_DCM fraction, for which the metabolites were elucidated via ultra-performance chromatography coupled to a mass spectrometer. In the present study, 17 secondary metabolites of Biscogniauxia sp., including nine polyketide derivatives, five terpenoids, and three isocoumarins, were putatively identified. This is the first study to report of the ability of Biscogniauxia sp. in the production of isocoumarin orthosporin; the terpenoids nigriterpene A and 10-xylariterpenoid; the polyketide derivatives daldinin C, 7'dechloro-5'-hydroxygriseofulvin, daldinone D, Sch-642305, curtachalasin A, cytochalasin E, epoxycytochalasins Z8, Z8 isomer, and Z17. Furthermore, this study has reported the biosynthesis of Sch-642305 by a Xylariaceae fungus for the first time. F_DCM significantly reduced the viability and proliferation of human melanoma cells at half-maximal inhibitory concentrations of 10.34 and 6.89 µg/mL, respectively, and induced late apoptosis/necrosis and cell cycle arrest in G2/M phase after 72 h of treatment. Given its ability to produce unique metabolites with promising cytotoxic effects, Biscogniauxia sp. of A. satureioides may be a reservoir of compounds with important therapeutic applications.

A catalytic hairpin assembly–based Förster resonance energy transfer sensor for ratiometric detection of ochratoxin A in food samples

Ochratoxin A (OTA) poses severe risks to the environment and human health, making the development of an accurate and sensitive analytical method for OTA detection essential. In this study, a catalytic hairpin assembly (CHA)-based Förster resonance energy transfer (FRET) aptasensor was developed to detect OTA using carbon quantum dots (CDs) and 6-carboxy-fluorescein (FAM) as dual signal readout. In the presence of OTA, the aptamer specifically interacted with OTA to release the helper DNA (HP), which could open the hairpin structure of FAM-labeled hairpin DNA 1 (H₁-FAM) modified on the surface of gold nanoparticles (AuNPs). CHA between H₁-FAM and hairpin H₂ labeled with CDs (H₂-CDs) can release HP for the next cycle, resulting in the occurrence of FRET with CDs as the energy donor and FAM as the energy acceptor. According to the ratio of F_CDs/F_FAM, the proposed aptasensor showed a wide linear range from 5.0 pg/mL to 3.0 ng/mL and a low detection limit of 1.5 pg/mL for OTA detection. Moreover, satisfactory results were obtained for OTA detection in rice, suggesting the potential application of this sensor in food safety analysis.

Partitioning behavior-oriented health risk assessment on internal organophosphorus flame retardants exposure

Though the partitioning behavior of organophosphorus flame retardants (OPFRs) has been recognized in vitro incubation assay, health risk assessment on those internal exposure with or without partitioning indexes in human blood is still unclear. In this study, nine commonly used OPFRs were quantified in 96 pairs of plasma and blood cell samples from Chinese volunteers. Non-carcinogenic and carcinogenic risk (CR) assessment building upon two distinct scenarios were conducted and compared. The dominant OPFRs in both plasma and blood cells were TBEP, TBP and TPHP. TCEP was the most enriched compound in plasma with F_plasma nearly to 1.0 (0.92), followed by TCPP, TBEP, TPHP, TBP and TEHP (from 0.61 to 0.76). The partitioning behavior of TCP in plasma was equivalent to blood cells with F_plasma at 0.50. When fully considered the F_plasma, the estimated average daily intake (DI) of ∑OPFRs (638.44 ng/kg BW/day) reached nearly 1.48-fold higher than the conventional calculation (dividing the concentration of plasma (C_plasma) by a factor of 2.0). Accordingly, we found the average hazard quotation (index) of TBP, TPHP and ∑OPFRs was underrated 1.50-fold when neglected the partitioning behaviors. Notably, the average CR of TCEP exceeded 10^-6 at the highest concentration (1.19 × 10^-6 ng/mL in plasma) only when the F_plasma was introduced. These data conjointly demonstrated that most of the DI levels and the corresponding risk index of OPFRs would be underestimated without factoring F_plasma into calculation, especially for those of low plasma partitioning. To our best knowledge, this study initially uncovered the gap between introducing F_plasma and dividing C_plasma by 2.0 during health risk assessment on internal OPFRs exposure.
