Spinal cord injury is an intractable traumatic injury. The most common hurdles faced during spinal cord injury are failure of axonal regrowth and reconnection to target sites. These also tend to be the most challenging issues in spinal cord injury. As spinal cord injury progresses to the chronic phase, lost motor and sensory functions are not recovered. Several reasons may be attributed to the failure of recovery from chronic spinal cord injury. These include factors that inhibit axonal growth such as activated astrocytes, chondroitin sulfate proteoglycan, myelin-associated proteins, inflammatory microglia, and fibroblasts that accumulate at lesion sites. Skeletal muscle atrophy due to denervation is another chronic and detrimental spinal cord injury–specific condition. Although several intervention strategies based on multiple outlooks have been attempted for treating spinal cord injury, few approaches have been successful. To treat chronic spinal cord injury, neural cells or tissue substitutes may need to be supplied in the cavity area to enable possible axonal growth. Additionally, stimulating axonal growth activity by extrinsic factors is extremely important and essential for maintaining the remaining host neurons and transplanted neurons. This review focuses on pharmacotherapeutic approaches using small compounds and proteins to enable axonal growth in chronic spinal cord injury. This review presents some of these candidates that have shown promising outcomes in basic research (in vivo animal studies) and clinical trials: AA-NgR(310)ecto-Fc (AXER-204), fasudil, phosphatase and tensin homolog protein antagonist peptide 4, chondroitinase ABC, intracellular sigma peptide, (-)-epigallocatechin gallate, matrine, acteoside, pyrvate kinase M2, diosgenin, granulocyte-colony stimulating factor, and fampridine-sustained release. Although the current situation suggests that drug-based therapies to recover function in chronic spinal cord injury are limited, potential candidates have been identified through basic research, and these candidates may be subjects of clinical studies in the future. Moreover, cocktail therapy comprising drugs with varied underlying mechanisms may be effective in treating the refractory status of chronic spinal cord injury.

Acaricidal activity and efficacy of oxymatrine-based biopesticide against peanut red spider mite

The acaricidal activity and efficacy of oxymatrine-based com. biopesticide on the peanut red spider mite Tetranychus ogmophallos Ferreira and Flechtmann, 1997 were assessed under laboratory, semi-field, and field conditions.In the laboratory, the oxymatrine-based biopesticide provided high acute toxicity for T. ogmophallos larvae, protonymphs, deutonymphs, and adult females, in a manner dependent on the concentration, developmental stage, and exposure time of mites to residues.Among the mite developmental stages, T. ogmophallos larvae were more susceptible to the biopesticide than adult females.The biopesticide also reduced fecundity and fertility, induced a low ovicidal effect, and provided a high repellent activity against T. ogmophallos.In semi-field, the biopesticide showed a short biol. persistence [mortality >80% up to 1 day after spraying (DAS)] compared to the synthetic acaricide fenpropathrin (mortality >80% up to 3 DAS).At 7 and 10 DAS, no significant mite mortality was observed on peanut plants treated with biopesticide compared to the control.However, the synthetic acaricide fenpropathrin caused mortality greater than 40% up to 10 DAS.In the field, the biopesticide showed high effectiveness in controlling the mite population, with a similar efficacy to the synthetic acaricide fenpropathrin.Therefore, the oxymatrine-based biopesticide constitutes an important management tool to be used in rotation with synthetic acaricides for population suppression of T. ogmophallos in conventional peanut crops or as an effective measure for controlling peanut red spider mite in organic peanut crops.

Matrine alleviates Staphylococcus aureus-induced acute lung injury in mice by inhibiting MLKL and NLRP3-mediated inflammatory activity

Acute lung injury (ALI) is a serious clinical condition with high incidence and mortality. The inflammatory response induced by gram-positive bacteria, especially Staphylococcus aureus (S. aureus), is a key factor contributing to ALI progression and other infectious diseases. Matrine, known for its diverse biological and pharmacological properties, has not been fully explored for its potential to prevent or treat S. aureus-induced ALI. Our study demonstrated that matrine exerts a protective effect against lung injury in mice infected with S. aureus. Specifically, matrine reduced pulmonary edema and decreased neutrophil infiltration in the infected lungs. Furthermore, matrine significantly reduced the expression of high-mobility group box protein 1 (HMGB1) and hyaluronic acid-binding protein 2 (HABP2) in the lungs of infected mice. Additionally, matrine modulated the production of inflammatory mediators, including tumor necrosis factor-α (TNF-α); interleukin-1β (IL-1β); regulated upon activation normal T cell expressed and secreted (RANTES) and Interleukin 10 (IL-10), in both infected lungs and macrophages, suggesting a protective role against tissue damage. Moreover, matrine influenced the secretion of proinflammatory cytokines and regulated the activation of the mitogen-activated protein kinase (MAPK) and nuclear factor kappa B (NF-κB) signaling pathways, along with the activation of Nod-like receptor pyrin domain-containing protein 3 (NLRP3) and Mixed-lineage kinase domain-like protein (MLKL) in macrophages. Notably, when MLKL, but not NLRP3, is deleted, the ability of matrine to regulate damage-associated proteins and prevent tissue injury is diminished. These findings suggest that matrine may be a promising therapeutic agent for the prevention and treatment of ALI.

ORION CORPORATION PRESS RELEASE 5 SEPTEMBER 2022 AT 8.00 EEST Orion recalls Devisol Salmiakki dietary supplement from consumers Orion is recalling all Devisol Salmiakki dietary supplement packages from consumers due to excessive levels of matrine in the product. Devisol Salmiakki has been on sale in Finland only starting from December 2021. Consumers are requested to return both opened and unopened Devisol Salmiakki products to the nearest pharmacy at the latest on 13 October 2022. The products will be reimbursed for the consumers. Empty packages or free samples will not be reimbursed. The recall does not concern Orion’s other Devisol products. The recall is due to levels of matrine in the licorice extract used in the Devisol Salmiakki product which have been found to exceed the maximum level allowed by the EU. Matrine occurs naturally in certain plants and is also used as a pesticide. Regarding matrine, there is no EU-level risk assessment and approval, and there is therefore no certainty about the safety of foodstuffs containing it. As a precaution, all batches of Devisol Salmiakki on the market are withdrawn. The daily recommended dose of Devisol Salmiakki is one tablet, in which case the exposure to matrine through this preparation is assumed to be low. Orion has not received any reports of adverse effects related to the Devisol Salmiakki product, and Orion is not aware of any consumer reports related to the safety of the Devisol Salmiakki product.
