A Two-part Trial Assessing the Effects of 7α-methyl-19-nortestosterone (MENT) on Blood Pressure in Normal Men: an Open-label Pilot Study Followed by a Randomized, Double-blind, Placebo-controlled Study

This is a two-part trial assessing the effects of MENT on blood pressure. The first part of the trial is an open-label pilot study to verify the MENT gel formulation delivers appropriate drug levels. Upon confirming MENT serum levels are within the required range, clinical trial material will be produced and labeled. Consequently, the second part of the trial, the main study, will proceed approximately two months after the conclusion of the pilot study. The main study, is a randomized, double-blind, placebo-controlled study.

Extraction and separation of pigments from Saccharina latissima using eutectic solvents

Harnessing the untapped potential of marine biomass has emerged as a pivotal strategy to address growing demands for natural bioactive compounds across many industries.Among these, Saccharina latissima, a prolific marine alga, is a rich source of fucoxanthin and chlorophylls - mols. of significant biotechnol. interest.This work used Saccharina latissima as a source of pigments to develop an integrated platform to promote the extraction and separation of chlorophyll and fucoxanthin using eutectic solvents (ES).Hydrophobic ES were investigated in the extraction of these pigments, and operational conditions were optimized.Among four hydrophobic ES screened, the Ment:LevA system, enhanced with 20 % (volume/volume) of water, exhibited the greatest potential, yielding an optimized extract rich in fucoxanthin with 137.2 ± 2.6 μg_fucoxanthin·g^-1_biomass.Addnl., a unique ES-ES biphasic system facilitated the selective partitioning of pigments: chlorophylls predominantly remained in the hydrophobic phase, while 95 % of fucoxanthin migrated to the hydrophilic.To further refine the quality of the extracted fucoxanthin, a subsequent purification step using water was implemented successfully, resulting in a concentrated pigment product.This work highlights ES as potential biocompatible solvents for the recovery of value-added compounds from marine biomass.

Neural modulation by nicotine aerosols and the role of flavor additives: insights from local field potentials in mice

Research on nicotine's neurobiological effects has rarely focused on aerosols, despite their primary role in tobacco product consumption. Here, we utilized in vivo electrophysiology to examine brain activity in mice exposed to nicotine aerosols, both alone and with flavor additives (citric acid and menthol). Local field potential (LFP) recordings from the nucleus accumbens (NAc), basolateral amygdala (BLA), ventral tegmental area (VTA), and ventral posteromedial nucleus (VPM) were analyzed under saline, nicotine, nicotine with citric acid(CA + NIC), and nicotine with menthol(MENT + NIC) conditions. Nicotine exposure significantly reduced power spectral density (PSD) in the NAc-Alpha, NAc-Beta, and BLA-Beta bands, unaffected by flavor additives. Coherence between key brain regions (e.g., VPM-VTA in Beta, VPM-BLA in Alpha) also decreased with nicotine but was restored with citric acid or menthol, suggesting their role in mitigating nicotine's disruptive effects on neural synchronization. Our findings show that LFPs can effectively capture nicotine's neural effects and highlight the modulatory role of flavor additives, offering new insights into nicotine exposure management and tobacco product design.

Fluornen-9-bisphenol increases Leydig cell proliferation but inhibits maturation in pubertal male rats via interacting with androgen receptor as an antagonist and estrogen receptor α as an agonist

Fluornen-9-bisphenol (BPFL) is used as one of the alternatives for bisphenol A. However, whether BPFL has deleterious effects to the male reproductive system and the underlying mechanism remain unknown. Here, we report the effects of BPFL on Leydig cell development in male rats in puberty. Male Sprague-Dawley (28 days old) rats were dosed with 0, 10, 100, 200 mg/kg/day BPFL via gavage for 28 days. BPFL significantly decreased serum testosterone levels at 200 mg/kg while increasing serum luteinizing hormone and follicle-stimulating hormone levels at 200 mg/kg. BPFL markedly increased Leydig cell number but down-regulated the expression of Cyp17a1 and its protein level in Leydig cells at 200 mg/kg. Further study showed that BPFL significantly increased Pcna and Cdk2 expression and increased Leydig cell proliferation at 200 mg/kg. BPFL treatment to immature Leydig cells isolated from 28-day-old male rats for 24 h significantly inhibited testosterone biosynthesis at 50 μM, which was completely reversed by the androgen receptor agonist 7α-methyl-nortestosterone and estrogen receptor α antagonist ICI 182,780. In conclusion, BPFL increases Leydig cell proliferation but inhibits its maturation in male rats in puberty by blocking androgen receptor and activating estrogen receptor α.
