Primary objectives: 1) To evaluate the safety and tolerability of SS-001 in patients with B-cell lymphoma. 2) To evaluate the maximum tolerated dose (MTD) and recommended dose (RP2D) of SS-001 for Phase II clinical trials. Secondary objectives: 1) To evaluate the human pharmacokinetic (PK) characteristics of single and multiple oral SS-001 in patients with B-cell lymphoma. 2) To evaluate the BTK target occupancy of SS-001 in peripheral blood mononuclear cells (PBMCs). 3) To preliminarily evaluate the efficacy of SS-001 in patients with B-cell lymphoma.

Preliminary assessment of microplastic in rhizosphere and non-rhizosphere region of mangrove at four locations along Karachi coast, Pakistan

Mangrove ecosystem faces significant threats from the various pollutants including microplastic (MPs). The aim of this study was to assess variations in MP distribution in mangrove sediments of rhizosphere (R) and non-rhizosphere (NR) regions. A total of 14,960 MP particles were identified from Sandspit backwater (SS-1 & SS-2) and Creek areas (PQ & KC). Notably, the NR showed higher MP counts (7848) compared to the R region (7112). Analysis revealed variations in MP types, with beads being predominant in both R and NR, followed by film, fiber, and fragments. KC exhibited highest MP contamination, followed by PQ, SS-2, and SS-1. Fourier-transform infrared (FTIR) analysis confirmed the presence of polyethylene terephthalate and polyethylene in sediments samples. This first detailed report on MP in mangrove sediments and other limited studies from Pakistan establishes the widespread distribution of MPs in the coastal area and provide a baseline for further elaboration in future.

Chinese herbal medicine SS-1 inhibits T cell activation and abrogates TH responses in Sjögren's syndrome

BACKGROUND/PURPOSE:

Sjögren's syndrome (SS) is an autoimmune disease and its conventional treatment has exhibited limited therapeutic efficacy. Traditional Chinese medicine has been demonstrated to ameliorate the sicca symptoms of SS by decreasing the level of T_H1 and T_H2 cytokines and increasing salivary flow rate. A newly designed traditional Chinese medicine, SS-1, showed improved efficacy in alleviating the dryness symptoms of SS patients in the National Taiwan SS cohort investigation. Here, we investigated the effect of SS-1 on T cell responses.

METHODS:

SS-1 was authenticated and its major compounds were verified by high-performance liquid chromatography. We examined the effects of SS-1 on the activation and T_H1, T_H2, and T_H17 polarization of murine T cells. We also determined the level of T_H1, T_H2, and T_H17 cytokine RNA in peripheral blood mononuclear cells of SS patients before and after SS-1 treatment.

RESULTS:

SS-1 treatment inhibits the activation and T_H1, T_H2, and IL-17A⁺IFNγ⁺ T_H polarization of murine T cells. SS-1 treatment also significantly reduces IFN-γ, IL-4, and IL-13 expression, and moderately reduces IL-17A expression in peripheral blood mononuclear cells of SS patients.

CONCLUSION:

Our results suggest that SS-1 inhibits T cell activation and diminishes T_H1, T_H2, and IL-17⁺IFN-γ⁺ T_H responses in SS patients.

Artemisia princeps Pampanini, which is called Ssajuarissuk in Korean (SS-1), was fermented with lactic acid bacteria (LAB) and their passive cutaneous anaphylaxis reaction-inhibitory activity was investigated. Of these fermented agents, SS-1 extract fermented with Bifidobacterium infantis K-525 (F-SS-1) most effectively inhibited the release of P-hexosamindase from RBL-2H3 cells induced IgE. In IgE-induced RBL-2H3 cells, F-SS-1 inhibited proinflammatory cytokines IL-6 and TNF-alpha mRNA expression. Oral administration of SS-1 and F-SS-1 to mice inhibited passive cutaneous anaphylaxis (PCA) reaction induced by IgE and scratching behaviors induced by compound 48/80. The inhibitory activity of F-SS-1 against scratching behaviors was more effective than that of SS-1. These findings suggest that the fermentation of SS-1 with LAB can increase its antiallergic activity.
