Currently, there is concern about food products to which new psychoactive substances (NPS) have been added illegally. However, rapid, high-sensitivity, and high-throughput screening methods for such harmful substances have not been reported yet. In this study, a high-throughput non-targeted screening strategy was developed based on UHPLC-Q/Orbitrap. First, optimized pretreatment methods for typical high-risk matrices were established. Second, an accurate, efficient, and scalable high resolution mass spectrometry database containing 206 NPS and related compounds was constructed. The fragmentation pathways of three different types of fentanyl-related compounds were then systematically investigated to enhance compound identification. Additionally, a non-targeted screening strategy was developed using warning ions derived from 24 fentanyl-related compounds, streamlining the identification of NPS. This strategy was applied to 100 food samples with possible illegal additives and demonstrated its effectiveness. In summary, this study provides robust technical support for regulatory authorities in combating the illegal addition of food products.

Passive sampling of contaminants of emerging concern in a Caribbean urban estuary of Puerto Rico

Contaminants of emerging concern (CECs) have not been extensively studied in Caribbean estuaries, particularly in Puerto Rico. Polar organic chemical integrative samplers were deployed in surface waters at five sampling sites in the San Juan Bay Estuary to investigate the occurrence and distribution of 137 CECs, resulting in the detection of 64 chemicals (51 pharmaceuticals and personal care products "PPCPs", 12 hormones plus sucralose) at least in one sampling site, and with 35 chemicals obtaining a 100 % detection frequency. Wastewater intrusion was implicated in the presence of CECs based on the detection of sucralose and other wastewater tracers. Condado Lagoon was the most polluted site impacted by untreated wastewater, showing the highest levels of 30 of the 64 detected CECs, and the largest proportion of detected CECs, particularly PPCPs. Levels of estrone and 17β-estradiol represent a high potential risk for adverse effects on aquatic organisms in this Caribbean urban estuary.

Predicting the skin sensitizing potential of pesticides using Pred-skin 3.0–A web-based prediction tool

Pesticide usage is increasing due to growing needs of agriculture and horticulture. Occupational dermal exposure to pesticides at an acute or chronic low-level could result in contact dermatitis and various skin cancers. Hence, detailed understanding about the Adverse Outcome Pathways (AOP) or Chemical Sensitization Pathway (CSP) behind pesticides belonging to various categories has to be investigated. Animal models of skin sensitization testing at times either over or under predict the human responses due to species-to-species variability. This necessitates the need for prediction tools for skin sensitizing potential of various chemicals. Pred-skin 3.0, is a consensus Naïve Bayes model-based prediction tool which utilizes various human, LLNA, and non-animal data to predict skin sensitization. Although, this tool was never used for predicting skin sensitizing potential of pesticides. Henceforth, the current study aims to test the applicability of this prediction tool in predicting skin sensitizing potential of 96 pesticides belonging to three Major classes. The Bayesian outcome of Pred Skin prediction tool provided a good concordance of 72.72 % with the existing animal skin sensitizing data as well as 63.46 % with the non-sensitizer data.
