Background:Irisquinone, an important compound extracted from Semen Irisis, has been used clinically as a radiotherapy sensitizer for lung, oesophageal, head and neck, breast and leukemia cancers. However, the mechanism by which it acts against cancer is still unclear.Objective:The present study aims to investigate the anti-tumor activity and mechanism of Irisquinone.Methods:The effect of Irisquinone on cell viability and proliferation was evaluated using the CCK-8 assay. Fluorescence probe (Fast-TRFS) and DTNB assay were used to observe the inhibitory effect of Irisquinone on both intracellular and extracellular thioredoxin reductase (TrxR). The level of reactive oxygen species (ROS) in tumor cells was assessed using the DCFH-DA probe. Annexin V-FITC/PI, staining and microscopy experiments, were used to examine the apoptosis and pyroptosis. Western blotting analyses confirmed that Irisquinone induced apoptosis and pyroptosis in cancer cells by inhibiting TrxR to increase ROS generationResults:Our research has shown that Irisquinone has anti-proliferative effects on several cancer cell lines while having low toxicity to normal cells. The amount of ROS induced by inhibition of TrxR activated the BAX (proapoptotic protein) and caspase-1(the pro-pyroptotic protein) to induce apoptosis and pyroptosis.Conclusion:Irisquinone showed anticancer activity through inhibiting TrxR. These results suggested that Irisquinone will be developed to be an anti-tumor drug possibility.

Development of an Efficient Synthetic Process for Irisquinone

AbstractIrisquinone is a tumor radiotherapy sensitizer and has been found to have broad-spectrum antitumor activity in recent years. The current acquisition method of extracting and purifying from semen irisis has greatly limited its wide application and activity study deeply. In this work an efficient route for the synthesis of the irisquinone was investigated to solve the source of it. The target compound was synthesized by 5-step reactions to Wittig reaction, reduction, oxidation, Wittig reaction, and oxidation using 3,5-dimethoxycarboxaldehyde as the starting material with an overall yield of 48%. The key factors such as the ratio of raw materials, temperatures, solvents, reaction times, and types of base for the main reactions were optimized. In addition, the deprotection and reduction were completed with Pd/C catalytic simultaneously when compound 2 was synthesized from compound 1. In the last reaction, the 3,5-dimethoxybenzene moiety of compound 4 was directly oxidized to 6-methoxy-1,4-benzoquinone by K3[Fe(CN)6]/H2O2 without the need to selectively remove the methyl protecting group, which were the innovative points in the experimental route design of the irisquinone synthesis. This work has opened new perspectives for the artificial synthesis and the development of irisquinone.

Physicochemical parameters combined flash GC e-nose and NIR for quality and volatile characterization of Zhenjiang vinegar with different aging time

In order to clarify the scientific connotation of the processing adjuvant vinegar of "the aging the better", the physicochem. indexes, antioxidant activity in vitro, odor characteristics and NIR spectrum characteristics of Zhenjiang vinegar with different aging time were comprehensively studied, and the compositional characterization by UPLC-MS.The results showed that the physicochem. quality of Zhenjiang vinegar increased rapidly during the 2-3 years, entered a rising stable period during the 5-8 years and entered a sudden increase period during the 8-10 years of fermentationOverall, it was in line with the trend that "the aging the better".The study further characterized the odor changes of different aging time samples, established a fast and non-damage SVM discriminant model based on NIR spectral characteristics (accuracy 100 %), and 753 components were characterized by UPLC-MS, in order to provide a reference for the quality evaluation and standard improvement of Zhenjiang vinegar and processing adjuvant.
