Work Physiological-Biomechanical Analysis of a Passive Exoskeleton to Support Occupational Lifting and Flexing Processes

BACKGROUND Industrial tasks that are characterized by high loads, a high repetition rate, and/or awkward body postures, put employees at higher risk to develop work-related musculoskeletal disorders (WRMSD), especially low back pain. To counteract the prevalence of WRMSD, human-robot interaction could improve the power of a person and reduce the physical strain. For the lower back, a reduction of spinal loading could be helpful. The passive upper-extremity exoskeleton Laevo® is developed to support physically heavy work: it supports the back during bending and should, consequently, result in less low back pain (Laevo®, the Netherlands).
OBJECTIVES The primary aim of this study is to assess to what extent wearing the exoskeleton changes:
* muscular activity of the erector spinae and biceps femoris muscles;
* knee compression force;
* posture of the upper and lower spine, trunk, hips and knees; ...in different tasks (static vs. dynamic), different trunk postures (trunk flexion vs. trunk flexion and rotation) and different knee postures (straight vs. stooped).
Secondary aims of this study are to assess to what extent wearing the exoskeleton changes:
* muscular activity of the trapezius descendens, rectus abdominis, vastus medialis and gastrocnemius medialis;
* perceived discomfort;
* heart rate;
* internal loadings on the spine, using a lumbar spine model;
* the performance of subjects during functional activities (e.g., stair climbing) when wearing the exoskeleton (either turned on or off); ...in different tasks (static vs. dynamic), different trunk postures (trunk flexion vs. trunk flexion and rotation), different knee postures (stoop vs. squat), and different static holding positions(0° vs. 30° vs. 60°) with different weights (0kg vs. 8kg vs. 16kg).

Egg production, nutrient utilization and metabolism in Lohmann LSL lite hens fed corn and soybean meal-based diet supplemented with multiple doses of single ß-mannanase from 63 to 83 weeks of age

The impact of supplementing multiple doses of single ß-mannanase (BM) in corn and soybean meal-based diets was investigated in Lohmann LSL lite hens. Corn and soybean meal-based diets were formulated to meet or exceed nutrient specifications and supplemented with BM at 0, 200, 400, 800 and 1,200 TMU/kg of feed. All diets contained titanium dioxide indigestible marker and were prepared in mash form. Three hundred and twenty hens at 63 weeks of age (woa) were placed in enriched cages (4 birds/cage). Diets were allocated to give 16 replicate cages/diet. Birds had free access to water and feed to 83 woa. Hen day egg production (HDEP), feed intake (FI), FCR, egg weight (EW), egg mass (EM), eggshell breaking strength (ESBS) and thickness (EST), Haugh unit (HU) and yolk color were reported at 4 weeks periods. Apparent retention (AR) of components and metabolizable energy (AME) were determined at 4th and 17th week of the experiment. Plasma metabolites, ceca digesta short chain fatty acids (SCFA), body and organ weight were determined at the end of the experiment. Interactions (P ≤ 0.05) between BM and period/sampling time point was observed for HDEP, EW, EM, FCR and AR of neutral detergent fiber (ARNDF) such that there were period dependent linear and non-linear responses to BM. Independent of the period, BM increased HDEP (P = 0.025, cubically), EW (P ≤ 0.018, linearly and quartically), EM (P < 0.01, quartically). The FCR reduced linearly (P = 0.032) in response to BM. Birds receiving BM800 had higher ARNDF than birds receiving BM0 at 4th wk sampling. The ESBS increased (P = 0.040, cubically) whereas HU decreased (P = 0.048, quadratically) in response to BM. The AR of DM, CP and AME, plasma glucose, cholesterol, lactate dehydrogenase, lipase Ca, P, and abdominal fat increased (P < 0.05) linearly in response to BM. There were no BM effects (P > 0.05) on SCFA. In conclusion, BM improved HDEP; egg quality and nutrient retention dose dependently. Moreover, plasma metabolites indicated positive effects on indices of carbohydrate and lipid metabolism.

Connecting academia and industry: Advancing the use of entomopathogenic nematodes to tackle emerging challenges and opportunities in modern agriculture

The collaboration among academia, industry, and government is crucial for scientific progress and innovation. Academia generates fundamental knowledge, which industry translates into sound applications, considering government policies. This partnership is vital to feed progress and constant development and address global challenges like climate change and food security. Sustainable crop protection is a topical theme, with efforts to reduce pesticide reliance and promote alternatives to chemical pest management, and it continues to grow and be accepted worldwide. In this respect, biopesticides such as entomopathogenic nematodes (EPNs) offer a promising solution for pest and disease management as an eco-friendly alternative. However, EPN continue to face adoption barriers due to regulatory, commercialization and basic and applied knowledge gaps. Thus, stronger collaborations are needed to unlock their full potential, as highlighted in the 2024 congress organized in La Rioja (Spain) to commemorate the 100 years since the discovery of the first EPN. This review examines the gap between academia and industry, suggesting strategies to bridge it, thereby promoting the advancement of EPN in 21st-century agriculture. Despite decades of research demonstrating their efficacy, EPN commercialization remains limited by production, formulation, and application challenges. Universities and government research agencies have driven fundamental innovation through the discovery of new EPN-bacteria partnerships in new regions of the world, which are helping us understand their distribution and habitant adaptations which are required for their registration and in establishing global regulations. Research conducted both in academia and the private sector (both big and small start-up companies) has and continues to play a key role in the characterization of EPN and in assessing their performance for their subsequent formulation, product optimization, and commercialization. These are fundamental steps to reach the ultimate goal, which is to provide growers with reliable products that are cost-effective and sustainable. In this review, we summarize key findings that have led to the commercialization and application of EPN, spanning from the characterization of EPN and their symbiotic bacteria to production, formulation, and the requirements for their registration. We also highlight critical knowledge gaps and opportunities for collaboration between academia, government agencies, and industry. Strengthening these partnerships will drive EPN adoption in agriculture, establishing them as a desirable biocontrol solution.

Discovery and biological characterization of a novel mesoionic insecticide fenmezoditiaz

Abstract:

BACKGROUND:

Many piercing‐sucking insects have developed resistance or cross‐resistance to many insecticides targeting insect neural nicotinic acetylcholine receptor (nAChR). Here we are aiming to present the discovery of a novel mesoionic insecticide, fenmezoditiaz, by BASF through structure‐based drug design (SBDD) approaches. It has recently been added to the Insecticide Resistance Action Committee mode of classification (IRAC 4E). It is being developed for plant protection against piercing‐sucking pests, especially rice hopper complex.

RESULTS:

The soluble acetylcholine binding protein (AChBP) from the sea slug Aplysia californica was modified using site‐directed mutagenesis and based on putative aphid nAChR subunit sequences to create soluble insect‐like AChBPs. Among them, insect‐like β1 AChBP and native aphid membrane preparation showed the highest correlated biochemical affinity toward structurally diverse ligands. This mutant AChBP was used to understand how insect nAChRs structurally interact with mesoionics, which was then utilized to design novel mesoionics including fenmezoditiaz. It is an excellent systemic insecticide with diverse application methods and has a broad insecticidal spectrum, especially against piercing/sucking insects. It lacks cross‐resistance for neonicotinoid resistant plant hoppers. Field‐collected brown plant hopper populations from Asian countries showed high susceptibility.

CONCLUSIONS:

Fenmezoditiaz is a systemic insecticide with a broad spectrum, lack of cross‐resistance and it could be an additional tool for integrated pest management and insecticide resistance management, especially for the rice hopper complex. © 2024 The Authors. (NASDAQ:MDGL), a clinical-stage biopharmaceutical company pursuing novel therapeutics for nonalcoholic steatohepatitis (NASH), today announced three new members of its leadership team: Mark Barrett joins Madrigal as Chief Business Officer, Clint Wallace joins as Chief Human Resources Officer and Tina Ventura joins as Chief Investor Relations Officer. “I am delighted to welcome Mark, Clint and Tina to the team,” said Bill Sibold, Chief Executive Officer of Madrigal. “As we prepare for a first-to-market launch of resmetirom in NASH with significant fibrosis, we are also building a foundation for enduring leadership. These new leaders are not only experts in their respective areas but have experience scaling teams and organizations. They will each play a pivotal role in Madrigal’s next phase of growth.” Mark Barrett brings more than 25 years of biopharmaceutical leadership and industry experience to Madrigal, with a track record of success completing a wide range of partnerships and acquisitions to build commercial portfolios and R&D pipelines across therapeutic areas. He was most recently co-founder, Director, and CEO of Lassen Therapeutics, a biotechnology company focused on discovery and development of novel antibody therapeutics in immunological diseases and cancer. Mr. Barrett has also served as Entrepreneur in Residence at Frazier Healthcare Partners, where he focused on starting and financing new biotech companies. Prior to Lassen, he held leadership positions in Strategy and Business Development at Johnson & Johnson and Sanofi. He holds an M.B.A from the University of California at Berkeley and a B.S. degree in Biology from Duke University. Clint Wallace joins Madrigal from Kenvue, where he served as Senior Vice President Human Resources, Global Functions and Organizations and was responsible for organizational design and talent succession strategy. Prior to Kenvue, he was the Senior Vice President Human Resources, North America, Sanofi, from 2016-2022, serving as a member of the Sanofi Global HR Leadership Team. Prior to Sanofi, Mr. Wallace served in HR leadership positions at BASF Corporation, GE Healthcare and Pratt & Whitney. He was a Commissioned Officer in the U.S. Air Force and retired in June 2008 after 21 years of service. He holds an M.B.A. from Rensselaer Polytechnic Institute and a B.S. degree in Manufacturing Engineering from Central Connecticut State University. Tina Ventura brings nearly 25 years of experience in global health care investor relations and communications to Madrigal. Before joining Madrigal, she was Chief Investor Relations Officer at Horizon Therapeutics (acquired by Amgen) where she led the company's investor relations efforts to shape its investment identity as a rare disease-focused biotech, build and develop its investor base and drive shareholder value creation.
