Introduction

Associates with and regulates the activity of the sodium/potassium-transporting ATPase (NKA) which catalyzes the hydrolysis of ATP coupled with the exchange of Na(+) and K(+) ions across the plasma membrane (By similarity). May increase NKA activity by increasing the apparent affinity for Na(+) (PubMed:18263667). Involved in down-regulation of E-cadherin which results in reduced cell adhesion. Promotes metastasis (PubMed:11756660).

FXYD5 regulates gastric cancer cell metastasis and drug resistance by EMT modulation

Gastric cancer (GC) is the third leading cause of cancer-related mortality and the fourth most prevalent malignancy globally. The high prevalence and mortality rates of GC are attributed to various factors, including drug resistance, local recurrence, and distant metastases. There is an urgent need to identify novel therapeutic targets for GC. Patient-derived xenografts (PDX) model offers unique advantages in maintaining the molecular heterogeneity and tumor microenvironment of primary tumors, offering significant advantages for the screening of personalized therapeutic targets. In this study, we established GC PDX models with metastatic potential through orthotopic transplantation and investigated the different gene expressions between primary and metastatic tumors using PCR-array analysis. We found that the metastatic tumors displayed elevated levels of FXYD domain-containing ion transport regulator 5 (FXYD5) compared to the primary tumors. Additionally, reducing FXYD5 expression was found to inhibit the invasion, metastasis, and proliferation of GC cells. Silencing FXYD5 also reversed the resistance of GC cells to doxorubicin and vincristine by modulating the epithelial-mesenchymal transition (EMT) process and the expression of multidrug resistance protein 2. This study indicates that FXYD5 is involved in GC progression and regulates chemotherapy resistance, suggesting its potential as a novel therapeutic target for the clinical treatment of GC.

Genetic parameters and identification of genomic regions and candidate genes associated with vaginal discharge score in Holstein cattle based on genomic and transcriptomic analyses

A healthy uterine environment is essential for establishing and maintaining pregnancy and normal embryo development after insemination. In this context, the primary objectives of this study were to assess the genetic background of vaginal discharge score (VDS) traits during the voluntary waiting period in Holstein cows and to identify genomic regions and candidate genes influencing postpartum uterine health based on the integration of phenotypic, genomic, and transcriptomic data sets. Genetic parameters of 5 VDS traits defined according to lactation stage (VDS1, VDS2, VDS3, VDS4, and VDS5) were estimated based on VDS records from 64,241 Holstein cows that calved between 2019 and 2023 and genomic information from 2,489 cows. Genome-wide association studies (GWAS) were performed aiming to identify genomic regions associated with VDS traits. Differentially expressed genes and modular genes were obtained through RNA-seq data of uterine secretion from 6 healthy and 6 diseased cows. The VDS traits had low heritability estimates ranging from 0.006 ± 0.002 to 0.081 ± 0.011. Among the VDS traits, there were relatively strong genetic correlations between VDS1 (0-14 DIM) and metritis (0.678-0.763) as well as VDS3 (29-55 DIM) and endometritis (0.579-0.628). A total of 190 genes harboring 32 significant SNPs were identified as candidate genes regulating VDS in primiparous cows. The candidate genes identified were significantly enriched for pathways involved in cytokine-cytokine receptor interaction, MAPK signaling, and oxytocin signaling. Based on RNA-seq data of uterine secretions, 2,803 differentially expressed genes and 3,570 modular genes were identified. Furthermore, 7 genes were identified based on GWAS, differential gene expression, and weighted gene co-expression network analysis. The genes VSTM1, IL10RA, FXYD5, C2CD5, CETN4, ALS2CL, and PBX1 were considered to be the most promising candidate genes influencing postpartum uterine health in Holstein cows. This study provides novel insights on the genetic background of postpartum uterine health in Holstein cows. The results obtained can contribute to further refinements of selection indexes for improving uterine health and fertility in dairy cattle.

A Novel Platinum-Resistance-related Gene Signature in Ovarian Cancer: Identification and Patient-derived Organoids Verification

Background:Platinum-based chemotherapy resistance is one of the main contributors to the mortality of Ovarian Cancer (OC). It is believed that sensitive biomarkers for identifying the population that is platinum-resistant are urgently needed. This study aims to develop a platinum-resistance gene-based signature to predict OC patients' responses to platinum drugs as well as survival outcomes.Methods:A platinum-resistance-related gene model was built by bioinformatics analysis. Then, its predictive power was internally validated. Continually, a nomogram was constructed to confirm the model's predictive ability. Afterward, GSEA was used to explore our model's potential functions. The ESTIMATE, CIBERSORT, TIMER, and ssGSEA were applied to estimate immune conditions. Then, somatic mutation and drug sensitivity were also analyzed. Finally, to gain insights into the roles of targeted genes in drug sensitivity, patient-derived tumor organoids (PDOs) validation was performed.Results:Nine platinum-resistance-related genes, including SLC22A2, TAP1, PC, MCM3, GTF2H2, FXYD5, SUPT6H, IGKC, and MATN2, were anchored to build the predictive model, which was well internally validated. Subsequently, GSEA unveiled that our model genes enriched in the Hedgehog signaling pathway. The predictive signature was associated with immune checkpoint inhibitors such as PD-1, PD-L1, and CTLA4, guiding immunotherapy applications for OC patients. Drugs such as dasatinib, midostaurin, metformin, MK-2206, and mitomycin C might also benefit OC patients with different risk scores. PDOs showed patients with high-risk scores were more resistant to cisplatin than patients with low-risk scores.Conclusion:The platinum-resistance-related gene signature (SLC22A2, TAP1, PC, MCM3, GTF2H2, FXYD5, SUPT6H, IGKC, and MATN2) is valuable for prognosis prediction and guidance of treatment choices for OC patients
