Introduction

Component of the NuA4 histone acetyltransferase (HAT) complex, a multiprotein complex involved in transcriptional activation of select genes principally by acetylation of nucleosomal histones H4 and H2A (PubMed:14966270). The NuA4 complex plays a direct role in repair of DNA double-strand breaks (DSBs) by promoting homologous recombination (HR) (PubMed:27153538). The NuA4 complex is also required for spermatid development by promoting acetylation of histones: histone acetylation is required for histone replacement during the transition from round to elongating spermatids (By similarity). In the NuA4 complex, EPC1 is required to recruit MBTD1 into the complex (PubMed:32209463).

A Di-aptamer-functionalized scaffold promotes bone regeneration by facilitating the selective retention of MSCs and EPCs and then promoting crosstalk between osteogenesis and angiogenesis

The crosstalk between osteogenesis and angiogenesis plays an important role in promoting the formation of a microenvironment that supports bone regeneration. This suggests that the retention of endogenous osteogenic and angiogenic cells in the bone defect area can promote tissue-engineered bone (TEB) osteogenesis and cell-cell interactions. In this study, a Di-Aptamer-functionalized HA/β-TCP (Di-Aptamer-H/T) scaffold was prepared by sequential modification of APTES and sulfo-SMCC and connected with aptamer HM69 and EPC1. We confirmed that aptamers HM69 and EPC1 can specifically identify mesenchymal stem cells (MSCs) and endothelial progenitor cells (EPCs), respectively. This process triggers the expression of adhesion-related genes in these cells and allows these cells to selectively stay coupled to Di-Aptamer-H/T. The osteogenic differentiation ability of MSCs treated with Di-Aptamer-H/T in vitro was significantly increased. Similarly, the ability of Di-Aptamer-H/T-treated EPCs to form blood vessels was also enhanced. Notably, the osteogenic and angiogenic abilities of cocultured MSCs and EPCs treated with the Di-Aptamer-H/T scaffold were significantly better than those of cells cultured individually. In vivo, the results of micro-CT angiography, H&E staining, Masson's staining and histochemical staining further confirmed that Di-Aptamer-H/T formed new bones and vessels more readily than those treated with a single aptamer linked to HA/β-TCP or with HA/β-TCP alone. In brief, our study demonstrated that crosstalk between osteogenesis and angiogenesis is promoted by the Di-Aptamer-H/T scaffold, which serves as a potential treatment strategy for bone defects and can improve outcomes.

Ossifying fibromyxoid tumours with lipomatous and cartilaginous differentiation: A diagnostic pitfall

AimsOssifying fibromyxoid tumour is a rare mesenchymal neoplasm predominantly affecting adults characterised by a multinodular growth pattern and the presence of a fibrous pseudocapsule with areas of ossification. Prompted by the recognition of a non‐ossifying ossifying fibromyxoid tumour with lipomatous differentiation which caused diagnostic difficulty, we sought to further explore cases of ossifying fibromyxoid tumour with non‐osseous heterologous elements.Methods and ResultsA search of our institutional and consultation archives revealed three additional cases that demonstrated lipomatous components and two cases with cartilaginous differentiation. RNA‐sequencing revealed fusions involving PHF1 (n = 4) or EPC1 (n = 1) in all (five of five) cases tested, including EPC1::PHC1 and JAZF1::PHF1 fusions, which have not been reported before in ossifying fibromyxoid tumour.ConclusionThese six cases expand the histomorphological spectrum of ossifying fibromyxoid tumour, introducing lipomatous differentiation as a hitherto undocumented feature. Awareness of these rare variants will ensure appropriate diagnosis and clinical management.

Effects of low temperature stress on cold-resistant substances and gene expression in Spodoptera frugiperda (Lepidoptera: Noctuidae)

The fall armyworm Spodoptera frugiperda (J. E. Smith) (Lepidoptera: Noctuidae) is a major agricultural pest. Low temperature is an important factor that limits S. frugiperda survival and reproduction. The physiological cold tolerance indexes of S. frugiperda larvae from October 2022 to February 2023 were investigated. The results showed that the cold-resistant substances in S. frugiperda differed significantly among different months. The water content was the lowest in January. Lipids were the highest in December and January, trehalose and protein contents were the highest in December, and glycerol content was the highest in January. The contents of lipid, protein, trehalose, and glycerol were the highest in the 6^th instar larvae, whereas the water content did not differ among the larva stages. The functions of two genes, including endothelial lipase (Eol1) and gastric lipase (Epl1) that were extracted from the transcriptome analysis of S. frugiperda in the cold resistance were assessed. After 24 h of cold stress, the mRNA levels of Eol1 and Epl1 RNA interference were significantly down-regulated by 60.76 % and 82.53 %, and the survival rate of S. frugiperda larvae decreased. Additionally, Eol1 and Epl1 silencing significantly decreased the lipid content and pupal weight of S. frugiperda. The results indicate that S. frugiperda larvae can change the contents in winter to response the cold conditions and the lipids plays an important role in the cold resistance of S. frugiperda.
