Synthesis, structural analysis, and systematic exploration of the antitumor activities of triphenyltin(IV) 2-hydroxy-5-(phenyldiazenyl)benzoates through the modulation of trifluoromethyl variants

By reacting 5-[(E)-2-(2-trifluoromethylphenyl)-1-diazenyl]-2-hydroxybenzoic acid (H'HL³), 5-[(E)-2-(3-trifluoromethylphenyl)-1-diazenyl]-2-hydroxybenzoic acid (H'HL⁴), and 5-[(E)-2-(4-trifluoromethylphenyl)-1-diazenyl]-2-hydroxybenzoic acid (H'HL⁵), with the triphenyltin source Ph₃SnOH, three triphenyltin(IV) 2-hydroxy-5-(phenyldiazenyl)benzoates [Ph₃Sn(HL³)] (3), [Ph₃Sn(HL⁴)] (4) and [Ph₃Sn(HL⁵)] (5) were obtained. The resulting tin complexes were characterized using standard spectroscopic techniques and single-crystal X-ray diffraction (SC-XRD). Triphenyltin complexes 3-5 exhibit a monomeric distorted tetrahedral configuration, with the fluoro substituted 2-hydroxy-5-(phenyldiazenyl)benzoates coordinating in a monodentate fashion. Additionally, the crystal structure of H'HL⁵ is reported. Alongside these, two triphenyltin compounds [Ph₃Sn(HL¹)] (1) and [Ph₃Sn(HL²)] (2), are included to evaluate and compare their anti-proliferative properties. Here, HL¹ and HL² represent 5-[(E)-2-(phenyl)-1-diazenyl]-2-hydroxybenzoate and 5-[(E)-2-(4-fluorophenyl)-1-diazenyl]-2-hydroxybenzoate, respectively. The in vitro antiproliferative activity of the triphenyltin(IV) compounds 1-5 was evaluated against MCF-7 (human breast cancer), HeLa (human cervical cancer), and HEK-293 (normal human embryonic kidney) cells and a mechanism of action is proposed on the basis of various biological assays.

Identification of high-risk non-synonymous SNPs (nsSNPs) in DNAH1 and DNAH17 genes associated with male infertility: a bioinformatics analysis

Male infertility is a significant reproductive issue affecting a considerable number of couples worldwide. While there are various causes of male infertility, genetic factors play a crucial role in its development. We focused on identifying and analyzing the high-risk nsSNPs in DNAH1 and DNAH17 genes, which encode proteins involved in sperm motility. A total of 20 nsSNPs for DNAH1 and 10 nsSNPs for DNAH17 were analyzed using various bioinformatics tools including SIFT, PolyPhen-2, CADD, PhD-SNPg, VEST-4, and MutPred2. As a result, V1287G, L2071R, R2356W, R3169C, R3229C, E3284K, R4096L, R4133C, and A4174T in DNAH1 gene and C1803Y, C1829Y, R1903C, and L3595P in DNAH17 gene were identified as high-risk nsSNPs. These nsSNPs were predicted to decrease protein stability, and almost all were found in highly conserved amino acid positions. Additionally, 4 nsSNPs were observed to alter post-translational modification status. Furthermore, the interaction network analysis revealed that DNAH1 and DNAH17 interact with DNAH2, DNAH3, DNAH5, DNAH7, DNAH8, DNAI2, DNAL1, CFAP70, DNAI3, DNAI4, ODAD1, and DNAI7, demonstrating the importance of DNAH1 and DNAH17 proteins in the overall functioning of the sperm motility machinery. Taken together, these findings revealed the detrimental effects of identified high-risk nsSNPs on protein structure and function and highlighted their potential relevance to male infertility. Further studies are warranted to validate these findings and to elucidate the underlying mechanisms.

Region-specific roles of oviductal motile cilia in oocyte/embryo transport and fertility

AbstractThe physiological and clinical importance of motile cilia in reproduction is well recognized; however, the specific role they play in transport through the oviduct and how ciliopathies lead to subfertility and infertility are still unclear. The contribution of cilia beating, fluid flow, and smooth muscle contraction to overall progressive transport within the oviduct remains under debate. Therefore, we investigated the role of cilia in the oviduct transport of preimplantation eggs/embryos using a combination of genetic and advanced imaging approaches. We show that the region of the oviduct where cumulus–oocyte complex circling occurs, around the time of fertilization, is correlated with asymmetrical mucosal fold arrangement and non-radially distributed ciliated epithelium. Our results suggest that motile cilia, as well as mucosal fold asymmetry, may contribute to the local flow fields that help steer luminal contents away from the epithelial walls. We also present, in vivo, volumetric evidence of delayed egg transport in a genetic mouse model with disrupted motile cilia function in the female reproductive system. Females with Dnah5 deleted in the oviduct epithelium are subfertile and demonstrate disrupted motile cilia activity within the oviduct mucosa. Fifty percent of Dnah5 mutant females have delayed egg transport where cumulus–oocyte complexes did not progress to the ampulla at the expected time point and remained within the ovarian bursa. The integration of advanced imaging with genetic dysfunction of motile cilia provides valuable insights into oviductal transport. Potentially, these data could be valuable for better understanding and management of tubal pathologies and human infertility.
