The purpose of this study is to assess the impact of TMP001 in the treatment of patients with relapsing-remitting multiple sclerosis (RRMS). Therefore the average total number of contrast enhancing lesions (CELs) on brain MRI scans at weeks 12, 16, 20, and 24 during treatment with TMP001 is compared to the average total number of CELs on brain MRI scans at week -4 and baseline in these patients .
Based on promising preclinical results, the investigators assume a comparable effect of TMP001 on reduction of contrast-enhancing lesions as shown for other immunomodulatory substances in recent clinical studies.

Bioavailability, Pharmacokinetics and Tissue Distribution of R-flurbiprofen Capsules in Healthy Subjects

For previous clinical trials, R-flurbiprofen has been prepared in tablet form. In this study R-flurbiprofen, will be available as gelatine capsules.
This study aims to show the bioavailability of R-flurbiprofen when administered in gelatine capsules. The serum availability will be determined by analysis of pharmakokinetic (pK)-blood samples at different time points. To assess the safety of the administered capsules adverse events will be documented.
Analysis of lipid signaling molecules in plasma will be done to assess the role of this molecules as variable for therapeutic effects.

Highly Efficient Chiral Separation Based on Alkali‐proof Protein Immobilization by Covalent Organic Frameworks

AbstractChiral separation plays a pivotal role in both practical applications and industrial productions. However, traditional chiral stationary phases (CSPs) exhibit inherent instability in alkaline environments, presenting a significant challenge despite their importance. Herein, basophilic alcalase is creatively developed to fabricate ultrastable protein‐based CSPs that can efficiently work under alkaline conditions. An in‐depth theoretical simulation is conducted to unveil the unique three‐dimensional conformation of alcalase, showing selective affinity towards various enantiomers of chiral amino acids and drugs, especially acidic substrates. Subsequently, an in situ assembly strategy is used to immobilize alcalase within a hydrazone‐linked covalent organic framework (COF) platform. The generated protein‐based CSPs enable successful baseline separation (Rs≥1.50) for various value‐added compounds (e.g., non‐steroidal drug, RS‐flurbiprofen; nucleotide analog, RS‐tenofovir) via high‐performance liquid chromatography, surpassing the commercial chiral column. Furthermore, a systematic study reveals that increasing hydrophilicity and pore sizes of COFs can enhance the separation performance. Remarkably, the obtained CSPs demonstrated exceptional durability, maintaining performance for >2,400 runs. This study provides a new member to the protein library for CSPs, and represents an innovative and effective platform for CSPs with immense potential for the enantioseparation of acidic drugs.

Preparation of novel chiral stationary phases based on chiral metal-organic cages enable extensive HPLC enantioseparation

BACKGROUNDThe metal organic cages (MOCs) are an emerging type of porous material that has attracted considerable research interest due to their unique properties, including good stability and well-defined intrinsic cavities. The chiral MOCs with porous structures have broad application prospects in enantiomeric recognition and separation. However, there are almost no relevant reports on chiral MOCs as chiral stationary phases (CSPs) for enantioseparation by high-performance liquid chromatography (HPLC).RESULTSHere, we report for the first time, the chiral MOCs, MOC-PA was "wrapped" onto the silica gel to create a novel HPLC CSP of chiral MOC-PA@SiO₂ through wrapped in net method. There are 24 and 20 chiral compounds or chiral drugs, including alcohols, diols, esters, ketones, alkene, epoxides, ethers, organic acids, phenols and amines, were enantioseparated by vary degrees on MOC-PA@SiO₂ packed column under both normal phase (NP) and reversed phase (RP) modes, respectively. Furthermore, compared with commercial Chiralpak AD-H and Chiralcel OD-H columns, the MOC-PA@SiO₂ packed column exhibits well chiral separation complementarity to them. It can separate some racemates that unable to be or not to be well separated by the Chiralpak AD-H and Chiralcel OD-H columns. Besides, there are four positional isomers were also well separated by MOC-PA@SiO₂ packed column.SIGNIFICANCEThe effects of temperature and injection volume on separation of MOC-PA@SiO₂ packed column were further investigated. This column also exhibited excellent repeatability, stability and column-to-column reproducibility on separation in HPLC. This work indicates that chiral MOCs are a promising class of chiral materials to prepare new CSPs for HPLC enantioseparation.

Redox Reactions of Organic Molecules Using Rotating Magnetic Field and Metal Rods

In recent years, redox reactions have harnessed light or mechanical energy to enable the formation of chemical bonds. We postulated a complementary approach that electromagnetic induction could promote the redox reaction of organic molecules using a rotating magnetic field and metal rods. Here, we report that electromotive force activates the redox-active trifluoromethylating reagents. This magnetoredox system can be applied to the trifluoromethylation of heteroarenes with high regioselectivity and hydrotrifluoromethylation of alkenes without the need for catalysts and organic additives.
