Approximately 21 subjects with amyotrophic lateral sclerosis (ALS) will be randomized (6 to 1) to receive by mouth seven morning doses of CC100 or placebo for 7 days. Subjects are required to stay in the Clinic for approximately 9 hours following the first and last dose. Subjects will also have a mid-week clinic visit and will be contacted by phone within 3 to 5 days after the last dose.

Matrix metalloproteinases (MMPs); activated by the application of adhesive systems on dentin cause degradation of the collagen proteins and loss of bond strength in adhesive restorations. Caffeic acid phenethyl ester (CAPE) showed inhibitory activities on dentinal MMPs. It is hypothesised that dentin pretreatment with CAPE, as an MMP inhibitor, would be beneficial for the bonding of resin material and tooth substrate over the course of time. This study evaluated the effect of 5 % CAPE pretreatment on the microtensile bond strength of a nanohybrid resin restoration applied with total-etch or self-etch adhesive systems on 10 patients (12-18 yrs). Two standard cavities (3 x 3 x 4 mm) prepared on the occlusal surfaces of one of the two premolar teeth in each patient were randomly assigned into four treatment groups (n=10); 5% CAPE pretreatment and total-etch adhesive (TC), Total-etch adhesive (control) (T), 5% CAPE pretreatment and self-etch adhesive (SC), Self-etch adhesive (control) (S) followed by restoration and 3 months of intra-oral functioning. After teeth extraction, the specimens were stored in distilled water+thymol solution at 4°C to inhibit microbial growth and used in one week period. Dentin-composite sticks were obtained from each restoration for µTBS tests and scanning electron microscopy (SEM) evaluation. Failure mode analysis was performed microscopically. Data were analysed statistically using Repeated Measure ANOVA (p < 0.05). Dentin pretreatment with CAPE solution (5%) for 60 seconds significantly increased the composite resin restorations' bond strengths to dentin applied with either total-etch or self-etch adhesive system. Thus it can be used as an MMP inhibitor prior to the the resin restorations. However further long term clinical studies are needed to support the use of CAPE pretreatment along with self-etch and total-etch adhesive systems.

Evaluation of Efficacy of Curcumin and Caffeic Acid Phenethyl Ester in Breast Cancer by Preclinical Studies

Aims::The aim of this study was to evaluate the combined and comparative efficacy of Caffeic acid phenethyl ester (CAPE) and curcumin in breast cancer.background:CAPE and curcumin are a class of phenolics. While curcumin is obtained from turmeric, CAPE is found in Baccharis sarothroides and Populus deltoides. Both agents are reported to produce activities in some cancer types. The combined and the comparative effects of two agents in breast cancer is not yet reported.objective:We evaluated the potential of CAPE and curcumin in both in vitro and in vivo breast cancer models.method:Human breast cancer cell lines, MDA-MB-231 and MCF-7 were exposed to CAPE and curcumin followed by functional assays such as cell cytotoxicity, cell proliferation and colony formation, cell cycle, mitochondrial membrane potential, apoptosis, and monodansylcadaverine (MDC) staining for autophagy. Computational analyses and mice model were also used.result:Employing computational analyses, both agents were found to exhibit drug like properties. Both molecules interacted with the key molecules of the NF-B pathway. CAPE and curcumin inhibited cell proliferation, colony formation, and invasion, and triggered apoptosis and autophagy in breast cancer cells. CAPE was found to be more effective in comparison to curcumin. Two agents working together were more effective than each agent working alone. Both agents suppressed the expression of survivin, Bcl-xL and GLUT-1. Further, cleaved PARP was increased by both agents. An induction in ROS generation was observed by both phenolics. Further, both molecules triggered a dissipation in mitochondrial membrane potential. In mice model implanted with Ehrlich-Lettre ascites carcinoma (EAC) cells, both drugs inhibited the growth of the tumour. The phenolics also modulated the metabolic parameters in tumour bearing mice.conclusion:The observations suggest that the combination of curcumin plus CAPE may be better in comparison to individual molecule.other:The study opens a window for analysing the efficacy of combination of CAPE and curcumin by more animal studies. This will provide a basis for examining the combined efficacy of two agents by clinical trial.

Periodontitis is an oral chronic inflammatory disease induced by pathogenic bacteria. Pathogens continuously activate immune cells to express a large number of pro-inflammatory factors and reactive oxygen species, eventually leading to periodontal tissue damage. Poor eradication of pathogenic bacteria, persistent inflammatory response and impaired periodontal tissue regeneration are the main challenges to control the progression of periodontitis. However, current clinical drug treatment fails to comprehensively address these issues. In this paper, a chitosan-P407-PNIPAM scaffold multi-crosslinked hydrogel (A/CQ-ML@Gel) encapsulating AgNPs and lipid complex (CQ-ML) is developed. This hydrogel shows thermo- and pH-sensitive properties, exhibits excellent injectability, high viscosity and reliable post-injection mechanical strength. A/CQ-ML@Gel possesses a significant antibacterial effect to Porphyromonas gingivalis after implantation, and then by virtue of the programmed release of quercetin microemulsion and caffeic acid phenethyl ester in CQ-ML, exerting excellent inflammation regulation and osteogenic differentiation of periodontal ligament stem cells. Notably, A/CQ-ML@Gel could activate mitophagy through PINK1/Parkin in inflammatory macrophages, thereby inhibiting the production of excess reactive oxygen species, ultimately reprogramming M1/M2 macrophages phenotype for inflammation suppression. In summary, we present an innovative insight into periodontal delivery system for trimodal synergistic therapy strategy in periodontitis.

Caffeic Acid Phenethyl Ester Inhibits Metastatic Properties of Acid-adapted Gastric Cancer Cells

BACKGROUND/AIMThe acidic tumor microenvironment promotes cancer invasiveness, epithelial-mesenchymal transition, and therapeutic resistance. This study aimed to investigate the long-term effects of acidic adaptation on gastric cancer cells and evaluate the anticancer properties of caffeic acid (CA) and caffeic acid phenethyl ester (CAPE) in this context.MATERIALS AND METHODSSNU601 gastric cancer cells were cultured in prolonged acidic conditions (pH 6.7) to establish an acid-adapted subline (SNU601-6.7). Invasion assays and qPCR were used to assess invasive potential and the expression of matrix metalloproteinases (MMPs). The effects of CA and CAPE on viability, apoptosis, invasion, and β-catenin expression were evaluated.RESULTSSNU601-6.7 cells exhibited increased invasiveness, along with upregulation of MMP2, MMP7, and MMP9. Both CA and CAPE reduced cell viability and invasion, with CAPE exerting a significantly stronger effect and inducing moderate apoptosis. Mechanistic studies revealed that CAPE decreased total and nuclear β-catenin levels, and inhibited AKT and GSK3β phosphorylation. Further, pharmacological inhibition of AKT pathway confirmed its role in β-catenin accumulation and cell invasiveness.CONCLUSIONThese findings identify CAPE as a potent inhibitor of invasion in acid-adapted gastric cancer cells by targeting the AKT/β-catenin pathway, highlighting its potential as a therapeutic candidate for gastric cancer in acidic tumor microenvironments.
