A recent international survey on aerosol therapy during mechanical ventilation (Ehrmann et al. 2013) reported that jet nebulizers remained the mostly used.

Uranium, once introduced into the human body, tends to accumulate in the kidneys and femurs. Its potent chemical toxicity and radioactivity can lead to severe damage, posing significant health risks. Consequently, there is an urgent need to develop low-toxicity materials capable of effectively detecting and removing uranium. In this study, a novel N^N-bidentate ligand modified with a tetraphenylethylene derivative, designated as HI, was synthesized. This bidentate ligand demonstrates dual functionality: it can selectively recognize UO₂^2 + both in vitro and in vivo, and it effectively facilitates the removal of UO₂²⁺. HI exhibits remarkable sensitivity for UO₂²⁺. Compared to a commercial decorporation agent diethylenetriamine pentaacetic acid (DTPA), HI shows lower toxicity and superior removal efficiency for UO₂²⁺. Furthermore, HI mitigates oxidative stress and DNA damage by chelating with UO₂²⁺, thereby significantly reducing the high rates of cell apoptosis induced by UO₂²⁺ exposure. Notably, prophylactic administration of HI resulted in the removal of 65.5 % and 39.8 % of UO₂²⁺ from the kidneys and femurs of mice, respectively. Given its promising properties, HI emerges as a potential candidate for both the detection and remediation of UO₂²⁺ pollution, offering a new avenue for addressing the health challenges posed by contamination of uranium.

Sulfur self-doped hierarchical porous carbon materials synthesized by one-pot method for efficient adsorption of thallium(I)

In this work, sulfur self-doped hierarchical porous carbons (KLC-X-Y) were fabricated via one-pot method using inexpensive sodium lignosulfonate as carbon and sulfur source and potassium hydroxide (KOH) as activator.Through detailed anal., it was found that the carbonization temperature and KOH addition exhibited significant effects on the porosity, phase composition, functional groups and morphol. of KLC-X-Y.Among them, KLC-700-2 possessed a hierarchical porous structure, high sp. surface area (851.3 m²·g^-1), large pore volume (0.53 cm³·g^-1), appropriate pore size distribution (D = 2.49 nm), and abundant oxygen- (-COOH and -OH) and sulfur-containing (-SO₃H and -SH) functional groups.Notably, the synthesized KLC-700-2 displayed excellent adsorption performance for Tl(I) with a maximum adsorption amount of 523.7 mg·g^-1.After five consecutive adsorption-desorption processes, the adsorption amount of Tl(I) by KLC-700-2 decreased slightly, which still reached 85.6% of the initial adsorption amount, and its own structural properties didn't change significantly, indicating that KLC-700-2 had good stability and reusability.The adsorption process of Tl(I) on KLC-700-2 was verified to be spontaneous and endothermic, and conformed to the pseudo second order and Langmuir models by adsorption kinetic, isotherm, and thermodn. studies.By comparing the physicochem. properties of KLC-700-2 before and after adsorption of Tl(I), it was confirmed that the adsorption of Tl(I) on KLC-700-2 was co-controlled by chem. and phys. adsorption, including ion exchange, precipitation reaction, surface complexation, pore filling and electrostatic attraction.

Comparative evaluation of chelators in the context of analysis of antibody oxidative stability

Oxidation is a critical post-translational modification for antibodies that naturally possess many solvent-exposed amino acid residues. Presence of oxidized methionine and tryptophan residues may potentially impact safety, efficacy, clearance, and immunogenicity of antibodies, emphasizing the importance of including oxidation assays as a part of the antibody control strategy. Sub-unit Reversed Phase HPLC is a commonly employed method to measure oxidation, wherein enzymatic digestion of antibodies followed by disulfide reduction is essential for sample preparation. Peaks resulting from intact sub-units i.e. light chain (LC), single-chain crystallizable fragment (Fc), and N-terminal half of heavy chain (Fd) and additional peaks arising due to subunit oxidation can be easily resolved to quantify oxidative degradation. Our data suggest that the performance of the method can be compromised due to sample oxidation during the analysis window imposing restrictions on sample throughput and increasing method variability. Drug substance batches of six monoclonal antibodies with variable subclasses and isoelectric points showed an increase of 5-50% in subunit oxidation levels when tested over 48 hours. With an aim to minimize oxidation during analysis we modified the sample preparation technique using a variety of metal chelators including Ethylenediaminetetraacetic acid (EDTA), Ethylene Glycol-bis (β-aminoethyl ether)-N,N,N',N'-Tetraacetic Acid (EGTA), Diethylene Triamine Pentaacetic Acid (DTPA), Nitrilotriacetic acid (NTA) and Hydroxyethyl Ethylenediamine Triacetic Acid (HEDTA). EGTA showed the most prevalent stabilizing influence on subunit oxidation for all drug substance batches screened. Overall, our results demonstrate that careful optimization of sample preparation during oxidation analysis can increase sample throughput significantly and highlight the feasibility of using EGTA as a more suitable alternative to EDTA, the most employed trace-metal chelator in the biopharmaceutical space.

Curium Announces Approval of Generic DTPA - Kit for the Preparation of Technetium Tc 99m Pentetate Injection in the U.S.

ST. LOUIS, April 02, 2025 (GLOBE NEWSWIRE) -- Curium announced today that the U.S. Food and Drug Administration approved its generic version of the Kit for the Preparation of Technetium Tc 99m Pentetate Injection, often referred to as DTPA (diethylenetriaminepentaacetic acid). Technetium Tc 99m pentetate is a radioactive diagnostic most often used for lung ventilation imaging and evaluation of pulmonary embolism when paired with perfusion imaging in adult and pediatric patients and administered by nebulizer for inhalation among other indications. Curium is now able to offer hospitals and nuclear pharmacies a suite of products used in ventilation/perfusion (V/Q) scans, streamlining product fulfillment and bolstering the supply chain to include a second supplier of this medication. Long-term cumulative radiation exposure is associated with an increased risk of cancer. Ensure safe handling and preparation procedures to protect patients and health care workers from unintentional radiation exposure. Use the lowest dose of Technetium Tc 99m pentetate necessary for imaging. Encourage patients to drink fluids and void as frequently as possible after intravenous administration. Radiation risks associated with the use of Technetium Tc 99m pentetate are greater in pediatric patients than in adults due to greater radiosensitivity and longer life expectancy. Bronchospasm in Lung Ventilation Studies: As with other inhaled medications, inhalation of Technetium Tc 99m pentetate solution may result in acute bronchoconstriction, especially in patients with heightened bronchoreactivity, such as patients with asthma or other lung or allergic disorders. Monitor all patients for bronchoconstriction. ADVERSE REACTIONSAdverse reactions are presented in decreasing order of reported frequency: Immune system disorders: allergic reaction, anaphylactic reaction, angioedema.Skin and subcutaneous tissue disorders: rash, itching, hives, erythema.Respiratory, thoracic and mediastinal disorders: throat irritation, wheezing.Vascular disorders: hypotension, hypertension.Nervous system disorders: headache, fainting, dizziness.General disorders and administration site conditions: chills.Gastrointestinal disorders: nausea, vomiting.Cardiac disorders: cyanosis, tachycardia. USE IN SPECIFIC POPULATIONSPregnancy: Limited available data with Technetium Tc 99m pentetate use in pregnant women are insufficient to inform a drug associated risk for major birth defects and miscarriage. Technetium Tc 99m pentetate is transferred across the placenta. No animal reproductive studies have been conducted with Technetium Tc 99m pentetate. All radiopharmaceuticals have the potential to cause fetal harm depending on the fetal stage of development and the magnitude of the radiation dose. If considering Technetium Tc 99m pentetate administration to a pregnant woman, inform the patient about the potential for adverse pregnancy outcomes based on the radiation dose from Technetium Tc 99m pentetate and the gestational timing of exposure. The estimated background risk of major birth defects and miscarriage for the indicated population is unknown. In the U.S., general population, the estimated background risk of major birth defects and miscarriage in clinically recognized pregnancies are 2% to 4% and 15% to 20%, respectively. Lactation: There are limited data available in scientific literature on the presence of Technetium Tc 99m pentetate in human milk. There are no data available on the effects of Technetium Tc 99m pentetate on the breastfed infant or the effects on milk production. Based on the United States Nuclear Regulatory Commission guidelines for breastfeeding interruption after exposure to radiopharmaceuticals, breastfeeding interruption is not recommended for Technetium 99m pentetate, at levels less than 1000 MBq (30 mCi). The developmental and health benefits of breastfeeding should be considered along with the mother’s clinical need for Technetium Tc 99m pentetate, any potential adverse effects on the breastfed child from Technetium Tc 99m pentetate or from the underlying maternal condition. Pediatric Use: Technetium Tc 99m pentetate is indicated for lung ventilation and evaluation of pulmonary embolism when paired with perfusion imaging and for renal visualization, assessment of renal perfusion, and estimation of glomerular filtration rate in pediatric patients ages birth to less than 17 years of age. Pediatric use is supported by evidence from controlled studies in adults and dosing and safety are based on clinical experience. The radiation risk of Technetium Tc 99m pentetate is greater in pediatric patients than adults. Geriatric Use: No formal studies of Technetium Tc 99m pentetate in the elderly were performed to determine whether they respond differently from younger subjects. Other reported clinical experience has not identified differences in responses between the elderly and younger patients. In general, dose selection for an elderly patient should be cautious, usually starting at the low end of the dosing range, reflecting the greater frequency of decreased hepatic, renal, or cardiac function, and of concomitant disease or other drug therapy.
