There are more and more herbal preparations that are used for the purpose of treatment and improvement of the clinical picture of vaginitis by patients themselves, but also by healthcare professionals. Plant species, St. John's wort, chamomile, calendula, yarrow, shepherd's purse and tea tree oil are well known for there anti-inflammatory, antimicrobial and wound healing activity. This paper presents the results of a clinical study in which three herbal formulations/vagitories, based on extracts of above mentioned plant species, were investigated for their effectiveness on non-specific vaginitis.
This was randomized controlled clinical study that included 210 women with diagnosed non-specific vaginitis. Patients were divided into two basic groups, women in reproductive stage and postmenopausal stage. Three subgroups, containing approximately 30 patients each, received one of three vagitorie formulations for 5 days during which the effects on subjective and objective symptoms were monitored.

Proton Pachinko: Probing Excited‐State Intramolecular Proton Transfer of St. John's Wort‐Derived Fluorescent Photosensitizer Hypericin with Ultrafast Spectroscopy**

AbstractHypericin from St. John's wort has been used as a potent photosensitizer, but its working mechanism remains elusive which hinders its rational design for improved functionality. We implement ultrafast spectroscopy and quantum calculations to track the excited‐state dynamics in an intricate hydrogen‐bonding network of hypericin in solution. Using femtosecond transient absorption (fs‐TA), we track excited state intramolecular proton transfer (ESIPT) via a previously unreported blueshift of a long‐wavelength stimulated emission (SE) band with excitation‐dependent dynamics in various solvents, owing to the dominant Q7,14 tautomer that undergoes bidirectional ESIPT. This finding is corroborated by ground‐state femtosecond stimulated Raman spectroscopy (GS‐FSRS) and density functional theory (DFT) calculations. Moreover, contrasting the neutral and anionic forms of hypericin enables us to reveal an intramolecular charge transfer step underlying ESIPT. We demonstrate UV and visible excitations as an integral platform to provide direct insights into the photophysics and origin for phototoxicity of hypericin. Such mechanistic insights into the excited state of hypericin will power its future development and use.

Thermoplastic Polyurethane/Polylactic Acid Blend Foams Loaded With Curcuma longa L. and Hypericum perforatum Extracts Towards Wound Dressing Applications

ABSTRACTThis study aimed to fabricate and evaluate TPU/polylactic acid foams loaded with natural extracts of Curcuma longa L. (C) and Hypericum perforatum (H) for WD application. In the frame of the structure/properties relationships study, different and complementary analyzes for WD materials, such as water contact angles measurements, water vapor transmission rate, water absorption, and extracts release studies were performed, in connection with the structural and thermal characterizations performed using scanning electron microscopy, differential scanning calorimetry and Fourier transform spectroscopy. The water absorption capacity of the blank blend foam and foams loaded with extracts (C and H) was found to be in the range of 8.31% ± 0.27, 90.7% ± 4.1 and 164% ± 4.0%, respectively. The loaded foams presented cumulative extracts release of 45.5% ± 2% (C) and 100% ± 1.1% (H) within 96 h and acceptable water vapor transmission rate in the range of 2088 ± 66 and 3520 ± 991 g·m−2·24 h−1, respectively. Summary, the investigated foams, enriched with both extracts, showed excellent performances in agreement with WD materials requirements, higher cell viability and proliferation compared to the reference foams, as well as non‐cytotoxic effects on human fibroblasts in experimental conditions.

Evaluation of binding interaction between compounds targeting peroxisome proliferator-activated receptor γ in Nelumbinis folium using receptor chromatography and molecular dynamic simulation

Despite considerable efforts invested in clinical trials aimed at treating obesity and enhancing the metabolic profiles of Nelumbinis Folium, the precise phytochemicals involved and their mechanisms of action remain unclear due to the absence of an efficient screening technique. Herein, Nelumbinis Folium serves as the focal point to elucidate the bioactive compounds that specifically bind to peroxisome proliferator-activated receptor γ using immobilized receptor chromatography. Following identification through liquid chromatography-mass spectrometry, the compounds were further evaluated using chromatographic techniques and molecular dynamics simulations. The results unveiled catechin and hypericin as the receptor-binding compounds present in Nelumbinis Folium, with hypericin exhibiting a stronger affinity and a faster dissociation rate constant compared to catechin. Molecular dynamics studies highlighted the crucial role of cysteine located at position of 285 in the receptor ligand binding domain during the initial ligand capture phase. Subsequently, Van Der Waals forces and electrostatic interactions facilitated the binding process. The calculated standard binding free energies were - 61.75 ± 2.61 kcal/mol for hypericin and - 43.19 ± 0.63 kcal/mol for catechin. Collectively, these findings provide valuable insights into receptor-drug interactions and confirm the effectiveness of immobilized receptor chromatography in screening potential lead compounds from complex systems.
