Characterization of Night Vision Impairment in Choroideremia and Short-Term Vitamin A Supplementation: The Dark-Adapted Retinal Function Response in Choroideremia (DARC) Study

Choroideremia (CHM) is an inherited retinal disorder that causes progressive vision loss, ultimately leading to complete blindness. The first symptom is generally night blindness, although, to date, little is known about the extent, type, pattern, and progression of dark-adapted visual function measures in CHM patients. We hypothesize that one of the key events causing night blindness in CHM is deficiency in the chromophore of the rod visual pigment, rhodopsin. We propose that this deficiency is at least in part due to inadequate delivery of vitamin A (all-trans-retinol) to the photoreceptors (PRs) from the ailing retinal pigment epithelium (RPE), characteristic of CHM. We hypothesize that increased availability of vitamin A would potentiate its entry into the RPE-mediated visual cycle, ultimately enabling delivery to the PRs. This would in turn allow rods to perform better by partially overcoming the RPE damage and the impaired chromophore recycling that we postulate exists in CHM. The goals of this proposal are: (1) to test the hypothesis that oral vitamin A supplementation can improve night time and peripheral vision in CHM patients, and (2) to provide detailed characterization of dark-adapted visual function outcome measures to guide interventional CHM trials.

Field-realistic cyfluthrin exposure alters multi-omics profiles in Bombus terrestris: Implications for wild pollinator health

Cyfluthrin is effective in mitigating crop damage caused by pests. However, its residues in bee colonies pose potential threats to the health and fitness of bumblebees. While previous studies have shown significant physiological effects of cyfluthrin on honeybees, its effects on bumblebees remain poorly understood. In this study, bumblebees were exposed to 30 and 120 μg/kg cyfluthrin (two field-level concentrations) for 10 days. We assessed the effects of chronic cyfluthrin exposure on bumblebee survival, sucrose solution consumption, body weight and midgut pathology. Multi-omics analyses were performed, encompassing the gut microbiome and metabolome, as well as the head transcriptome and proteome. The results demonstrated that exposure to cyfluthrin at a concentration of 120 μg/kg significantly reduced the survival probability of bumblebees. Hematoxylin-Eosin staining revealed damage to the midgut microvilli. 16S rRNA sequencing indicated a significant decrease in the relative abundance of the core gut microbiota, Bifidobacterium. Metabolomic analysis of the gut further revealed a significant reduction in multiple beneficial metabolites (crustecdysone, creatine and retinol palmitate). Head transcriptomics showed significant downregulation of genes associated with the tyrosine metabolism pathway and fatty acid β-oxidation. Furthermore, proteomic analysis of the head revealed a significant upregulation of apoptosis-related proteins, alongside a significant downregulation of proteins involved in immune responses and glycosaminoglycan biosynthesis. These findings highlight the need for cautious pesticide application in agricultural systems and emphasize the importance of considering sublethal effects on pollinators - particularly through disruptions to the gut and head.

An Encapsulated Vitamin A Palmitate Powder With Improved Stability for Use in Food Fortification

ABSTRACT:

Vitamin A deficiency (VAD) is a serious health concern, particularly in low‐ and middle‐income countries. Food fortification is hindered by the instability of vitamin A. The objective was to develop a dry form of vitamin A palmitate (VAP) with improved stability compared to marketed products. This powder could be employed for large‐scale food fortification to help prevent VAD. VAP is emulsified with a variety of food‐grade ingredients, including basic methacrylate copolymer (BMC). A spray‐dried BMC‐encapsulated VAP (PFH‐VAP) was produced using laboratory‐scale equipment. Subsets were assessed for determination of the VAP content and VAP recovery after cooking in water. Another subset was subjected to accelerated stability testing. Two high‐performance formulations were produced at the pilot scale. The two most promising PFH‐VAP formulations and a commercially available vitamin A product were each incorporated into bouillon cubes, stored (40°C, 75% relative humidity), and subjected to stability testing. The optimized laboratory‐scale formulations exhibited > 90% VAP recovery after cooking. The pilot‐scale batches showed 95% and 65% VAP recoveries after cooking and accelerated stability testing, respectively. After 12 months of stability testing, 70% and 68% VAP recoveries were achieved for the pilot‐scale batches of PFH‐VAP‐fortified bouillon. A 15% VAP recovery was obtained from the bouillon fortified with a commercial product. PFH‐VAP demonstrated a substantial stability advantage over an existing commercial formulation of VAP in a proof‐of‐concept food vehicle (bouillon). The use of PFH‐VAP could increase the nutritional and health benefits of vitamin A‐fortified foods and condiments.

Abstract:

Preschool-aged children and non-pregnant women of reproductive age particularly in countries of a lower socio-demographic index may experience insufficient dietary vitamin A. Fortifying staple foods such as edible oils with vitamin A is a useful approach to reduce malnutrition. To enable better quality oil fortification, producers need reliable and inexpensive tools for assessment of the vitamin A contents of their products. A simple reversed-phase high-performance thin-layer chromatography fluorescence detection (RP–HPTLC–FLD) method was developed and studied for the quantification of vitamin A palmitate (VAP) in five edible plant oils. The challenging matrices of non-refined palm kernel oil and soybean oil, which are known to interfere with the accuracy of some easy-to-use test kits, were separated from VAP using the new RP–HPTLC–FLD method. The standard addition method provided good accuracy and acceptable calibration curves (coefficients of determination R2 between 0.9874 and 0.9999) even for manual application. The developed RP–HPTLC–FLD method proved simple, robust, sustainable, cost-efficient, and suitable for low-tech (minimum equipment) on-site analysis. Graphical Abstract

Particles for Humanity Celebrates Codex Adoption of Key Ingredient in PFH-VAP for Use in Sugar and Rice

New Delhi: In a significant move aimed at safeguarding public health, the Indian government has banned 156 fixed-dose combination (FDC) drugs, including a variety of antibiotics, painkillers, multivitamins, and medications for treating fever and hypertension. This decision marks the largest crackdown on FDCs since 2016, when 344 such drugs were prohibited. The Union Ministry of Health and Family Welfare issued a gazette notification on Thursday, officially prohibiting the manufacture, sale, and distribution of the banned FDCs. The ban was based on the recommendations of an expert panel that reviewed a total of 324 FDCs.
