The aim of the proposed study is to investigate the effect of increased plasma lactate concentrations on the heart and brain's metabolism of other energy sources, primarily glucose in the brain and glucose and FFA in the heart.
The investigator hypothesize that:
* An acute increase in lactate plasma concentration will decrease cerebral glucose uptake and increase cerebral blood flow
* An acute increase in lactate plasma concentration will decrease palmitate and glucose uptake in the myocardium, and increase myocardial perfusion

Oral sodium lactate ingestion does not increase blood lactate concentrations and is accompanied by moderate-to-severe gastrointestinal side effects

Lactate has diverse roles in physiology, and a variety of methods have been used to explore its effects on metabolism. Here, we explored whether oral sodium lactate (Na-Lactate) ingestion could increase blood lactate concentrations. Oral Na-Lactate ingestion using a variety of dosing protocols did not increase blood lactate concentrations and was coupled with gastrointestinal issues. These results demonstrate that oral ingestion of Na-Lactate is not an effective method for studying lactate’s role in metabolism.

Effects of sodium lactate injection on meat quality and lactate content in broiler chickens: emphasis on injection method and dosage

This study aims to develop an experimental model of high lactate levels in broilers to mimic the condition of birds under stress or diseases and evaluate its consequent effects on meat quality. The injection sites and dosage effects were compared separately in 2 experiments. Experiment 1 includes 3 injection sites: intraperitoneal injection, intramuscular injection, and subcutaneous injection. Experiment 2 was a dosage experiment based on the results of Experiment 1: sodium lactate intraperitoneal injection group with 1.5, 3, 6 mM concentration. The results showed that injecting sodium lactate intraperitoneally, intramuscularly, or subcutaneously all significantly decreased body weight and breast muscle weight while elevating lactic acid levels in both the blood and breast muscle of broilers. Moreover, all 3 injection methods caused a significant reduction in pH_24h and an increase in the shear force value of breast muscle. In addition, dose-response experiments of intraperitoneal injection showed that a concentration of 3 mM and 6 mM were significantly decreased body weight and breast muscle weight in broiler chickens, accompanied by a notable increase in breast muscle lactate content. Compared to the control group, intraperitoneal injections of 1.5 mM, 3 mM, and 6 mM sodium lactate treatments significantly reduced the yellowness values of the breast muscle. As the dose of sodium lactate increased, the shear force value of the breast meat exhibited linear and quadratic increments, while the drip loss decreased linearly. Intraperitoneal injection of 3 mM sodium lactate also significantly reduced the pH_24h of broiler breast muscle. In addition, an increased dose of lactate injections up-regulated the glycolytic pathway responsible for endogenous lactate production in the breast muscle by upregulating the expression of phosphofructokinase, pyruvate kinase and lactate dehydrogenase A. In conclusion, intraperitoneal injection of sodium lactate at 3 mM directly increased breast muscle lactate levels, providing a valuable method for establishing a high-level lactate model in poultry.

Intravenous fluid therapy compared to no treatment following blood donation in cats: a randomised controlled trial

ObjectivesThere is currently no consensus regarding the use of intravenous fluid therapy in feline patients post‐blood donation in veterinary medicine. The primary aim of this study was to determine whether blood donation can be performed safely without post‐donation intravenous fluid therapy. The secondary aim was to report owner‐noted post‐donation changes.Materials and MethodsThe study aimed to enrol 100 conscious feline blood donations by client‐owned cats performed at a veterinary teaching hospital. Donors were randomised to either receive intravenous compound sodium lactate (twice the volume of blood donated over 2 hours) immediately after blood donation, or to receive no post‐blood donation intravenous fluid therapy. Systolic blood pressure was measured non‐invasively at 0, 60 and 120 minutes post‐donation. Median blood pressures were compared between the two groups using a Shapiro‐Wilk test. Owners were called the day following the donation to collect information on changes in their cat post‐donation.ResultsOne hundred cats were enrolled and the data of 97 cats were analysed; 46 who received intravenous fluid therapy and 51 who did not. Mean donation volume was 9.95 mL/kg for the intravenous fluid therapy group and 9.72 mL/kg for the non‐intravenous fluid therapy group. At each time point, the median blood pressure did not differ significantly between the two groups. The main reported changes in both goups were bruising at the venepuncture site with 27.3% (12/44) in the IVFT group and 23.4% (9/37) in the no‐IVFT group, and mild lethargy up to a maximum of 24 hours post donation with 11.4% (5/44) in the IVFT group and 18.9% (7/37) in the no‐IVFT group.Clinical SignificanceThis study suggests that the use of intravenous fluid therapy post‐feline blood donation may not be necessary. This could mean reduced hospitalisation time for feline donors, possibly decreasing feline stress.
