Delving into the Latest Updates on Amino Acids with Synapse

Cow's milk allergy (CMA) is a common food allergy in infants and young children that can have a significant impact on the individual and their family due to dietary restrictions, risk of nutritional deficiencies, social limitations, and decreased quality of life. It also represents a financial burden for families and healthcare resources.
There is an ongoing debate about the significance of early exposure to cow's milk proteins within hours or days after birth and its relationship to the risk of developing CMA later in life. Current recommendations for early introduction of cow's milk proteins in infants who cannot be breastfed vary and are inconsistent due to a lack of clear evidence. This knowledge gap underscores the need for further research to provide a definitive understanding of the relationship between early exposure to cow's milk proteins and the development of CMA, which will ultimately inform evidence-based prevention strategies to improve the health and well-being of affected individuals and their families.
This trial aims to investigate whether early supplementation with various nutritional interventions (cow's milk formula [CMF], amino acids formula [AAF], donor human milk [DHM], or high-pressure processed "pascalized" donor human milk [DHM-P]) could serve as an effective strategy for the primary prevention of CMA in breastfed neonates.
This study is an open-label randomized, controlled, head-to-head trial with four parallel arms and allocation 1:1:1:1.

Re-evaluation of the Muscle-full Effect During Continuously Elevated Amino Acid Availability in Healthy Young Males

Muscle tissue consists of proteins. These proteins are built up of a collection of smaller building blocks: amino acids. When protein is consumed, it gets digested and absorbed into the blood. The body can use these amino acids, by taking them up from thecirculation. By consuming sufficient protein through our diet, we ensure that the body is provided with enough amino acids to enable muscle protein building. Sufficient muscle protein synthesis is important for maintaining muscle function and strength.
Previous research has shown that when 20 to 25g of protein is eaten, muscle protein synthesis is maximized. It is therefore recommended to eat 20g of protein per meal. However, it is currently unclear what happens to muscle protein synthesis rates if multiple meals are eaten. When multiple meals are consumed, amino acids appear in the circulation for prolonged period of time.
Theoretically, when there are a high amino acid concentrations in the blood, muscle protein synthesis rates will increase. Contrary to this theory, a study more than 20 years ago showed otherwise. It was observed that muscle protein synthesis rates are only elevated for2 hours afterwhich they decrease again. This phenomenon was referred to as the "muscle-full" effect. Because this phenomenon is in contrast with more previous studies, the objective is to replicate that study. This is important so that nutritional advice for healthy, but also clinical populations in the future can be improved.

Amino acid-mTOR pathway-associated transcription factor GATAβ4 regulates storage protein expression in Bombyx mori

Storage proteins (SPs) are hexameric macromolecular protein, an important component of insect serum protein, which plays a variety of roles in insect metamorphosis and development. However, their regulatory mechanisms remain unclear. Our previous studies revealed that the expression of SPs is regulated by nutritional signals and identified FoxO as a negative regulator of SPs in the silkworm Bombyx mori (B. mori). In this study, amino acids upregulated BmSP expression, whereas Rapamycin downregulated it in fat body cultured in vitro. Rapamycin also reduced BmSP expression in B. mori larvae. Overexpression of the nutrient transcription factor GATA family in BmE cells revealed that only BmGATAβ4 significantly upregulated BmSP expression. Furthermore, the amino acid-mTOR signaling pathway modulated BmGATAβ4 expression. Overexpression of BmGATAβ4 resulted in increased BmSP expression in B. mori larvae. Luciferase reporter assays, electrophoretic mobility shift assays, and chromatin immunoprecipitation identified GATA-like CRE 1-1 and GATA-like CRE 2-2 of the BmSP1 promoter as binding sites for BmGATAβ4. These findings provide new insights into the regulation of nutrient protein expression in insects.

Factors affecting the intracytoplasmic sperm cell injection outcomes: A meta-analysis of porcine studies

BACKGROUND:

Intracytoplasmic sperm cell injection (ICSI) has the potential to produce gene-edited (GE) pigs for biomedical research, but its success is limited. The factors impeding ICSI in pigs are impractical in-vivo oocyte production, incomplete cytoplasmic maturation of in-vitro matured (IVM) oocytes, inefficient methods for sperm selection and membrane removal, abnormal sperm nucleus decondensation, substandard protocols for oocyte stimulation, suboptimal in-vitro culture (IVC) systems, and high embryonic/fetal losses.

AIM OF REVIEW:

The aim of this review is to investigate the effects of interventions in ICSI on oocyte activation, fertilization, cleavage, blastocyst, blastomere count, and live birth by means of robust statistical meta-analytical methods.

KEY SCIENTIFIC CONCEPTS OF REVIEW:

A total of 61 studies published between 1905 ∼ 2024 met the inclusion criteria. The results of the meta-analysis suggested that manipulation in the IVM media did not improve oocyte developmental competency to blastocysts but increased the blastomere count, especially with the addition of thiol compounds. Consistently, manipulation with sperm was beneficial only for increasing the cleavage and blastomere count. Exogenous stimulation increased the relative risk (RR) for oocyte activation (10 %), fertilization (33 %), cleavage (18 %), and blastocyst formation (71 %) but did not affect the blastomere count. Chemical stimulation either pre- or post-ICSI was more beneficial than electrical stimulation. Manipulation of the culture increased the RR for oocyte activation (14 %) and fertilization (37 %) but did not benefit cleavage, blastocyst formation, or blastomere count. The subgroup analyses revealed that supplementation with thiol compounds was indeed beneficial. Our network meta-analysis also supported the findings of classical meta-analyses showing that cysteine, cysteamine, epidermal growth factor, amino acid supplementation in maturation and culture media, and Triton treatment of sperm improved blastocyst formation. The overall success rate of live births from total embryos transferred after ICSI was not greater than 2 %. Although, manipulations that were beneficial for ICSI outcomes were identified in this meta-analysis, however, areas where more robust data are needed to reach a conclusive decision are highlighted.
