Secnidazole, a second-generation nitroimidazole, is widely used to treat various infections, including bacterial vaginosis, amoebiasis, and giardiasis, particularly in the treatment of female genital infections. Herein, a smartphone-based sensing platform has been developed in conjunction with a microfluidic-based paper device, enabling the visual, quick, sensitive, and quantitative analysis of secnidazole in real-time through fluorescence "Turn off". In addition to its high sensitivity for label-free and on-site detection of secnidazole, the proposed sensing device demonstrated a prominent anti-interference capability, ensuring accurate on-site identification. The N-CDs@WS₂-based fluorescent sensor exhibited a limit of detection (LOD) of 0.117 μM (117 nM) and a limit of quantification (LOQ) of 0.3573 μM (357 nM) for secnidazole. The portable sensor effectively enabled real-time monitoring of secnidazole in serum samples, yielding encouraging outcomes and paving the way for its accurate on-site detection. The detection efficiency of the sensor is validated by the World Health Organization (WHO) recommended LC-MS and UPLC methods. As indicated by the recoveries from UPLC, LC-MS, and N-CDs@WS₂-based sensing device, the proposed sensor can quantitaively analyze secnidazole in real samples.

Preparation and characterization of a secnidazole-mandelic acid coamorphous system: enhanced dissolution profile, antibacterial properties, and theoretical druglikeness assessment

This work describes the successful preparation of the first secnidazole (SNZ) coamorphous system using mandelic acid (MND) as a coformer via the slow solvent evaporation method, aiming to develop improved antimicrobial dosage forms. The coamorphous nature of the SNZ-MND system was confirmed by powder X-ray diffraction (PXRD), which revealed that the 1:1 ratio remained stable and amorphous for over 180 days. Its hygroscopicity and the structural, spectroscopic, and thermal properties were thoroughly investigated, supported by computational calculations, including Hirshfeld surface analysis, density functional theory (DFT), and in silico pharmacokinetic simulations. DFT calculations in the gas phase and ethanol provided insights into the thermodynamic, geometric, electronic, and vibrational parameters of the SNZ-MND heterodimer. The optimized geometry indicated that a hydrogen bond between the imidazole ring of SNZ and the carboxylic group of MND is a key stabilizing interaction. Thermodynamic data confirmed the spontaneity of this interaction, with an interaction energy of -2.02 kcal/mol in the gas phase. Vibrational modes were securely assigned by correlating experimental and calculated infrared spectra. Thermoanalytical analysis revealed a glass transition temperature of 106.5 °C, highlighting the system's thermostructural stability. Dissolution studies in phosphate buffer (pH 6.80) showed that the SNZ-MND coamorphous system exhibits a 1.22-fold higher dissolution rate than crystalline SNZ, a critical factor for bioavailability. The final plateau concentrations reflected the experimental setup under sink conditions. Similarly, apparent solubility measurements indicated a 22 % (39.5 ± 1.2 mg/mL) increase in dissolved concentration compared to the crystalline form of the drug. Importantly, the SNZ-MND (1:1) coamorphous system exhibited enhanced antibacterial activity against Staphylococcus aureus, Klebsiella pneumoniae, and Pseudomonas aeruginosa compared to the isolated compounds. These findings underscore the potential of the newly developed SNZ-MND coamorphous system as a promising antimicrobial binary composition with improved physicochemical and biological properties to be used in pharmaceutical dosage forms.

The residues of nitroimidazoles (NIIMs) in pork incurred by their overuse in feed pose a potential threat to human health.Therefore, it is necessary to establish effective anal. methods for the determination of them in pork.In this study, a carboxyl-functionalized three-dimensional conjugated microporous polymer (Try/BPDA(1:3)-CMP) was prepared with triptycene and 3,3'4,4'-biphenyl tetracarboxylic dianhydride as monomers by a simple Friedel-Crafts reaction.A good adsorption property of the polymer for the NIIMs was achieved probably via π-stacking, hydrogen boding and polar interactions.A simple method for the quantitation of NIIMs was established by using Try/BPDA(1:3)-CMP based solid-phase extraction combined with high performance liquid chromatog. detection.The method demonstrated a good performance and was successfully applied to the anal. of five NIIMs antibiotics in pork samples.Under the optimized conditions, the linear quantification response range for the analytes was 3.60-150 ng g^-1 with the determination coefficients larger than 0.9923, the limits of detection of the method for NIIMs at S/N = 3 were 1.20-1.60 ng g^-1, and the method recoveries for pork samples were in the range of 80.0%-118%, with the relative standard deviations between 2.1% and 12% (n = 5).
