The aim of our study is to investigate whether patients with moderate-to-severe VKC and keratitis will benefit from the addition of Diquafosol ophthalmic solution in terms of symptoms and signs of VKC, tear film metrics, ocular surface inflammation and quality of life. Potential beneficial effect of Diquafosol treatment on top of current standard use of topical immunosuppressant therapy in VKC would be studied.
Subject at least aged 6 and above with clinical diagnosis of vernal keratoconjunctivitis would be invited to join this study and perform the following assessments:
1. best corrected visual acuity,
2. slit lamp biomicroscope examination,
3. tear meniscus height and non-invasive keratographic tear breakup time(NIKBUT) measurement,
4. cornea fluorescein staining (CFS) and
5. Schirmer's I test. Doctor will prescribe eyedrop, 3% Diquafosol ophthalmic solution and 0.1% Cyclosporin A cationic ophthalmic emulsion or 0.1% Cyclosporin A cationic ophthalmic emulsion at the end of examination. Subjects will be followed up at 4 weeks, 8 weeks, 16 weeks after treatment.
All participants will be required to answer a standardised questionnaire relating to severity of VKC and dry eye disease.

Effects of 3% Diquafosol on Tear Film and Vision-related Quality of Life in Orthokeratology Lens Related Dry Eye

Investigate the effect of 3% Diquafosol on tear film and vision-related quality of life in dry eye patients wearing orthokeratology lenses(OK lens), and to provide reference for clinical treatment.This prospective, open label study will include 60 eyes of 30 OK lens related dry eye patients.Participants will receive 3% Diquafosol ophthalmic solution. Modes of Binding of Small Molecules Dictate the Interruption of RBD‐ACE2 Complex of SARS‐CoV‐2

AbstractThe spike protein is a vital target for therapeutic advancement to inhibit viral entrance. Given that the connection between Spike and ACE2 constitutes the initial phase of SARS‐CoV‐2 pathogenesis, obstructing this interaction presents a promising therapeutic approach. This work aims to find compounds from DrugBank that can modulate the stability of the spike RBD‐ACE2 protein‐protein complex. Employing a therapeutic repurposing strategy, we conducted molecular docking of over 9000 DrugBank compounds against the Spike RBD‐ACE2 complex, on ten variants, including the wild‐type. We also evaluated the intricate stability of the RBD‐ACE2 proteins by molecular dynamics simulations, hydrogen bond analysis, RMSD analysis, radius of gyration analysis, and the QM‐MM approach. We assessed the efficacy of the top ten candidates for each variant as an inhibitor. Our findings demonstrated for the first time that DrugBank small molecules can interact in three distinct modalities inside the extensive protein‐protein interface of RBD and ACE2 complexes. The top ten analyses identified specific DrugBank candidates for each variant and molecules capable of binding to multiple variants. This comprehensive computational technique enables the screening and forecasting of hits for any big and shallow protein‐protein interface drug targets.

AbstractPurposeTo investigate temporal changes in parameters associated with tear film stability after instillation of long-acting 3% diquafosol ophthalmic solution (DQS LX), which contains polyvinylpyrrolidone.Study DesignProspective crossover study design.MethodsWe enrolled 23 eyes of 23 soft contact lens (SCL) wearers (aged 25.3 ± 4.4 years). One-day disposable silicone hydrogel lenses (narafilcon A) were used in this study. DQS LX was instilled without a SCL on the first day. On the other two days, subjects received DQS or DQS LX at 7 h after wearing SCLs. Tear meniscus height (TMH), non-invasive tear break-up time (NIBUT) and ocular surface temperature (OST) were measured before and at 5, 15, 30, 45, 60, 80 and 120 min after instillation.ResultsWhen not wearing SCLs, DQS LX instillation significantly improved TMH for up to 120 min and NIBUT for up to 80 min. When wearing SCLs, DQS and DQS LX instillation significantly increased TMH for up to 45 and 80 min, and NIBUT for up to 15 and 30 min. Compared to DQS, DQS LX administration resulted in a significantly higher TMH at 5, 60 and 80 min, as well as a significantly prolonged NIBUT at 5 and 60 min. OST with and without SCLs also varied depending on the changes in the parameters associated with tear film stability.ConclusionCompared to DQS, when wearing SCLs, DQS LX was found to increase the amount of tear fluid and improve tear film stability for longer periods.

Glia-like taste cells mediate an intercellular mode of peripheral sweet adaptation

The sense of taste generally shows diminishing sensitivity to prolonged sweet stimuli, referred to as sweet adaptation. Yet, its mechanistic landscape remains incomplete. Here, we report that glia-like type I cells provide a distinct mode of sweet adaptation via intercellular crosstalk with chemosensory type II cells. Using the microfluidic-based intravital tongue imaging system, we found that sweet adaptation is facilitated along the synaptic transduction from type II cells to gustatory afferent nerves, while type I cells display temporally delayed and prolonged activities. We identified that type I cells receive purinergic input from adjacent type II cells via P2RY2 and provide inhibitory feedback to the synaptic transduction of sweet taste. Aligning with our cellular-level findings, purinergic activation of type I cells attenuated sweet licking behavior, and P2RY2 knockout mice showed decelerated adaptation behavior. Our study highlights a veiled intercellular mode of sweet adaptation, potentially contributing to the efficient encoding of prolonged sweetness.

Yuyu Pharma, Dry Eye Pipeline Mechanism of Action and Pre-clinical Results Published

SEOUL, South Korea, April 27, 2023 /PRNewswire/ -- At ARVO 2023, Yuyu Pharma unveiled the results of its mechanism of action and head-to- head studies for its dry eye treatment peptide (project name: YP-P10). ARVO is the world's largest ophthalmic society and held its annual meeting in New Orleans, Louisiana, USA in April of this year. A study conducted by Dr. Virginia Calder of the University College London (UCL) found that YP-P10 significantly reduces inflammatory Th2 and Th17 cells. In particular, the YP-P10 model reduced Th17 cells 25% more effectively when compared to the control group. The YP-P10 pre-clinical head-to-head study, jointly conducted by five people including Dr. Kyungho Park of Naason Science, showed statistically significant reduction of corneal damage by measuring Corneal Fluorescein Staining (CFS). The study compared the efficacy of YP-P10 to that of several competing dry eye treatment products such as Lifitegrast, Cyclosporine, Diquafosol, and Hyaluronic Acid. The results showed that YP-P10 had 12% better efficacy than Lifitegrast, 7% better than Cyclosporine, 9 % better than Hyaluronic Acid, and showed similar efficacy to Diquafosol.
