Alkaline hydrolysis-UV activation strategy for rapid separation and screening of methyl salicylate in complex samples

BACKGROUND:

Methyl salicylate is an important volatile biomarker related to plant stress responses and is also widely used as an active ingredient in analgesic products. Rapid screening of methyl salicylate in complex samples is therefore valuable for agricultural monitoring and product quality assessment. In this study, a portable analytical method integrating alkaline hydrolysis, ultraviolet activation, gas membrane separation, and colorimetric derivatization was developed for methyl salicylate detection.

RESULTS:

Methyl salicylate was hydrolyzed under alkaline conditions to generate methanol, which was subsequently oxidized by titanium dioxide under ultraviolet irradiation to produce formaldehyde. The generated formaldehyde was enriched through a gas membrane and derivatized with acetylacetone to form a yellow chromophore for ultraviolet-visible spectrophotometric quantification. Under optimized conditions, the method showed a linear response from 20 to 200 μg/mL, with a correlation coefficient of 0.9962 and a detection limit of 10.0 μg/mL. Recovery values were 105.1-113.0% for medical adhesive bandages and 85.5-115.0% for plant samples. Matrix blank tests showed no detectable background signal in the investigated real samples.

SIGNIFICANCE AND NOVELTY:

The proposed method converts methyl salicylate into a detectable volatile derivative through a cascade reaction and combines chemical transformation with gas membrane enrichment in a portable workflow. Compared with conventional high-performance liquid chromatography, this method requires simpler instrumentation and is more suitable for rapid on-site screening. It provides a practical and field-deployable strategy for methyl salicylate analysis in medical products and plant samples.

Perceptual interactions and predicted mechanism in binary odorant mixtures of baked green tea (Camellia sinensis cv. Zhongcha 302)

INTRODUCTION:

Tea is globally prized for its flavor, with aroma being a key quality indicator. To fully understand tea aroma, which arises from complex perceptual interactions, systematic molecular sensory omics that integrate both perceptual complexity and matrix effects are required. This critical integration continues to be unmet as a key challenge in the field.

OBJECTIVES:

Baked green tea produced from 'Zhongcha 302' (Camellia sinensis cv. Zhongcha 302, ZC302) is prized for its superior flavor, yet its key odorants and their formation mechanisms remain unclear. This study aimed to identify these key odorants and elucidate the perceptual interactions and predicted mechanism in binary mixtures.

METHODS:

Key odorants were identified through an integrated sensomics approach. Perceptual interactions between binary odorant pairs were evaluated using 3‑AFC tests, S‑curves, and odor activity value calculations, with further validation conducted in tea blank matrix. Molecular docking and molecular dynamics simulation were performed to characterize odorant binding to olfactory receptors and predict binding modes.

RESULTS:

Seven key odorants yielded 21 binary pairs, classified as 13 synergistic, six masking, one additive, and one non-interactive. The additive (geraniol & methyl salicylate) and masking (geraniol &trans-β-ionone) pairs consistently modulated the "floral" attribute in both aqueous and tea matrix. Molecular docking and molecular dynamics simulations demonstrated that the additive odor pair forms a highly compatible and stable complex with OR1G1 via strengthened hydrogen bonding, hydrophobic interactions, and favorable electrostatic complementarity. In this binary odorant system, methyl salicylate stably anchors the receptor extracellular domain, whereas geraniol dynamically modulates receptor structural flexibility.

CONCLUSION:

The key odorants in ZC302 baked green tea primarily interact through additive or masking effects, with their perceptual outcomes governed by distinct binding modes to olfactory receptors. The tea matrix critically modulates odor interaction patterns and substantially affects the authenticity of aroma perception.

Functional characterization of CaspOR20 involved in repellency of 1-penten-3-ol, sulcatone, and methyl salicylate against the rape stem weevil Ceutorhynchus asper

The rape stem weevil (Ceutorhynchus asper) is a devastating pest of cruciferous crops, and its management heavily relies on insecticides, leading to resistance development and environmental concerns. CaspOR20 is a highly expressed odorant receptor in the antennae of C. asper, yet its functional role remains unclear. In this study, by using the transgenic Drosophila expression system combined with single sensillum recordings, we identified six volatile compounds (1-penten-3-ol, 2-heptanone, sulcatone, methyl salicylate, benzaldehyde, and ethyl acetate) as ligands for CaspOR20. Subsequent electroantennography (EAG) assays revealed that all of these ligands except ethyl acetate elicited significant electrophysiological responses in both male and female C. asper adults. Behavioral assays using an H-tube olfactometer demonstrated that three compounds (1-penten-3-ol, sulcatone, and methyl salicylate) exhibited significant repellent effects on both male and female C. asper. Molecular docking analysis indicated strong binding affinity between CaspOR20 and the three repellent compounds, with key residues (CYS-129, ALA-176, and ASN-179) involved in hydrogen bond interactions. Functional validation via RNA interference targeting CaspOR20 in vivo indicated that CaspOR20-silenced individuals displayed significantly reduced EAG responses to the repellent compounds and lost their behavioral avoidance. This study successfully identified repellent compounds against C. asper, and provided insights into the molecular mechanism underlying C. asper's detection of these compounds. Our findings established a solid foundation for the future development of effective repellents and eco-friendly control strategies against this pest.

Paracetamol to PAN-D: CDSCO finds over 50 sub-standard drugs. Check full list of medicines

New Delhi: The Central Drugs Standard Control Organisation ( CDSCO ) has identified over 50 drugs which have failed in the drug test. In its latest report, the controller said 50 drugs including Paracetamol are "not of standard quality" (NSQ). The report has also flagged calcium and vitamin D3 supplements, anti-diabetes pills, and high blood pressure medicines. Paracetamol tablets from Karnataka Antibiotics & Pharmaceuticals Ltd were specifically mentioned for quality concerns. The report listed a total of 48 drugs in the NSQ category, including a Gauze Roll Non Sterile Roller Bandage. NSQ alerts come from monthly random sampling conducted by state drug officers. Some of the medicines were produced by companies like Hetero Drugs, Alkem Laboratories, Hindustan Antibiotics Limited (HAL), Karnataka Antibiotics & Pharmaceuticals Ltd, Meg Lifesciences, and Pure & Cure Healthcare. Additionally, the CDSCO shared another list that features five more drugs that failed quality tests. The pharmaceutical companies denied responsibility for these drugs. They claim they are "spurious," and one response from a company stated, "The actual manufacturer (as per label claim) has informed that the impugned batch of the product has not been manufactured by them and that it is a spurious drug. The product is purported to be spurious; however, the same is subjected to outcome of investigation." Govt bans 156 medicines including antibiotics, painkillers, multivitamins: Here is the full list

New Delhi: In a significant move aimed at safeguarding public health, the Indian government has banned 156 fixed-dose combination (FDC) drugs, including a variety of antibiotics, painkillers, multivitamins, and medications for treating fever and hypertension. This decision marks the largest crackdown on FDCs since 2016, when 344 such drugs were prohibited. The Union Ministry of Health and Family Welfare issued a gazette notification on Thursday, officially prohibiting the manufacture, sale, and distribution of the banned FDCs. The ban was based on the recommendations of an expert panel that reviewed a total of 324 FDCs. Impact on Pharmaceutical Industry The banned FDCs include popular combinations such as mefenamic acid and paracetamol injections, commonly used for pain relief and fever, and omeprazole magnesium with dicyclomine HCl, used to treat abdominal pain.
