Testosterone Supplementation in Patients in Best Supportive Care: Impact on Quality of Life

This is a monocentric, single-arm prospective pilot study that will enrol hypogonadal (testosterone < 231 ng/dL) male subjects in best supportive care, with no further therapeutic options and no need for resuscitation.
Currently, testosterone formulations for intramuscular (IM) injection and subcutaneous injection as well as for oral and transdermal administration have been approved for androgen therapy. To date, injectable testosterone is the most commonly used formulation. To increase serum testosterone levels to the physiological range IM injections of testosterone every 2-3 weeks are required, which lead to supraphysiological peaks shortly after administration, followed by a sharp fall in levels thereafter. Testosterone levels before the next injection are frequently in the hypogonadal range. For this reason, Sustanon 250® (1 ml, IM) will be administered on day 0 after confirmation of hypogonadism by blood test and then every 15 days in this trial given the limited life expectancy of the subjects and to maximise the effect and benefit of testosterone supplementation.
The Edmonton questionnaire and ADL questionnaire will be completed before the injection on day 0 and then every 15 days at the time of injection until subject's death or if a subject is discontinued from the study treatment/procedures for any other reasons than death. In parallel, the EQ-5D-3L questionnaire will be completed by a family member or a proxy.

Pharmacokinetic, Clinical, Hormonal and Therapeutic Effects of the Use of Estradiol and Testosterone Hormonal Subdermal Implants in Women With Natural Menopause, Premature Ovarian Failure or Surgical Menopause Due to Cervical Cancer.

Estrogen and androgen deficiencies negatively impact the quality of life of women at different stages of life, especially after menopause. New modalities and new therapeutic alternatives have been researched. Parenteral administration of estradiol and testosterone could be effective to treat symptoms secondary to estrogen and androgen deficiencies and minimize these adverse events. This study evaluates the efficiency of subdermal implant-bioabsorbable use in women with menopausal symptoms associated with secondary estrogen and androgen deficiencies in women with natural menopause, premature ovarian failure or surgical menopause due to cervical cancer. Pharmacokinetic, biochemical, metabolic, thromboembolic and hormonal data will be evaluated, as well as the effects on quality of life, menopausal symptoms and sexual function after treatment.

Physicians who treat children with idiopathic short stature (ISS) are challenged in deciding whether the intervention to improve final height is truly efficacious, given the heterogeneity of the diagnosis of ISS. There are numerous studies and reviews that have attempted to assess and demonstrate the benefit of various interventions, such as growth hormone therapy and aromatase inhibitors. Recently, Zhang et al. published a well-written meta-analysis entitled "Comparative effects of growth hormone, testosterone, and aromatase inhibitors on height gain in children and adolescents with idiopathic short statures: a network meta-analysis," attempting to stratify the benefit of several interventions. Our commentary seeks to highlight the importance of more research needed in this field and the complexities of treating children with ISS.

Elevated serum uric acid (SUA) has been linked to adverse outcomes in polycystic ovary syndrome, yet its broader role in female fertility is unclear. This retrospective cohort study of 16,223 women undergoing a first in vitro fertilisation (IVF)/intracytoplasmic sperm injection (ICSI) cycle investigated associations between pre-treatment SUA and reproductive parameters. Higher SUA levels were independently associated with elevated luteinizing hormone (LH), testosterone, and anti-Müllerian hormone (AMH), and with lower estradiol and follicle-stimulating hormone. Each 50 µmol/L SUA increase correlated with a 16-gram reduction in neonatal birth weight and a 0.06-week decrease in gestational age, with a threshold effect near 350 µmol/L. Bidirectional Mendelian randomisation analyses supported a causal effect of elevated SUA on increased LH and testosterone, while reverse analysis suggested lower AMH may elevate SUA. These findings identify SUA as a biomarker influencing ovarian reserve and neonatal outcomes through bidirectional mechanisms, including a potential SUA-AMH feedback loop. Routine SUA monitoring in fertility treatment is supported, and interventions for individuals with SUA ≥350 µmol/L warrant clinical investigation.

Testosterone (TES) has complex roles in cardiovascular disease, influencing not only atherosclerosis development in general, but also atherosclerosis-related processes associated with hypertension, cholesterol metabolism, vascular calcification, and arterial stiffness. This review examines TES's effects, particularly its atheroprotective role in various mice and minipig model systems and cell cultures. TES modulates the renin-angiotensin system (RAS), contributing to hypertension and vascular dysfunction, but its deprivation can mitigate these effects. TES also impacts cholesterol metabolism by regulating liver X receptor (LXRα) pathways, promoting both cholesterol clearance and synthesis. Moreover, TES is involved in vascular calcification via androgen receptor (AR) signalling, a process that contributes to arterial stiffness, especially in females. The review highlights gaps in understanding TES's specific molecular mechanisms in cardiovascular disease, emphasising the need for further research to explore sex-specific responses and potential therapeutic interventions.

Rapid and accurate method from VTT for detecting hormones, toxins and other small molecules

ESPOO, Finland, Feb. 24, 2026 /PRNewswire/ -- In the field of diagnostics, there has long been a search for solutions to measure low concentrations of small molecules accurately, quickly and easily, for example from food, the environment or a patient sample. Immunocomplex antibody technology offers several advantages compared to conventional methods. Typically, laboratories use the so-called sandwich method utilizing two antibodies to measure large molecules, such as proteins. Many environmental toxins, pharmaceuticals, and hormones are so small molecules that only one antibody can bind at a time. As a result, detecting these small molecules has traditionally required a multi-step, slow and imprecise method. VTT has developed an immunocomplex antibody-based method that is suitable for the precise and rapid detection of small molecules. Innovative solution for rapid detection of small molecules A distinctive feature of immunocomplex antibodies is that they do not bind to the small target molecule or to the antibody that recognises the target molecule. Instead, they bind to the pair they form together - this combination is called the immunocomplex. The mechanism works because, when the first antibody binds to the small target molecule, it causes a slight structural change, which the second immunocomplex antibody is able to recognise. Technology offers several advantages that have enabled the use of immunocomplex antibodies in rapid diagnostics: It provides greater specificity and sensitivity than other conventional methods on the market and has the ability to detect both low and high concentrations of the target molecule. In addition, result can be measured within a few minutes after sample addition. At VTT, the antibodies are screened from vast antibody libraries, enabling the identification of rare immunocomplex-specific antibodies with exceptional selectivity. Testosterone measurement with LFA rapid test "There is a continuously growing global need for metabolic health monitoring.
