This is a Phase 2 dose-titration study designed to evaluate the safety and efficacy of MANP subcutaneous injection compared to placebo in reducing baseline daytime systolic blood pressure (SBP), derived from 24-hour ambulatory blood pressure monitoring (ABPM), in subjects with hypertension who are taking 3 or more antihypertensive medications with different mechanisms of action.

A Phase 1B, Multiple Ascending Dose Trial Examining The Safety And Tolerability Of Subcutaneous MANP In Difficult To Control/Resistant Hypertensive Subjects

A randomized, double-blind, placebo-controlled multiple ascending dose study in hypertensive subjects on stable doses of at least three hypertensive drugs for at least 6 weeks prior to Screening. The study will consist of screening, PK-unit admittance, and safety follow up periods.
Subjects will be randomized at a 6:2 ratio of either MANP or placebo and will be stratified by race in each dosage cohort. The entire first Cohort will be given the lowest dosage with subsequent cohorts progressing sequentially to the higher doses depending on safety and tolerability of the previous cohort.
Endpoints not related to the safety reviews will be analyzed after the last patient last visit (LPLV).

Multifunctional type lll recombinant human collagen incorporated sodium alginate hydrogel with sustained release of extra cellular vehicles for wound healing multimodal therapy in diabetic mice

The effective promotion of wound healing poses a substantial challenge for clinical treatment. Despite evidence supporting the role of extracellular vesicles (EVs) in this process, their therapeutic potential is currently restrict by challenges in targeting and maintaining them. The manufacturing process for rhCol III, or recombinant human collagen III, is stable, and the rejection rate is low. We used a cross-linking method to prepare a rhCol III incorporated sodium alginate (SA) hydrogel, which enabled to accomplish an EV sustained release that was site-specific. Cell viability through MTT assay, proliferation and ROS generation were performed with MC3T3-E1cell lines. In addition, diabetic wounds are characterised by an environment of hyper-inflammation and elevated oxidative stress. The rhCol III/SA-EVs hydrogel, which is a delivery vehicle with anti-inflammatory and antioxidant characteristics, promotes wound healing in this setting. The In vivo effectiveness of the created wound dressing on a diabetic wound model was examined in this study. After 21 days of treatment, the wound dressing significantly (p < 0.05) expedited wound healing compared to the control group, and wound closure was approximately 95% without any negative systemic reactions.

The aim of the study was to develop an oral targeting drug delivery system (OTDDS) of oxymatrine (OMT) to effectively treat ulcerative colitis (UC). The OTDDS of OMT (OMT/SA-NPs) was constructed with OMT, pectin, Ca²⁺, chitosan (CS) and sialic acid (SA). The obtained particles were characterized in terms of particle size, zeta potential, morphology, drug loading, encapsulation efficiency, drug release and stability. The average size of OMT/SA-NPs was 255.0 nm with a zeta potential of -12.4 mV. The loading content and encapsulation efficiency of OMT/SA-NPs were 14.65% and 84.83%, respectively. The particle size of OMT/SA-NPs changed slightly in the gastrointestinal tract. The nanoparticles can delivery most of the drug to the colon region. In vitro cell experiments showed that the SA-NPs had excellent biocompatibility and anti-inflammation, and the uptake of SA-NPs by RAW 264.7 cells was time and concentration-dependent. The conjugated SA can help the internalization of NPs into target cells. In vivo experiments showed that OMT/SA-NPs had a superior anti-inflammation effect and the effect of reducing UC, which was attributed to the delivery most of OMT to the colonic lumen, the specific targeting and retention in colitis site and the combined anti-inflammation of OMT and NPs.

APTES-mediated Cu2(OH)3(NO3) nanomaterials on the surface of silicone catheters for abscess

Metal-based nanomaterials have remarkable bactericidal effects; however, their toxicity cannot be disregarded. To address this concern, we developed a simple synthesis route for antibacterial catheters using metal-based nanomaterials to reduce toxicity while harnessing their excellent bactericidal properties. The grafting agent (3-aminopropyl)triethoxysilane (APTES) forms -NH₂ groups on the catheter surface, onto which copper ions form a nanomaterial complex known as Cu₂(OH)₃(NO₃) (defined as SA-Cu). The synthesized SA-Cu exhibited outstanding contact antibacterial effects, as observed through scanning electron microscopy (SEM), which revealed cell membrane crumbing and bacterial rupture on the catheter surface. Furthermore, SA-Cu exhibited excellent biosafety characteristics, as evidenced by the cell counting kit-8 (CCK-8) assay, which showed no significant cytotoxicity. SA-Cu demonstrated sustained antimicrobial capacity, with in vivo experiments demonstrating over 99% bactericidal efficacy against methicillin-resistant Staphylococcus aureus (MRSA) for two weeks. The transcriptome sequencing results suggested that SA-Cu may exert its bactericidal effects by interfering with histidine and purine metabolism in MRSA. This study presents a straightforward method for synthesizing antimicrobial silicone catheters containing copper nanomaterials using copper ions.
