Primary objective: To evaluate the efficacy and safety of fluocinolone cream in the treatment of moderate to severe chloasma. Secondary objective: To evaluate the clinical equivalence of fluocinolone cream and the reference preparation by clinical endpoint method; to clarify the systemic exposure of the test preparation and the reference preparation by monitoring the systemic absorption of the three components of retinoic acid, fluocinolone acetonide and hydroquinone in clinical trials.

Does Tri‐Staple™ technology reduce the risk of anastomotic leakage in colorectal surgery? A propensity score matching analysis

AbstractBackground and ObjectivesThis study evaluates the Tri‐Staple™ technology in colorectal anastomosis.MethodsPatients who underwent rectosigmoidectomy between 2016 and 2022 were retrospectively evaluated and divided into two groups: EEA™ (EEA) or Tri‐Staple™ (Tri‐EEA). The groups were matched for age, sex, American Society of Anesthesiologists (ASA), and neoadjuvant radiotherapy using propensity score matching (PSM).ResultThree hundred and thirty‐six patients were included (228 EEA; 108 Tri‐EEA). The groups were similar in sex, age, and neoadjuvant therapy. The Tri‐EEA group had fewer patients with ASA III/IV scores (7% vs. 33%; p < 0.001). The Tri‐EEA group had a lower incidence of leakage (4% vs. 11%; p = 0.023), reoperations (4% vs. 12%; p = 0.016), and severe complications (6% vs. 14%; p = 0.026). There was no difference in complications, mortality, readmission, and length of stay. After PSM, 108 patients in the EEA group were compared with 108 in the Tri‐EEA group. The covariates sex, age, neoadjuvant radiotherapy, and ASA were balanced, and the risk of leakage (4% vs. 12%; p = 0.04), reoperation (4% vs. 14%; p = 0.014), and severe complications (6% vs. 15%; p = 0.041) remained lower in the Tri‐EEA group.ConclusionTri‐Staple™ reduces the risk of leakage in colorectal anastomosis. However, this study provides only insights, and further research is warranted to confirm these findings.

The conditions for the preparation of pectic oligosaccharides (POS) by high hydrostatic pressure-assisted enzymatic (E-HHP) method were explored. The optimal conditions consisted of the pressure of 350 MPa for 20 min, and enzymolysis for 60 min with 0.011 U/mL enzyme. The products were isolated by ion exchange chromatography, galacturonic acid, di- and tri-galacturonides (Tri-GalA) with high purity were obtained. Additionally, the effects of POS on activation and degranulation of RBL-2H3 mast cells were investigated. It was found that Tri-GalA and POS could attenuate the release of β-hexosaminidase and histamine, reduce the production of IL-4 and inhibit the extracellular Ca²⁺ influx of RBL-2H3 cells. Notably, 150 μg/mL POS significantly alleviated the IgE-mediated allergic reaction of RBL-2H3 cells. These results indicate that POS could be used as an inhibitor in regulating mast cell-mediated allergic inflammatory responses.

Efficacy of a single administration of afoxolaner (NexGard®) or fipronil plus permethrin (Frontline® Tri-Act) against Hyalomma marginatum ticks in dogs

This study was conducted to assess the acaricidal efficacy of afoxolaner (NexGard®, Boehringer Ingelheim), and fipronil - permethrin (Frontline® Tri-Act, Boehringer Ingelheim) administered once to dogs experimentally infested with Hyalomma marginatum ticks. Twenty-four Beagle dogs were randomly allocated based on a pre-treatment H. marginatum infestation to an untreated control group, a NexGard® or a Frontline® Tri-Act treated groups. Treatments were administered once on Day 0 as per the products' labels. For the efficacy evaluation, dogs were experimentally infested with 30 adult H. marginatum ticks on Days -2, 7, 28 and 36. In-situ counts were performed at 48 h post-treatment on Day 2 and post-infestations on Days 9, 30 and 38. Ticks were removed and counted at 72 h post-treatment on Day 3 and after each tick infestation on Days 10, 31 and 39. The numbers of live ticks counted in the treated groups were significantly different than in the control group at all time-points (p ≤ 0.0006). The efficacy was at least 97% after 48 h, and at least 99% after 72 h for both treatments. In this study both afoxolaner and fipronil/permethrin formulations demonstrated a high efficacy against adult H. marginatum ticks in treated dogs for at least five weeks.
