Linear Force Scaling in Kinesin-Driven Microtubule Swarms Revealed by Electromagnetic Tweezers

Scalability refers to the ability of a system to enhance performance with increasing size. This ability is a defining feature of natural swarms such as ant and bee colonies. Extending this principle to artificial active matter has motivated the creation of molecular swarms composed of myosin-driven actin filaments and kinesin-driven microtubules (MTs). While these swarms exhibit cooperative transport behaviors that surpass the abilities of individual filaments, their collective force generation has remained unquantified. Here, we introduce a straightforward electromagnetic tweezer approach to directly measure the forces generated by MT swarms propelled by surface-bound kinesin motors. We find that force output changes with swarm size, demonstrating quantitative scalability characterized by a linear dependence of collective force on the inferred number of kinesin motors, linking collective organization to mechanical performance. These results characterize the scaling of collective force with swarm size in kinesin-driven MT swarms and highlight the link between collective organization and mechanical performance.

The Effect of Three Key Administrative Errors on Patient Trust in Physicians: Prescription Errors, Confidentiality Breaches, and Appointment Scheduling Omissions

ABSTRACT:

Background:

Understanding how administrative errors, such as prescription and appointment scheduling omissions, and patient confidentiality breaches impact trust in physicians is crucial for improving patient‐physician relationships and healthcare outcomes. To investigate the association between administrative errors, general trust in physicians, and interpersonal trust in a physician, we surveyed adults across Japan.

Methods:

Participants were adults aged ≥ 20 years who had received treatment at least twice for non‐communicable diseases within the past 6 months. The exposure variables were past experiences with prescription errors, confidentiality breaches, and appointment scheduling omissions by personal physicians treating their non‐communicable diseases. General trust and interpersonal trust in a physician were measured using the Japanese version of the Wake Forest Physician Trust Scale.

Results:

Among the 661 participants, nearly 14% reported experiencing at least one type of administrative error. Prescription errors were associated with a significant decrease in general trust in physicians (−9.78 points, 95% confidence interval [CI]: −13.74 to −5.81). Confidentiality breaches had the most significant negative impact on interpersonal trust (−14.09 points, 95% CI: −24.35 to −3.83), followed by appointment scheduling omissions (−13.56 points, 95% CI: −22.48 to −4.65). Mediation analysis revealed that the association between prescription errors and reduced general trust was partially mediated by decreased trust in personal physicians.

Conclusions:

Administrative errors during care for non‐communicable diseases significantly undermine patients' trust in physicians. Physicians should prioritize improving their practices, particularly regarding prescription errors, as these errors have broader implications for the public's perception of physicians.

Development and evaluation of a self-assembled nanoparticle-based prodrug for sustained delivery of 4-phenylbutyric acid

4-Phenylbutyric acid (PBA) is a small molecule with promising therapeutic potential for treating various diseases, including cancer and neurodegenerative disorders, due to its dual ability to reduce endoplasmic reticulum stress and inhibit histone deacetylases. However, its clinical application is hindered by rapid clearance from the body, necessitating frequent dosing that increases the risk of adverse effects. To address these limitations, we developed a nanoparticle-based prodrug (Nano^PBA) utilizing the amphiphilic block copolymer poly(ethylene glycol)-b-poly(vinyl 4-phenylbutyrate) [PEG-b-P(VPBA)]. This system self-assembles into micelles, enabling controlled and sustained PBA delivery. The synthesis and characterization of Nano^PBA revealed its high stability under physiological conditions and enzyme-responsive PBA release. Nano^PBA demonstrated a controlled release profile in vitro, reducing burst release while maintaining therapeutic efficacy. Cytotoxicity assays using normal cell lines, including endothelial cells (BAEC), macrophages (RAW264.7), and rat gastric cells (RGM-1), showed minimal cytotoxic effects compared to the parent low-molecular-weight PBA. Furthermore, in vivo studies conducted in healthy C57BL/6J mice confirmed Nano^PBA's biocompatibility, with no significant adverse effects observed at therapeutic doses ranging from 200 to 500 mg-PBA/kg via oral administration. In conclusion, Nano^PBA offers a controlled release profile, enhanced biocompatibility, and reduced toxicity, addressing the limitations associated with conventional PBA administration. These attributes make Nano^PBA a promising candidate for improving the therapeutic efficacy and safety of PBA in clinical applications, particularly in diseases where maintaining consistent drug levels is crucial for treatment outcomes.
