In oncology, in September 2025, the U.S. Food and Drug Administration (FDA) approved KEYTRUDA QLEX injection for subcutaneous (SC) administration for use in adults across most solid tumor indications for KEYTRUDA, based on results from the Phase 3 MK-3475A-D77 trial. In October 2025, the FDA subsequently approved KEYTRUDA QLEX for the treatment of certain adult patients with resectable locally advanced head and neck squamous cell carcinoma (LA-HNSCC), based on results from the Phase 3 KEYNOTE-689 trial. KEYTRUDA QLEX is now approved for use in adults across all solid tumor indications approved for KEYTRUDA and is the first and only subcutaneously administered immune checkpoint inhibitor that can be given by a health care provider in as little as one minute. In addition, the European Medicines Agency’s Committee for Medicinal Products for Human Use (CHMP) adopted a positive opinion recommending approval for SC administration of KEYTRUDA for all adult indications; a final decision is expected in the fourth quarter of 2025. The European Commission (EC) also approved KEYTRUDA as part of a perioperative regimen for the treatment of PD-L1+ resectable LA-HNSCC. At the European Society for Medical Oncology (ESMO) Congress 2025, the Company announced new research from its broad and differentiated portfolio and pipeline, highlighting progress in new tumor types and earlier stages of disease. This included positive results from the Phase 3 KEYNOTE-905 trial (also known as EV-303) in cisplatin-ineligible patients with muscle-invasive bladder cancer (MIBC), the Phase 3 KEYNOTE-B96 trial in platinum-resistant recurrent ovarian cancer and the Phase 2/3 REJOICE-Ovarian01 trial in collaboration with Daiichi Sankyo in certain types of platinum-resistant ovarian cancer, long-term follow-up data from the Phase 3 KEYNOTE-775 trial in advanced endometrial cancer, as well as long-term data for KEYTRUDA in both earlier-stage and metastatic NSCLC. In vaccines and infectious diseases, in August 2025, the Company received two approvals in Japan: its nine-valent HPV vaccine for use in males ages 9 and older that will be marketed under the trademark SILGARD 9, and CAPVAXIVE for use in the elderly or adults who are at an increased risk of pneumococcal disease. At the European AIDS Clinical Society 2025 conference, the Company also presented new data from Phase 3 trials evaluating the once-daily, oral, two-drug regimen of doravirine/islatravir in adults with virologically suppressed HIV-1 infection, which showed minimal changes in weight and body composition and no clinically meaningful effect on fasting lipids and the homeostatic model assessment of insulin resistance across both clinical trials. In cardiovascular disease, the Company announced positive topline results from the Phase 3 CORALreef Lipids trial evaluating the safety and efficacy of enlicitide decanoate, an investigational, once-daily oral proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitor being evaluated for the treatment of adults with hypercholesterolemia. The trial met all primary and key secondary endpoints. Enlicitide has the potential to be the first approved oral PCSK9 inhibitor. This revised non-GAAP EPS outlook reflects several items not previously included, such as a benefit from an amended collaboration agreement with AstraZeneca related to Koselugo, and operational improvements, including a more favorable estimated tax rate and lower estimated costs related to the impact of tariffs, partially offset by an estimated negative impact related to the acquisition of Verona Pharma and an incremental negative impact from foreign exchange. The midpoint of this revised non-GAAP EPS range reflects a net improvement of $0.04 per share compared to the midpoint of the Company’s prior outlook. US-based startup Bambusa Therapeutics has secured $90m in Series A financing to advance its pipeline of bispecific antibodies for immunological and inflammatory disorders into the clinic. The funding round was led by RA Capital Management, with participation from Janus Henderson Investors, Redmile Group, Invus, and ADAR1 Capital Management, as well as existing investors. Bambusa was launched in May 2024 and previously raised $15m in seed financing, which closed in September 2024. That round was co-led by BVF Partners and KKR’s Dawn Biopharma, with additional backing from Salvia GmbH and INCE Capital. The company is being led by founder and CEO Shanshan Xu, who was previously vice president for external innovations at BioNTech. The company’s pipeline includes four bispecific antibody candidates, with two programmes – BBT001 and BBT002 – set to enter the clinic later on in 2025. BBT001 is being investigated for dermatologic indications and is expected to enter a Phase I trial (NCT06808477) by the end of March. The trial aims to enrol 98 healthy volunteers and adult patients with atopic dermatitis. BBT002, designed for respiratory, dermatology, and gastroenterology indications, has completed preclinical studies and is slated to enter human trials by mid-2025. Bambusa describes BBT002 as a “platform in a molecule,” with potential applications across multiple inflammatory disease areas. Bambusa’s additional candidates, BBT003 and BBT004, are in earlier stages of development. BBT003 is being developed for inflammatory bowel disease, while BBT004 is focused on rheumatology. The company has not shared timelines for these programmes but describes them as having “best-in-disease” potential. In the announcement accompanying the funding, RA Capital partner Derek DiRocco said: “The next wave of therapeutics for patients with immunology and inflammation disorders will be both more efficacious and convenient than currently available options. “The Bambusa Therapeutics pipeline fits this profile, as their half-life extended bispecific antibodies were designed to target complementary disease-driving signalling pathways, affording the potential for a best-in-disease profile for patients.” The bispecific antibody market is one that is already crowded. It has seen increasing investment from pharmaceutical companies looking to expand their immunology and inflammation portfolios. In August 2024, MSD announced plans to acquire a CD3 and CD19-targeting T cell engager bispecific antibody, CN201, from Curon Biopharmaceutical. The deal is worth approximately $1.3bn in upfront and milestone-based payments. In May 2024, Johnson & Johnson (J&J) paid $1.25bn to acquire global rights for Numab Therapeutics’ IL-4R and IL-31-targeting bispecific antibody.

SAN DIEGO — AstraZeneca is getting ready to step on the gas in the world of bispecific T cell engagers. The company presented some of its first Phase 1 data for its CD3xCD19 bispecific known as AZD0486 — which is being tested in multiple B cell malignancies — at the American Society of Hematology’s annual meeting this weekend. Data come from a larger study where AstraZeneca is testing the program against a broad range of blood cancers, where the company tested four dose levels. Researchers touted results in later-line follicular lymphoma and diffuse large B cell lymphoma (DLBCL). In 27 patients with follicular lymphoma who received any of the three highest doses, AZD0486 induced a 96% objective response rate and an 85% complete response rate, AstraZeneca said. Additionally, in the two middle dose levels, 17 of 19 patients achieved undetectable minimal residual disease — an endpoint where the FDA is proving increasingly receptive toward in other blood cancers — by 12 weeks. In DLBCL, a much harder disease to treat, the responses were less potent, according to last month’s abstract. At the dose expected to move into further studies, nine of 19 patients (47%) responded, with eight of 19 (42%) reaching a complete response. AstraZeneca is expected to break down additional DLBCL results in a presentation Monday evening Pacific time. But most promising about these data is how even patients who had previously taken CAR-T or anti-CD20 therapies were seeing responses, AstraZeneca’s executive VP for oncology R&D Susan Galbraith told Endpoints News . Among 14 individuals who progressed after CAR-T in the study at the target dose level, the ORR was 36% and the complete response rate was 29%. Patients whose cancers progress after taking CAR-T, in particular, have extremely limited treatment options, Galbraith said, but bispecifics as a whole appear to be emerging as a new alternative. “I think that bodes well, not just for an option for those patients, but also the potential for those to be really quite efficacious in the early lines of treatments as well,” she said. “We’ve got work to do to deliver all of that data, but so far, I’m quite excited about potential for this market.” AZD0486 came to AstraZeneca in its buyout of TeneoTwo, acquired in 2022 for up to $1.27 billion, including milestones (AstraZeneca paid only $100 million upfront). TeneoTwo is itself an offshoot from Teneobio, which spun it off after being bought by Amgen in 2021 for $900 million upfront and another $1.6 billion in milestones. The only other approved CD3xCD19 bispecific is Amgen’s Blincyto, which was first approved in 2014. Blincyto has since racked up half a dozen other approvals, but they’ve all come in different settings for acute lymphoblastic leukemia (ALL). In 2023, Amgen reported $861 million in worldwide Blincyto sales. Galbraith said the lack of other such drugs has given AstraZeneca the right opportunity to design their compound in a way where it can best take advantage of the market. There is still the potential for combination therapies, particularly for the DLBCL setting, that have yet to be explored in the clinic. Other companies have taken note recently, with a slew of CD19 bispecific deals announced in recent months. In August, Merck paid $700 million upfront for Curon Biopharmaceutical’s CD3xCD19 compound, and GSK spent $300 million upfront in October for a bispecific from Chimagen Biosciences to test it in autoimmune disease. But Galbraith also believes AZD0486 can be more efficacious, too. “I don’t think you get to the same level of exposure with [Blincyto] as we can achieve with this,” Galbraith said. “And it’s early days yet. But I’m encouraged by the profile that we’re seeing and the potential to have a better CD19 bispecific option for patients in ALL.”
