Tert-butylphenol exposure alters cartilage and bone development in zebrafish

Phenolic antioxidants, such as 2,4-di-tert-butylphenol (2,4-DTBP), 2-tert-butyl phenol (2-BP), and 4-tert-butyl phenol (4-BP), are additives used in domestic water pipes, food packaging, paints, and other industrial products. As additives, they can leach from products and are frequently found in both environmental and human biological samples. Previous studies have demonstrated that 2,4-DTBP exposure can impair the differentiation of human iPS cells into somite- and sclerotome-like cells, and reduce key processes involved in osteoblast formation. Therefore, the goal of this study is to determine if 2-BP, 4-BP, 2,4-DTBP, and its metabolite 3,5-di-tert-butylcatechol (3,5-DTBC) impacts the development of cartilage and bone in vivo, using zebrafish as a model organism. Zebrafish embryos were exposed to increasing concentrations of each of the four chemicals from 1 h post fertilization (hpf) until 5 days post fertilization (dpf), and analyzed for markers of bone and cartilage development. At their highest concentrations tested, both 2-BP and 2,4-DTBP altered axial skeleton formation, with 76% and 61% of the zebrafish showing spinal curvatures, respectively. To corroborate these changes, the expression of marker transcripts were examined. 2-BP exposure reduced mRNA expression of the bone mineralization marker sparc by 1.6-fold. In contrast, 2,4-DTBP increased sparc transcript expression by 1.4-fold. All four compounds significantly upregulated sox9a, a chondrogenesis marker, between 1.4- to 5-fold. Changes in tail cartilage formation were noted using Alician blue staining, with 2,4-DTBP reducing width, length, and cartilage area of the tail, while 2-BP reduced the tail width but with increased the tail base, yielding a more straightened tail. Principle component analysis (PCA) demonstrated associations between sox9a, sparc, nrf2a, reactive oxygen species (ROS), and tail cartilage measurements, particularly in the 2,4-DTBP exposures, suggesting the involvement of nrf2a signaling in impairing cartilage formation. Overall, the study shows that each of the phenolic antioxidants differentially affects the development of bone and cartilage structures in zebrafish.

Green tea fermented by Ganoderma lucidum presented anti-obesity properties via enhanced thermogenesis in vitro and on C57BL/6J mice

High-fat diets contribute to obesity and metabolic disorders. Ganoderma lucidum is renowned for its abundant bioactive compounds and diverse pharmacological effects. Green tea fermented by G. lucidum (TFG) has been shown to enhance lipid-lowering activity in vitro significantly. Using UPLC-MS/MS and GC-MS/MS, we identified 78 active lipid-lowering compounds in TFG. We explored their potential targets and pathways through network pharmacology, validated by in vivo experiments. In a 4-week trial, 70 mice were randomly assigned to 7 groups: ND (normal diet), HFD (high-fat diet), PC-HFD (HFD with orlistat), NFT1 (HFD with 200 mg/kg/day non-fermented tea), NFT2 (HFD with 400 mg/kg/day NFT), TFG1 (HFD with 200 mg/kg/day TFG), and TFG2 (HFD with 400 mg/kg/day TFG). TFG treatment significantly reduced body weight, hepatic lipid droplets, and epididymal adipocyte size in mice compared to the HFD group. TFG also increased the abundance of lipid-lowering bacteria, such as Lactococcus and Lachnospirales. Liver transcriptomic and fecal metabolomic analyses revealed that TFG reduced triglyceride (TG), diglyceride (DG), monoglyceride (MG), and free fatty acid (FFA) levels and differentially regulated key genes (Dpf3, Atp5k, ND3) involved in the thermogenesis pathway. RT-PCR confirmed that TFG upregulated the mRNA expressions of AMPK, UCP1, PGC1α, and PPARγ in dorsal fat. In conclusion, TFG enhances thermogenesis via the AMPK-PGC1α pathway and increases the abundance of lipid-lowering bacteria, thereby reducing fat accumulation in mice. These findings offer insights into TFG's anti-obesity mechanisms, providing a scientific basis for developing new weight loss methods or products.

BRAF-induced EHF Expression Affects TERT in Aggressive Papillary Thyroid Cancer

AbstractContextBRAFV600E and TERT promoter mutations in papillary thyroid carcinoma (PTC) have a synergistic effect on prognosis. This effect is believed to arise from MAPK activation triggered by BRAFV600E, leading to the upregulation of ETS transcription factors that bind to the mutant TERT promoter.ObjectivesTo explore the role of ETS factors in relation to clinical features, BRAFV600E, and TERT promoter mutations in PTC.DesignTranscriptomic data for 28 ETS factors were analyzed in the PTC cohort of The Cancer Genome Atlas (n = 399) and subsequently validated in a local cohort (n = 93). In vitro experiments were performed to investigate the regulatory role in relation to BRAFV600E and TERT expression.ResultsThe Cancer Genome Atlas identified ETS1, ERG, FLI1, GABPA, EHF, ETV6, and SPDEF as differentially expressed genes between stages I + II and III + IV. In both cohorts, EHF was consistently associated with adverse clinical features, BRAFV600E and TERT promoter mutation/expression. Notably, in BRAFV600E mutated PTC, high EHF expression was associated with shorter disease-free survival. Cases harboring concurrent BRAFV600E, TERT promoter mutations, and high EHF expression exhibited the shortest disease-free survival. In cells harboring concurrent BRAFV600E and TERT promoter mutation, overexpression of EHF significantly increased TERT expression, whereas knockdown or pharmacological inhibition of BRAF significantly decreased both EHF and TERT expression. In addition, chromatin immunoprecipitation and quantitative PCR analysis suggested a potential binding of EHF in TERT promoter mutant cells but not in TERT promoter wild-type cells.ConclusionThe ETS transcription factor EHF is associated with poor prognosis in PTC. This is potentially mediated by BRAF-induced upregulation of EHF, which in turn increases TERT expression in TERT promoter mutated cells.
