Introduction

Negatively regulates MAP kinase activation by limiting the formation of Raf/MEK complexes probably by inactivation of the KSR1 scaffold protein. Also acts as a Ras responsive E3 ubiquitin ligase that, on activation of Ras, is modified by auto-polyubiquitination resulting in the release of inhibition of Raf/MEK complex formation. May also act as a cytoplasmic retention protein with a role in regulating nuclear transport.

Insight into flavor changes in bighead carp (Aristichthys nobilis) fillets during storage based on enzymatic, microbial, and metabolism analysis

The flavor alterations in bighead carp subjected to varying storage temperatures and the underlying metabolic mechanism were elucidated. Analysis of volatile flavor compounds, electronic nose, free amino acids, ATP-related compounds, and sensory evaluations uncovered a progressive flavor deterioration during storage, especially at 25 °C. Metabolomics-based flavor relating component profiling analysis showed that free fatty acids formed various fatty aldehydes including (E, E)-2,4-heptadienal and nonanal under lipoxygenase catalysis. Alcohol dehydrogenase and alcohol acyltransferases were intimately involved in alcohol and ester generation, while alkaline phosphatase, 5'-nucleotidase, and acid phosphatase were closely associated with IMP, Hx, and HxR conversion, respectively. Aeromonas, Serratia, Lactococcus, Pseudomonas, and Peptostreptococcus notably influenced flavor metabolism and enzyme activities. The metabolism disparities of valine, leucine, isoleucine, lysine, and α-linolenic acid could be the primary factors contributing to flavor metabolism distinctions. This study offers novel insights into the flavor change mechanisms and potential regulation strategies of bighead carp during storage.

Dismantling of necrotroph Alternaria alternata by cellular intervention of Peppermint Oil Nanoemulsion (PNE)

Alternaria alternata, a common necrotrophic fungal pathogen, poses a significant threat to various crops, causing substantial yield losses and quality deterioration. In the present study, we explore the potential fungicidal properties of Peppermint Oil Nanoemulsion (PNE) against A. alternata and investigate its impact on the fungal phenotype. Our previous study synthesized the PNE using a nanoemulsion approach, optimizing its formulation for enhanced stability and efficacy. The present study extended the assessment of a multidisciplinary approach to comprehensively analyze the fungicidal efficacy of PNE against A. alternata. Notably, in a liquid growth medium, 0.5 % of PNE could reduce A. alternata's biomass by 96 %. PNE-treated mycelia were stained with a nitro-blue tetrazolium (NBT) dye to assess ROS accumulation during oxidative stress induced by PNE. A higher degree of ROS generative potential of PNE has appeared in 72 h treated mycelia. PNE-treated mycelium showed cell wall alterations, with red fluorescence peaking at 0.5 %, indicating a dose-dependent effect compared to the untreated control. Consequently, PNE treatment led to a significant early hour increase in electrical conductivity (EC), extended to 306.03-353.33 μS/cm compared to 277.67-280.33 μS/cm untreated control. Scanning Electron Microscopy (SEM) analysis of A. alternata reflects the osmotic imbalance and structural damage in mycelia as the obvious cause of fungal inhibition. In addition, a phenotype microarray analysis of PNE-treated A. alternata mycelia revealed a significant phenotypic loss in 37 out of 708 substrates, potentially impacting metabolic pathways essential for fungi's functional processes. The study found that downregulation of genes like Cre A, NmrA, SOD, IMP, EfP, and Erg, which are linked to A. alternata's stress coping mechanisms, leads to alterations in survival and adaptation. Additionally, understanding the phenotypic changes induced by PNE contributes to our knowledge of the mode of action of this nanoemulsion against A. alternata. In conclusion, this study provides a comprehensive analysis of the fungicidal and phenotypic effects of PNE, offering a promising avenue for sustainable fungal control. The implications of our research extend to the development of novel, natural fungicidal agents for agricultural applications.

In-vitro activities of essential antimicrobial agents including aztreonam/avibactam, eravacycline, colistin and other comparators against carbapenem-resistant bacteria with different carbapenemase genes: A multi-centre study in China, 2021

OBJECTIVECarbapenem-resistant bacteria (CRB), including carbapenem-resistant Acinetobacter baumannii (CRAB), carbapenem-resistant Pseudomonas aeruginosa (CRPA) and carbapenem-resistant Enterobacterales (CRE), pose a considerable threat to public health in China. Eravacycline, aztreonam/avibactam and colistin are important antimicrobial agents for the treatment of serious infections caused by CRB. This study aimed to evaluate the prevalence of CRB strains, and the susceptibility of commonly used clinical antimicrobial agents against strains with different carbapenemase genes.METHODSIn total, 7194 gram-negative bacteria strains were collected from different regions of China, and 924 carbapenem-resistant strains were identified. All strains were from confirmed infections. Antimicrobial susceptibility testing, covering 21 antimicrobial agents including aztreonam/avibactam, eravacycline, colistin and other comparators, was performed using the broth microdilution method. Carbapenemase genes (bla_KPC, bla_NDM, bla_OXA, bla_IMP and bla_VIM) were screened using polymerase chain reaction amplification and sequence analysis. All statistical analyses were performed using Statistical Package for the Social Sciences Version 23.0.RESULTSThe isolation rates of CRE, CRAB and CRPA were 6.31% (332/5265), 62.95% (440/699) and 15.20% (152/1000), respectively. The predominant carbapenemase in carbapenem-resistant Escherichia coli (CRECO) was NDM, while in carbapenem-resistant Klebsiella pneumoniae (CRKP), it was KPC. All CRAB produced OXA-23, and 85.52% of CRPA did not produce any of the following carbapenemases: NDM, KPC, VIM, IMP and OXA. Aztreonam/avibactam, colistin and eravacycline exhibited high antimicrobial activity against different species producing various carbapenemases. Compared with ceftazidime/avibactam, aztreonam/avibactam demonstrated superior antimicrobial activity, particularly pronounced in CRECO and strains producing metallo-beta-lactamases. In comparisons between tigecycline and eravacycline, the latter maintained higher antimicrobial activity across different species. Antimicrobial agents exhibited varying levels of activity against strains with different resistance mechanisms.CONCLUSIONSUsing aztreonam/avibactam, eravacycline and colistin to treat infections caused by CRB offers significant advantages. These findings will guide clinical practice and optimize antimicrobial administration.
