Introduction

One HIP oligomer binds the ATPase domains of at least two HSC70 molecules dependent on activation of the HSC70 ATPase by HSP40. Stabilizes the ADP state of HSC70 that has a high affinity for substrate protein. Through its own chaperone activity, it may contribute to the interaction of HSC70 with various target proteins (By similarity).

Differential effect of acute versus persistent insect-specific flavivirus infection on superinfection exclusion of West Nile, Zika and chikungunya viruses in RNAi-competent and -deficient mosquito cells

Millions of people are annually infected by mosquito-transmitted arboviruses including dengue virus (DENV), West Nile virus (WNV), Zika virus (ZIKV) and chikungunya virus (CHIKV). Insect-specific flaviviruses (ISFs), which only infect mosquitoes and cannot replicate in vertebrates, can offers a potential one health strategy to block the transmission of arboviruses by reducing the mosquito's susceptibility for subsequent arbovirus infections through superinfection exclusion (SIE),. Most SIE studies focus on acute ISF infections in RNAi-deficient Aedes albopictus C6/36 cells. Because ISFs are known to persistently infect mosquitoes, acute infections in C6/36 cells may not accurately reflect natural interactions between ISFs and arboviruses. To study the underlying mechanisms for SIE, we persistently infected C6/36 and RNAi-competent Aedes aegypti Aag2 cells with the ISF Binjari virus (BinJV) and a BinJ-ZIKV chimera that contains the ZIKV prME structural genes. SIE of WNV, ZIKV and CHIKV by BinJV was more pronounced in acute than in persistently infected cells and much stronger in acutely infected C6/36 cells compared to Aag2 cells. The viability of RNAi-deficient mosquito cells was severely reduced upon acute ISF infection, which correlated to the observed SIE. However, persistently infected mosquito cells still inhibited subsequent arbovirus replication. Moreover, RNAi-competent Aag2 cells were better protected against ZIKV superinfection when they were pre-infected with BinJ-ZIKV as compared to BinJV. Therefore, acute ISF infections and strong cytopathic effects in RNAi-deficient cells augment SIE, while in persistently infected cells SIE is established through RNAi-dependent and independent mechanisms. This highlight the importance of using more representative in vitro models.

Pathological and bacteriological investigations of Pasteurella multocida-induced epididymitis in calves

Between June 2017 and October 2023, five cases of epididymitis caused by Pasteurella multocida were confirmed in four Japanese Black calves and one Japanese Black-Holstein crossbreed calf across five farms in three prefectures in Japan. Although P. multocida is primarily known to cause respiratory diseases in cattle, its role in reproductive disorders, particularly in epididymitis, has not been extensively studied. Histopathological examination showed pyogranulomatous lesions in the epididymal duct and surrounding areas, consisting of ductal extension, immune cell infiltration, epithelial degeneration, abscess formation, and fibrosis with angiogenesis. In the immunohistochemical analysis, positive reactions to anti-P. multocida serotype A antibodies were observed in macrophages and neutrophils in the epididymal ducts and surrounding areas, suggesting an ascending infection rather than hematogenous spread. This report is the first to directly confirm P. multocida involvement in bovine epididymitis through immunohistochemical examination. P. multocida was isolated in pure culture from the scrotal contents in all cases. The isolates were grouped into sequence types ST79 and ST13, commonly found in the respiratory tract and oral cavity of cattle, using the Rural Industries Research and Development Corporation multilocus sequence typing scheme; pulsed-field gel electrophoresis revealed unique band patterns in the isolates from each case. These results suggest that multiple genetically distinct strains, commonly found in the nasal and oral cavities, incidentally infected the epididymis via an ascending route. This bacterium warrants further investigation as a potential cause of reproductive disorders, particularly in Japanese Black cattle.

Mechanism of chaperone recruitment and retention on mitochondrial precursors

Nearly all mitochondrial proteins are imported into mitochondria from the cytosol. How nascent mitochondrial precursors acquire and sustain import competence in the cytosol under normal and stress conditions is incompletely understood. Here, we show that under normal conditions, the Hsc70 and Hsp90 systems interact with and redundantly minimize precursor degradation. During acute import stress, Hsp90 buffers precursor degradation, preserving proteins in an import-competent state until stress resolution. Unexpectedly, buffering by Hsp90 relies critically on a mitochondrial targeting signal (MTS), the absence of which greatly decreases precursor–Hsp90 interaction. Site-specific photo-cross-linking and biochemical reconstitution showed how the MTS directly engages co-chaperones of Hsc70 (St13 and Stip1) and Hsp90 (p23 and Cdc37) to facilitate chaperone retention on the mature domain. Thus, the MTS has a previously unappreciated role in regulating chaperone dynamics on mitochondrial precursors to buffer their degradation and maintain import competence, functions that may facilitate restoration of mitochondrial homeostasis after acute import stress.
