The Sequence Alignment for Multiple Antibody CDRs in Antibody Drug Research and Development

3 min read

The practice of antibody sequence retrieval often necessitates the alignment of multiple antibodies' sequences within the same series for thorough research analysis. Conducting such a study manually — downloading each antibody sequence individually and inserting them into an offline document for subsequent comparison— is a laborious and time-consuming task.

A more efficient technique involves utilizing the Multiple Sequence Alignment (MSA) antibody sequence comparison tool offered by Patsnap Bio’s biological sequence database. This mechanism facilitates real-time online sequence comparisons, offering easy operation and clear results. Instructions for utilizing this tool are as follows:

Initially, sign-up for a free Bio database account at https://bio-patsnap-com.libproxy1.nus.edu.sg. Once registered, select the Drug/Gene Index in the left search input field and key in 'Cemiplimab monoclonal antibody' for sequence retrieval. In the resultant detailed search page, tick the relevant sequence and click "Save Sequence." This action will redirect the user to the workspace page where the sequences can be reserved. At this juncture, create a novel customized folder for convenient sequence storage and comparison.

To further continue, visit the database homepage and access the workspace from either the "Workspace" link at the page bottom or the top-right corner of the homepage.

Within your workspace, locate and select the specific sequence intended for comparison, then click "Compare Sequence." The system will automatically highlight the chosen sequences for comparison.

It is also possible to juxtapose multiple sequences simultaneously or make a pairwise comparison. Choose one sequence as a point of reference and couple it with other sequences for comparison purposes. Alternatively, the CDRs can be used for sequence comparison in the highlight section. To save the comparison report, simply click "Save" in the upper right corner of the "Alignment Report" section for later reference.
