Introduction

Mucocellular carcinoma of the ovary, usually metastatic from the gastrointestinal tract, characterized by areas of mucoid degeneration and the presence of signet-ring-like cells. It accounts for 30%-40% of metastatic cancers to the ovaries and possibly 1%-2% of all malignant ovarian tumors. The lesions may not be discovered until the primary disease is advanced, and most patients die of their disease within a year. In some cases, a primary tumor is not found. A Phase 1 First-In-Human Study of the Anti-CD73 IPH5301 Alone or in Combination With Chemotherapy and Trastuzumab in Patients With Advanced Solid Tumors

CHANCES-IPC 2021-008 is First In Human, Phase I, multicenter, European study evaluating an anti-CD73, IPH5301 in advanced and/or metastatic cancer.
The trial will be conducted in two parts, Part I- Dose escalation: This part aims to identify the maximum tolerated dose (MTD) of IPH5301 agent in monotherapy and recommended phase 2 dose (RP2D) for future trials, followed by a safety expansion study part cohort.
Part II- Expansion cohort: A total of 12 HER2+ cancer patients, respectively 6 breast cancer patients and 6 gastric cancer patients, is planned to be enrolled into the next expansion cohort to select a recommended dose of IPH5301 to be administered in combination with chemotherapy and trastuzumab for evaluation in future trials with selected advanced solid tumors.

Krukenberg's tumor diagnosed in pregnancy is an uncommon situation that raises both diagnosis and medical management issues. We performed a review of the existing literature regarding this pathology, diagnostic means and therapeutic approaches, motivated by a case in our own practice. A 35-year-old primigravida was diagnosed with an adnexal mass during the first trimester prenatal ultrasound. Ultrasound revealed a 10 cm right adnexal mass with multiple septae, richly vascularized, whose presence and characteristics were confirmed by magnetic resonance imaging. Due to the progressively increasing tumor size, laparoscopy was performed with right adnexectomy and peritoneal biopsies. Histopathology diagnosed a metastatic ovarian tumor from a mucinous colorectal adenocarcinoma. After delivery the patient was further investigated and diagnosed with sigmoid cancer. Even though ovarian cancer in pregnancy is rare, adnexal ultrasound is mandatory when scanning during the first trimester to rule out the presence of associated fallopian or ovarian masses.

Tumor specific delivery and radiation-enhanced tumor penetration of mesoporous silica nanoparticles for effective radionuclide therapy of ovarian peritoneal metastasis

Advanced ovarian cancer with peritoneal metastasis is challenging to treat. Limited tumor delivery and penetration of the therapeutics to deep tumor regions are significant barriers to effective treatment. The rising radiopharmaceuticals offer hopes for patients through targeted delivery. However, site specific delivery avoiding off target effect remain critical and challenging. We have developed radioactive ¹⁶⁶Holmium loaded mesoporous silica nanoparticles (¹⁶⁶Ho-MSNs) that exhibited predominant accumulation to peritoneal metastases of ovarian cancer upon intraperitoneal administration. It was observed that fluorescence labeled radioactive ¹⁶⁶Ho-MSNs distributed throughout the tumor tissues, while non-radioactive fluorescent ¹⁶⁵Ho-MSNs showed mainly tumor surface deposition of MSNs. The deep penetration leads to uniform therapeutic radiation distribution and absorbed doses within tumors as demonstrated by the dosimetry analysis. The radiation dosing regimen consisting of two 100 µCi ¹⁶⁶Ho-MSN doses separated by 7 days decreased tumor activity and increased the overall lifespan and ascites free survival in several models of IP tumor-bearing mice. These findings illustrate that ¹⁶⁶Ho-MSN is promising for the treatment of ovarian peritoneal metastasis, with selective targeting advantage of the nanoparticles and limited off-target radiation exposure.

Diagnostic Challenges in Invasive Mole With 18F-FDG PET/CT

AbstractA 34-year-old woman underwent 18F-FDG PET/CT scan following a diagnosis of an invasive mole. Although CT indicated multiple lung metastases, FDG uptakes were not evident. In contrast, FDG uptakes were observed in the pelvic lesion including the left ovary, suggestive of ovarian metastasis. MRI disclosed a ring-shaped enhancement typical of a corpus luteum. This corpus luteum was supposed to be maintained by the elevated human chorionic gonadotropin excreted from the invasive mole. This case underlines the importance of careful interpretation of 18F-FDG PET/CT in the evaluation of invasive moles, highlighting potential false positive in corpus luteum.
