This is a national, multicenter, open-label, randomized, phase II/III trial that evaluates the efficacy and safety of favipiravir and ribavirin in the treatment of patients with confirmed COVID-19 observed within 72 hours. Approximately 100 patients will be randomized in 1:1 ratio and divided into two groups.

STREAM: The Evaluation of a Standard Treatment Regimen of Anti-tuberculosis Drugs for Patients With MDR-TB

Tuberculosis (TB) is a common, infectious, bacterial disease that is spread when an infected person transmits their saliva through the air by coughing or sneezing. Despite the availability and effectiveness of affordable six-month treatments for tuberculosis (TB), the worldwide control of this disease is currently being impacted by the emergence of multidrug resistant TB (MDR-TB). MDR-TB refers to TB that is resistant to at least isoniazid and rifampicin. These are the two most powerful first-line drugs used to treat pulmonary TB. MDR-TB usually develops while a person is taking TB treatment due to either inappropriate treatment or failure of patients to comply with their treatment. This strain of drug-resistant bacteria can also be spread to other people through the air. With the incident rate of MDR-TB on the rise, there is a need to investigate optimal treatment regimens using effective drugs.

A novel aptasensor integrating the DNA rolling nanomachine and Tb/COF/KB as dual signal amplifiers for kanamycin detection in foods

A novel aptasensor for the detection of kanamycin (KNM) was developed based on dual signal amplification through nanomaterials and DNA rolling machines. Firstly, a composite consisting of the covalent organic framework (COF), ketjen black (KB) and toluidine blue (Tb), was synthesized for signal generation. Tb molecules were loaded into the composite and combined with AuNPs to serve as tracer label. Secondly, the wDNA-loaded Fe₃O₄ magnetic beads (wDNA@MBs) as the DNA walking legs, with cleavage by Mg²⁺-driven DNAzyme, the DNA walking legs can move along the sDNA-decorated MBs surface, converting the target KNM into numerous output DNA strand, achieving sensitive amplification. With a low detection limit of 0.66 fM and a wide linear dynamic range from 10 fM to 100 nM, this aptasensor showed good results in the detection of KNM in real samples. Therefore, we envision it can provide a useful method for KNM detection in actual samples.

A signal-on photoelectrochemical aptasensor based on WO3/CdS heterojunction for the ultrasensitive detection of kanamycin

In this study, a signal-on photoelectrochemical (PEC) aptasensor for the ultrasensitive determination of kanamycin (KANA) was constructed using WO₃/CdS heterojunction as photoactive material. Firstly, WO₃/CdS heterojunction with excellent photoelectric response was successfully prepared by simple co-precipitation method, resulting in a strong and stable initial photocurrent. In addition, amino modified aptamers were immobilized on the electrode surface by glutaraldehyde as biological recognition components. In the presence of the target KANA, it is specifically recognized and captured by the aptamers. More importantly, KANA can act as a signal amplifier to enhance the photocurrent due to the oxidation of KANA by photogenerated holes. Therefore, a signal-on PEC aptasensor based on WO₃/CdS heterojunction with high selectivity was obtained for the detection of KANA. Under optimized experimental conditions, the PEC aptasensor demonstrated a wide linear range of 10 pM to 400 nM, with a detection limit of 6.77 pM. Meanwhile, the designed PEC aptasensor had been successfully utilized for the analytical examination of milk, fish, serum, and water samples.

DNA nanowire-enhanced polymerization isomerization cyclic amplification toward photocurrent polarity switching-based PEC biosensing

The photocurrent-polarity-switching signal transduction strategy has provided an effective approach to improve the performance of photoelectrochemical (PEC) biosensors. Herein, we prepared a MXene/CdS nanocomposite based PEC biosensor and combined it with the polymerization and isomerization cyclic amplification (PICA)-assisted capture of a Fe³⁺ and Cu²⁺ co-coordinating 5,10,15,20-tetrakis(4-aminophenyl)porphyrin (TFC) based signal probe to develop a novel photocurrent-polarity-switching PEC biosensing method for the detection of kanamycin antibiotic. The PICA arose from the aptamer recognition-triggered release of an Mg²⁺-dependent DNAzyme (MNAzyme) strand to cleave its substrate. This reaction also triggered the assembly of a DNA nanowire decorated with abundant MNAzymes through hybridization chain reaction. Due to the DNA nanowire-enhanced PICA to extremely increase the probe capture at the biosensor and the good energy level matching between TFC and MXene/CdS, a strong polarity-switching based cathodic photocurrent response was produced to construct the signal transduction strategy. This not only excluded the possible false positive or negative signal interferences but also endowed a seven-order magnitude wide linear range and a very low detection limit of 0.03 fg mL^-1 to the method. In addition, it also has convenient assay manipulation, high selectivity, outstanding repeatability, and excellent reliability, which determine its high potential promising for practical applications.
