Lactoferrin in cancer: Focus on mechanisms and translational medicine

Lactoferrin is an iron-binding glycoprotein that provides natural protective effects to the human body. Its biological properties, including antibacterial, antiviral, anti-inflammatory, immune-regulatory, and iron metabolism-regulating functions, have been extensively studied. With further research, lactoferrin's impact on tumorigenesis and tumor microenvironment has become increasingly evident, as it inhibits tumor proliferation, invasion, and metastasis through multiple pathways. This article summarizes the molecular mechanisms underlying lactoferrin's anticancer effects, explores its association with the malignant progression of various cancers, and highlights its clinical translational potential as a potential cancer biomarker and drug delivery carrier to enhance anticancer therapy efficiency. Due to the high safety profile of lactoferrin, its widespread application in the field of cancer treatment is highly anticipated.

Protective effects and mechanisms of lactoferrin and HIF-1α on dry eye syndrome in mice

The objective of this study was to investigate the protective effects and related mechanisms of lactoferrin and HIF-1α on dry eye syndrome (DED) in mice. The expression levels of lactoferrin and HIF-1α in tears of DED patients and normal controls were detected. A DED mouse model received lactoferrin (50 mg/kg dissolved in 2 mL PBS) or DMOG (40 mg/kg dissolved in 2 mL PBS) orally daily for 28 days. DMOG (dimethyloxaloylglycine) is a hypoxia-inducible factor prolyl hydroxylase inhibitor. Various tests conducted in this study were phenol red thread test, corneal fluorescein sodium staining, hematoxylin-eosin (HE) staining, PAS staining of conjunctiva, TUNEL staining, and Western blotting. Compared to normal controls, DED patients showed significantly decreased expression of lactoferrin and increased expression of HIF-1α in tears (P < 0.05). Compared to normal mice, DED model mice exhibited significantly decreased tear secretion, goblet cell count, Bcl-2, lactoferrin, and STAT3 protein expression levels, and significantly increased corneal fluorescein sodium staining grade, TUNEL positivity rate, Bax, HIF-1α, p21, and p27 protein expression levels (P < 0.05). Treatment with lactoferrin or DMOG significantly increased tear secretion, goblet cell count, Bcl-2, lactoferrin, HIF-1α, and STAT3 protein expression levels, and significantly decreased corneal fluorescein sodium staining grade, TUNEL positivity rate, Bax, p21, and p27 protein expression levels in DED model mice (P < 0.05). Normal mice showed normal corneal morphology. Compared to normal mice, DED model mice exhibited rough surface of corneal epithelial cell layer with vacuolated cells and inflammatory cell infiltration. Treatment with lactoferrin or DMOG significantly alleviated corneal lesions in DED model mice. Lactoferrin and HIF-1α exert protective effects on DED in mice.

Titanium nanostructure mitigating doxorubicin–induced testicular toxicity in rats via regulating major autophagy signaling pathways

Doxorubicin (DOX) is a powerful antineoplastic FDA-approved anthracycline-derived antibiotic and is considered as the most suitable intervention for solid tumors and hematological cancers therapy. However, its therapeutic application is highly limited due to acute and chronic renal, hematological and testicular toxicity. Oxidative stress, lipid peroxidation and apoptosis in germ cells as well as low sperm count, motility and disturbing steroidogenesis are the principal machineries of DOX-induced testicular toxicity. Nevertheless, the comprehensive molecular pathways responsible for DOX-induced testicular damage are not yet fully understood. The current study aims to clarify the role of autophagy and apoptotic signaling pathways in testicular toxicity induced by DOX in the rat model. The study also investigates the potential role of both titanium dioxide nanoparticles (TiO₂NP) loaded with DOX and Lactoferrin in combination with DOX in mitigating testicular toxicity induced by DOX the standard antitumor drug. In the present study, male Wister albino rats were intoxicated with a total cumulative dose of DOX (18 mg/kg) via intra-peritoneal injection and served as positive control group. The other two groups administered either TiO₂NP-DOX or lactoferrin-DOX. Furthermore, biochemical and molecular analyses were then performed. DOX intoxication induced testicular toxicity, revealing mineral imbalance as indicated by an increase in both calcium and magnesium concentrations. Administration of either TiO₂NP-DOX or lactoferrin-DOX resulted in a significant modulation of disrupted mineral concentrations, with TiO₂NP-DOX showing superiority in modulating both magnesium and calcium concentrations. Acid Phosphatase level significantly increased upon DOX-induced testicular damage. Molecular analysis of EGFR and K-RAS gene expression showed significant overexpression, while p53 and JAK-2 gene expression was significantly reduced post-DOX intoxication. Protein expression of both AKT and PI3K significantly increased upon DOX administration. Results showed a remarkable modulation of all disrupted gene and protein expressions upon treatment with TiO₂NP-DOX or Lactoferrin-DOX with the superiority of TiO₂NP-DOX in modulating these parameters. In conclusion, TiO₂NP-DOX could be a promising drug delivery system to improve bioavailability and drug release, as well as reducing DOX's adverse effects particularly on testicular function.

Koch argued that TB is a contagious disease, spreading from one person to the next through the air. Based on the notion that TB is caused by a contagious bacterium, conventional treatment for TB involves vaccination and administration of multiple antibiotics over a long period. Price Foundation Logo The incidence of TB has declined dramatically since the 19th century, especially in the West, but the disease still kills an estimated 3,000 people per day, mostly in lower-income countries. "We propose looking elsewhere for the cause of this terrible illness," says Sally Fallon Morell, president of the Weston A. Price Foundation. "The symptoms of TB are identical to those of a disease called siderosis, which is poisoning by toxic iron fumes." Siderosis occurs most frequently today in welders. Symptoms of both TB and siderosis include breathlessness, coughing (including coughing up blood), decreased lung function, miliary (millet-seed-like) lung lesions, and darkening of the eyes due to iron deposits, a unique symptom. Koch found that TB bacillus grew only on ox blood or occasionally on meat, confirming that it is an iron-loving organism. "These facts beg the question: is the organism the cause of TB or the therapy, appearing in the body when excess iron needs clearing?" asks Morell. During the Industrial Age, exposure to iron fumes was widespread, especially in trench warfare, where the heavy pollutant would linger. TB was a huge problem in the Crimean War, for example, with iron cannons, hand grenades, gun barrels, cookware, all of which release toxic iron oxide when heated, and coal smoke (which is high in iron). Soldiers living in these trenches suffered massively from TB. In 1790, a new invention increased exposure to iron oxide and brought it into the drawing room: the cast iron stove. With its large surface area, the cast iron stove released large amounts of iron oxide, which exposed not only the cooks and maids in unventilated basements but also the rest of the household enjoying warm rooms upstairs. As stainless steel and aluminum replaced cast iron in stoves, furnaces, and industrial equipment—and as oil and natural gas replaced coal—exposure to iron oxide gradually declined. Today, TB deaths occur mainly in countries where cast iron stoves and cookware are still widely used, often in unventilated hovels, and where iron mining and iron works are common. Coal is also frequently used in these areas' power plants and domestic settings. In houses where cast iron pans are the only exposure, there is not a concern. "If the cause of TB is toxic exposure to iron fumes, it makes little sense to treat the disease with vaccinations and antibiotics," says Morell. "Rather, appropriate treatment modalities may include chelating agents such as DSMO and lactoferrin, plus removal from exposure to sources of ferrous oxide.
