Non-targeted and targeted detection of hydrophilic compounds in fu brick tea: A study on samples from major Chinese production regions and different processing stages

Systematic research is still lacking on the content of hydrophilic compounds in Fu Brick Tea (FBT) from major Chinese production regions and their variation patterns during the processing of FBT. This study utilized optimized non-targeted (UHPLC-Q-Exactive Orbitrap-MS) and targeted (UHPLC-QqQ-MS) metabolomics to analyze 73 FBT samples from six regions of China and 30 samples from different stages of FBT processing. 573 and 74 hydrophilic compounds were respectively relatively and absolutely quantified for the first time. Among the 73 FBT samples, alkaloids were the most abundant (26221.76-36701.75 mg/kg), followed by soluble sugars (211372.59-26112.85 mg/kg), organic acids (17014.07-23589.20 mg/kg), amino acids (6664.59-9791.46 mg/kg), nucleotides (1020.33-2175.16 mg/kg), and nucleoside (650.86-1446.26 mg/kg). During the processing of FBT, sugars, acids, nucleotides, and nucleosides increased, while amino acids decreased, and alkaloids slightly increased. This study enhances the understanding of hydrophilic compounds in FBT.

Enhanced intracellular calcium detection using dopamine-modified graphene quantum dots with dual emission mechanisms

The tracking of calcium ions (Ca²⁺) is of great significance in clinical medicine. Dopamine-functionalized graphene quantum dots (DA-GQDs) have been developed as a novel fluorescence sensor for detecting and imaging Ca²⁺ with high selectivity and sensitivity. Upon the addition of various concentrations of Ca²⁺, the fluorescence intensity of DA-GQDs notably enhanced and exhibited a redshift. This behavior was regulated by the photoinduced electron transfer and internal charge transfer mechanisms, as elucidated by fluorescence titration and density functional theory calculations. The coordination of Ca²⁺ with the oxygen groups on DA-GQDs enhanced its sensitivity and selectivity. The detection limit was determined to be 0.05 µM, with a robust linear response range of 4.93-10.61 µM. Even in the presence of chelating agents such as gluconate, the DA-GQDs accurately quantified Ca²⁺. Competition experiments demonstrated that the DA-GQDs exhibited a high fluorescence response specifically for Ca²⁺, with negligible selectivity toward other ions and biomolecules. The CCK8 assay confirmed that the DA-GQDs are nontoxic and biocompatible. Moreover, DA-GQDs have been successfully employed for imaging intracellular Ca²⁺. This study provides new insights into the design of Ca²⁺ fluorescence sensors using biocompatible molecular components.

Shelf-life quality evaluation and smartphone-based phenolic analysis of cassava treated with varied chitosan formulations

Phenolic compounds exhibit potent antioxidant properties and defense mechanisms in plants, serving as quality biomarkers. This study investigates the effect of chitosan formulations, incorporated with 1 % and 2 % ascorbic acid and quercetin, for extending cassava shelf life and improving nutritional quality. The PhotoMetrix® smartphone App was employed to analyze the impact of these treatment-formulations on their total phenolic contents (TPC) using RGB (Red-Green-Blue) color scale, validated by statistical analysis. Treated cassava samples exhibited higher levels of TPC (up to 70 %) and antioxidant activity (up to 35.39 %). Treated cassava samples with high TPC demonstrated better antioxidant capacities, visual appearances and starch contents, alongside reduced dry matter and H₂O₂ levels. These outcomes highlight the effectiveness of chitosan-antioxidant formulations in boosting TPC, mitigating oxidative stress, and delaying cassava deterioration. The study also demonstrates PhotoMetrix® as a cost-effective, efficient alternative for phenolic assessment, and a tool for shelf-life study towards food security.

Vitamin boost: Half of Millennials are using vitamins and supplements more often than they did before the pandemic

CHICAGO, Jan. 31, 2023 /PRNewswire/ -- The use of vitamins, minerals, and dietary supplements (VMS) among Americans is on the rise. According to new research from Mintel, among the 78% of Americans who are vitamin* users, a third (34%) have increased their usage since the start of the COVID-19 pandemic, including nearly half (47%) of Millennials. In line with consumers overall, Millennials say supporting their overall physical health (66%) and immune system (62%) are the top two reasons for VMS use. However, Millennials (43%) prioritize VMS use to support their mental well-being more than consumers overall (34%). While sales of VMS surged during the pandemic, increasing an impressive 22% between 2019-20 to reach $31.52 billion in 2020, growth leveled out in 2022 when sales grew an estimated 4.1% to reach $35.64 billion. Sales are set to grow a further 5% in 2023 when the market is expected to hit an estimated $37.44 billion. Dorothy Kotscha, Health and Wellness Analyst, Mintel Reports US, said: "While the pandemic negatively impacted multiple areas of consumers' lives, the shift it created in how consumers view and approach their health has benefitted health and wellness brands, particularly within the vitamin, mineral, and dietary supplements (VMS) space. Immune health concerns no longer only hold seasonal significance and consumers have a heightened focus on the importance of both physical and mental well-being. Consumers show interest in single-letter vitamins and supplements The majority of consumers take some sort of vitamin or supplement: 78% take a vitamin and 64% take a supplement. While use of vitamins remained flat over the past two years (77% in 2021), usage of supplements increased by 13 percentage points, up from 51% in 2021. Specifically, single-letter vitamins and mineral supplements have seen a gradual uptick in usage over the last five years: single-letter vitamin use increased from 47% in 2018 to 52% in 2022, while mineral supplement use increased from 33% to 42%. Mintel research shows that consumers are looking to optimize their health by focusing on vitamins and minerals they may be lacking for a more personalized approach, rather than a one-size-fits-all multivitamin. "Customization and personalization are more important than ever as consumers are increasingly interested in broadening their approach to health. Single-letter vitamins may erode the widespread usage of multivitamins as consumers desire greater control over their VMS regimens. Major VMS players should take a close look at ways to offer customized products in a variety of formats and distribution channels in order to optimize the way that they meet their customers' unique needs," concluded Kotscha.
