Abstract:
Water hyacinth (Eichhornia crassipes (Mart.) Solms) is a highly invasive aquatic weed native to the Amazonia basin, known for its rapid propagation, adaptability, and utilization in traditional medicine. The study aims to unveil the therapeutic potential of water hyacinth flowers methanolic extract (EC CME) and its four kupchan fractions (EC PESF, EC DCMSF, EC EASF, EC ASF) through diversified chemical‐pharmacological approaches. GC‐MS/MS of EC‐CME uncovered a rich tapestry of 72 phytochemical components. In vitro DPPH scavenging assay and total phenolic content determination assay deciphered promising antioxidant assays with remarkably low IC50 values of 0.353 and 0.485 μg/mL, respectively for EC‐ESF and EC‐ASF. Besides, different in vivo tests, including tail emersion, acetic acid‐induced writhing, and thiopental‐induced sleeping test of EC‐CME, yielded a remarkable 8.61±0.29 min of tail immersion time compared to the control's 2.05±0.11 min at the highest dose (600 mg/kg). The best % inhibition of writhing was recorded as 47.96 % accrued in 400 mg/kg dose, indicating robust pain‐relieving properties. The onset and duration of sleep are significantly ameliorated for EC‐CME, unveiling its antidepressant potential. Besides, molecular docking studies along with ADME/T analysis also validated the wet lab findings as well as their safety, efficacy, and drug‐likeliness profile. Finally, this work can be an essential hint for utilizing aquatic weeds in drug development research.

Focused mutagenesis in non-catalytic cavity for improving catalytic efficiency of 3-ketosteroid-Δ1-dehydrogenase

Non-catalytic sites are commonly present in many enzymes, but how to classify and modulate such sites for activity improvement is not well-explored.Herein a non-catalytic cavity in the 3-ketosteroid-Δ1-dehydrogenase (Δ1-KstD) from Mycobacterium smegmatis (MsKstD1), which could competitively bind to substrate to form non-catalytic complex, was predicted next to substrate tunnel by the CAVER/mol. dynamics.Based on that, a rational design was performed by Focused Site-directed Iterative Saturation Mutagenesis (FSISM) for blocking the non-catalytic cavity to enhance substrate enter active site to improve the Δ1-dehydrogenation activity of MsKstD1.We obtained the best quadruple mutant (H132M\L113F\V419W\M51L) with 10-fold higher specific activity toward hydrocortisone than that of wild type enzyme.The Δ1-dehydrogenation space time yield toward hydrocortisone reached 36.0 g/L/h to produce prednisolone.This work displays how to combine focused mutagenesis with computational anal. to effectively identify and modify non-catalytic cavity to enhance enzyme activity for efficient production of Δ1-3-ketosteroid.

Metabolic Profiling of Serum for Osteoarthritis Biomarkers

Osteoarthritis is a prevalent aging disease in the world, and in recent years it has shown a trend toward younger age, which is becoming a major health problem in the world and seriously endangers the health of the elderly. However, the etiology and pathogenesis of osteoarthritis are still unclear, causing great trouble for treatment. To screen out candidate biomarkers that could be used for the identification of osteoarthritis and explore the pathogenesis of osteoarthritis, we performed an untargeted metabolomics analysis of nine New Zealand rabbit serum samples by LC-MS/MS, including three normal serum samples (control group) and six osteoarthritis serum samples (case group). Finally, 44 differential metabolites were identified, and the ROC analysis results indicated that a total of 36 differential metabolites could be used as candidate biomarkers. Further metabolic pathway enrichment analysis was performed on these differential metabolites, and we found that a total of 17 metabolic pathways were affected, which may provide directions for the study of osteoarthritis mechanisms.
