The goal of this clinical trial is to learn more about using phenazopyridine (Pyridium) for pain control during bladder onabotulinumtoxinA ("botox") injections for surgical management of overactive bladder (OAB) for patients at Montefiore Medical Center in The Bronx. It will also learn about if using the phenazopyridine pill affects how satisfied patients are with their experience, how much pain patients feel afterwards, and if doctors think using the Pyridium pill affected how they performed the procedure. The main question it aims to answer is:
- Is oral phenazopyridine at least as good as intravesical lidocaine is for managing pain with bladder botox injections for OAB in Montefiore patients?
Researchers will compare phenazopyridine to intravesical lidocaine, a liquid put into the bladder, to see if phenazopyridine is at least as good at controlling pain with bladder botox injections as intravesical lidocaine is.
Participants will be assigned, by chance, to receive either the oral phenazopyridine pill or the intravesical lidocaine as their pain control for the procedure.

Management of Lower Urinary Tract Symptoms during the Treatment for Non-Muscle Invasive Bladder Cancer

PURPOSE OF REVIEWThis narrative review aims to report upon the existing treatment evidence and strategies for managing lower urinary tract symptoms (LUTS) during treatment, including transurethral resection and intravesical therapy. This review also attempts to examine novel approaches to mitigate treatment-related lower urinary tract symptoms and improve treatment adherence.RECENT FINDINGSThere is sparse but promising evidence in improving LUTS secondary to intravesical therapy. Oral agents including phenazopyridine and hyaluronic acid, Bacillus Calmette-Guerin dose reduction, and emerging therapies including beta-3 agonists as well as Onabotulinumtoxin A injections all have demonstrated encouraging improvement in LUTS in limited research. Although recent literature explores new medications and potential strategies for managing intravesical therapy-related LUTS, further research is required to establish efficacy and new consensus on treatment strategies. Further research is also required to establish effective LUTS mitigation strategies with other emerging intravesical therapy regimens.

Discovery and optimization of phenazopyridine hydrochloride as novel SARS-CoV-2 RdRp inhibitors

The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is the pathogen of coronavirus disease (COVID-19) causing a pandemic with growing global transmission. The viral RNA-dependent RNA polymerase (RdRp) is conserved especially for variants of concern (VOCs), making it as an effective antivirals target. Due to the proofreading activity of coronavirus nsp14/nsp10, limited the efficacy of nucleoside analogs in vivo. Herein, we identified that Phenazopyridine hydrochloride (PAP) inhibits SARS-CoV-2 with EC₅₀ of 5.37 μmol/L. Furthermore, PAP can effectively inhibit SARS-CoV-2 RdRp with EC₅₀ value of 7.37 μmol/L, after further optimization, compound PAP-22 exhibits the most potential inhibition, with EC₅₀ of 1.11 μmol/L. PAP and its derivatives can bind directly to SARS-CoV-2 RdRp, fully resistance to the exoribonuclease (ExoN) and exhibit broad spectrum anti-CoV activities. Combined with the current data available on the safe and pharmacokinetics of PAP as an approved drug in clinical use, these results provide a path for the urgently needed antivirals to combat SARS-CoV-2.

Enhanced sonophotocatalytic degradation of phenazopyridine using mesoporous nickel oxide nanoparticles synthesized by a solvothermal method in deep eutectic solvent

NiO nano-photocatalysts have demonstrated good photocatalytic performance for the degradation of wastewater pollutants.The purpose of this study is to investigate the effect of Deep Eutectic Solvent (DES) on the structure, morphol., and photocatalytic activity of synthesized nanoparticles.Herein, mesoporous NiO nanoparticles (NPs) were synthesized in the presence of DES by a solvothermal method.The synthesized nanostructures were characterized using XRD (X-ray diffraction), BET (Brunauer-Emmett-Teller), and HRTEM/TEM (High-Resolution Transmission Electron Microscopy) to evaluate their crystallinity, size, composition, and morphol.The anal. shows that the NiO NPs synthesized in DES have high porosity and sp. surface area which results in better photocatalytic performance for the degradation of organic pollutants.NiO NPs can remove 90%, 81%, and 0% of organic model pollutant phenazopyridine (PhP) during sonophotocatalytic, photocatalytic, and sonocatalytic processes, resp.The simultaneous use of ultrasonic radiation, UV lamp, and prepared catalyst in the sonophotocatalytic reaction had a pos. synergistic effect of about 28.36%.The mechanism of the sonophotocatalytic process was studied through the addition of scavengers and enhancers.Scavengers like formic acid (14%), isopropanol (15%), and sodium bicarbonate (76%) significantly declined removal efficiency.Conversely, the presence of potassium persulfate (97%) as an enhancer improved degradation efficiency by 7%.The degradation of PhP exhibits pseudo-first-order kinetics across all experiments (R² > 0.97).The reusability test shows a negligible decline in sonophotocatalytic performance after four consecutive uses.This feature improves the application of nano-photocatalysts in the industrial field, like wastewater treatment.
