Design and synthesis of azole derivatives of echinocystic acid as α-glucosidase inhibitors with hypoglycemic activity

Diabetes mellitus necessitates strict control of postprandial hyperglycemia via α-glucosidase inhibitors. In this study, novel azole derivatives of Echinocystic acid (EA), a natural pentacyclic triterpenoid, were synthesized through molecular hybridization to enhance hypoglycemic potential. Compound A4 exhibited superior α-glucosidase inhibition (IC₅₀ = 2.72 μM) than that of EA (IC₅₀ = 59.91 μM), and acarbose (IC₅₀ = 342.0 μM). Kinetic analysis revealed mixed-type inhibition (Ki = 15.31 μM, Kis = 10.23 μM), suggesting ternary complex formation. Analysis by spectroscopic studies confirmed A4 altered the enzyme's microenvironment and secondary structure. The combination of molecular docking and molecular dynamics simulations further elucidated the interaction between A4 and α-glucosidase. A4 enhanced binding stability through π-cation and π-π stacking interactions, with low RMSD values indicating structural stability. In vivo studies showed that A4 has an excellent safety profile, with no organ damage observed at a single dose of 0.5 g/kg. In a sucrose loading test in normal mice, A4 demonstrated glucose control comparable to that of acarbose at the same dose. Diabetic mice treated with A4 exhibited reduced fasting blood glucose, improved glucose tolerance, lipid normalization, and antioxidant effects, underscoring its therapeutic promise.

Combination of citrus peel-derived essential oils with acarbose to inhibit amylolytic enzymes – A potential type II diabetes treatment approach

Type 2 diabetes (T2D) can be managed by inhibiting amylolytic enzymes, α-amylase and α-glucosidase, reducing the impact of dietary carbohydrates on blood glucose elevation. Acarbose, a current α-glucose inhibitor (AGI), has excessive α-amylase inhibition, resulting in side effects associated with large amounts of undigested starch being fermented in the colon. This study evaluated the AGI efficacy of citrus peel-derived essential oils, where they were first tested in silico against the target amylolytic enzymes, and then their AGI activity was tested in vitro. The synergistic effects of the essential oils with acarbose against amylolytic enzymes were also determined. In silico and in vitro data of the efficacy of the essential oils as AGIs correlated positively; lower Ki values correlated with more negative binding affinity. Furthermore, molecular dynamic simulations of the most potent compounds were evaluated and indicated relative flexibility and stability induced upon ligand interactions with the protein. The standard AGI drug, acarbose, had the lowest Ki (0.10 ± 0.01 mg/mL) and more negative binding affinity (-7.5 kcal/mol) than the essential oils for α-glucosidase; however, the essential oils only showed potent inhibition against α-glucosidase, with the most potent essential oils being valencene (Ki = 0.33 ± 0.04 mg/mL), carveol (Ki = 0.53 ± 0.02 mg/mL) and geraniol (Ki = 0.56 ± 0.02 mg/mL). The essential oils and acarbose displayed competitive inhibition of α-glucosidase. Furthermore, a combination of acarbose with carveol or geraniol at a ratio of 12.5 μg/mL: 2 mg/mL exhibited antagonistic (CI > 10) and synergistic (CI < 0.7) effects on α-amylase and α-glucosidase inhibition, respectively. Carveol or geraniol can be considered as potentially therapeutic in managing T2D, as it may display lowered AGI-associated side effects.

Studies on the inhibition mechanism of α-glucosidase by kaempferide: Enzyme kinetic, multi-spectroscopy and molecular docking techniques

α-Glucosidase (α-Glu) is an enzyme that lowers postprandial blood glucose after breaking down complex carbohydrates. Kaempferide is the principal flavonoid active ingredient in plants and is widely found in fruits, vegetables, and beverages. This study found that kaempferide has the potential to inhibit α-Glu activity to treat type 2 diabetes. The results showed that kaempferide (IC₅₀ = 55.35 ± 0.27 μM), serving as a mixed-type inhibitor for α-Glu, exhibited sensibly superior inhibition of α-Glu than acarbose (IC₅₀ = 414.08 ± 10.73 μM). In addition, the outcomes from fluorescence quenching, 3D fluorescence, synchronous fluorescence, CD spectroscopy, and molecular docking analysis showed that kaempferide can not only chelate with α-Glu by hydrogen bonding and Van der Waals forces, but also affect the secondary structure and activity of the enzyme. After oral administration of sucrose in mice, kaempferide effectively reduces postprandial blood glucose (PBG) and without any other adverse symptoms. In summary, this study has the potential to contribute to the development of functional foods for the prevention and management of type 2 diabetes (T2DM).

BioDuro-Sundia plants its flag at former Par manufacturing site in CA

A former Par Pharma site in Irvine, CA, will now have a new tenant. BioDuro-Sundia will acquire a 40,000-square-foot oral solid dose manufacturing site in Irvine, the company announced Monday. The site will allow the company to scale up and cater to companies that need larger-scale production, Kent Payne, the president of development and manufacturing, said in a press release. The new site should be operational by the start of 2022. It will be able to produce 1 billion tablets and capsules a year, the company said. “BioDuro-Sundia is a pioneer in enhancing bioavailability, with 25 years of experience applying stabilized amorphous solid dispersion technology to poorly soluble drug product candidates,” he said. “Our clients have requested our support to provide commercial scale manufacturing to save time and maximize success, and it is a pleasure to take this next step with them.” BioDuro-Sundia is a CDMO with 2,000 employees in 10 sites across the country, including China and the US. The company specializes in small- and large-molecule discovery, IND submission support and the scaling of batches. It bills itself as a one-stop shop for its clients. The site is located at 72 Fairbanks in Irvine and was previously used by Par Pharmaceuticals. They acquired the site from Anchen Pharmaceuticals back in 2011, when they acquired the company. In 2020, BioDuro invested in a 150,000-square-foot R&D site in Wuxi City, China, and says that the integrated system can help save time and produce a better quality drug. In April, the company invested in adding a cGMP manufacturing site at the facility. That location handles oral dosage tablets and capsules. That added to its already-existing site in San Diego. Among the drugs manufactured by the company is Gliclazide, a sustained release tablet to treat diabetes from Tianjin Jun’an Bio. The drug is often used with metformin tablets, acarbose tablets or pioglitazone.
