The protective effects of Da Chai Hu Granules (DCHKL) on islet cells which were incubated with 4 mmol x L(-1) alloxan (AXN) were studied. The viability of islet cells were measured with MTT. Insulin released into medium and in islets was detected by radioimmunoassay. Cell apoptosis rate was determined by flow cytometry. The expression of anti-apoptotic gene Bcl-2 and pro-apoptotic gene Bax in islet cells were measured with RT-PCR (reverse transcription polymerase chain reaction). Serum containing DCHKL can promote the activity of islet cells significantly (P < 0.01). Basal insulin secretion and high glucose-stimulated insulin secretion increased significantly (P < 0.01). Serum containing DCHKL can inhibit apoptosis of islet cells, the ratio of apoptosis was decreased. Serum containing DCHKL increased expression of Bcl-2 mRNA and decreased expression of Bax mRNA. DCHKL can significantly promote proliferation of islet cells and increase the amount of basal secretion of pancreatic islet cells and high glucose-stimulated insulin secretion. The expression of Bcl-2 increased significantly. The expression of Bax decreased significantly. DCHKL have a protective effect on the islet cells.

A quality control method for the determination of related substances in 5-fluorocytosine API was established.The related substances in 5-fluorocytosine API were determined by HPLC.The specificity, precision, accuracy, linearity range and robustness of the method were validated.The linear relationship was good in the range of 0.02 ∼ 0.12 μg/mL with the correlation coefficient greater than 0.999, the limit of detection was 0.007 0 μg/mL, the limit of quantification was 0.02 μg/mL, the average recovery was 97.76%∼97.87%, the RSD of repeatability was 0.02%, and the results of robustness test were within the specified limits.The method can effectively determine the related substances in fluorocytosine API.

The objective of this paper is to develop a method for the determination of oleanolic acid and ursolic acid in different batches of Fructus ligustri lucid.A RP-HPLC method was developed for the determination of oleanolic acid and ursolic acid by using Kromasil RP C₁₈ (250 mm × 4.6 mm × 5 μm) column, a mixture of 0.5% trifluoroacetic acid acetonitrile (10:90) as mobile phase at the flow rate of 0.8 mL/min and detective wavelengths of 210 nm.The content of oleanolic acid was ranged from 6.39 to 13.40 mg/g, and ursolic acid was ranged from 1.68 to 3.77 mg/g among the 10 patches of tested samples.The established method is simple, stable, and repeatable, and can be applied for the quality control of the crude Fructus ligustri lucidi and compound prescriptions contained it.
