Multicenter, Randomized, Double Blind, Placebo-Controlled Phase 2a Study, to Evaluate Safety, Tolerability and Pharmacokinetics of Resveratrol (JOTROLTM) in Parkinson's Disease

This is an exploratory phase 2a study to investigate two doses of resveratrol (JotrolTM) vs placebo in individuals diagnosed with Parkinson's Disease (PD). Participants (n=30) will be randomized 1:1:1 into 3 groups and will receive oral JotrolTM vs placebo. Study drug will be titrated to reach a final maximal dose per group and will be administered orally once daily (QD) for 3 months. The study will primarily evaluate the safety, tolerability, pharmacokinetics, and pharmacodynamics (PK/PD) of JotrolTM.

Effectiveness of Resveratrol Addition in Improving Endothelial Dysfunction and Metabolic Control in Type 2 Diabetes Mellitus Patients: A Randomized Controlled Trial

Type 2 diabetes mellitus is associated with endothelial dysfunction, which contributes to increased cardiovascular risk. Chronic hyperglycemia induces oxidative stress and reduces nitric oxide bioavailability, leading to impaired endothelial function. Resveratrol, a natural polyphenol with antioxidant and anti-inflammatory properties, has been shown to improve endothelial function in experimental studies.
This randomized controlled trial aims to evaluate the effectiveness of resveratrol supplementation in improving endothelial dysfunction and metabolic control in patients with type 2 diabetes mellitus. Endothelial function will be assessed using flow-mediated dilatation (FMD) of the brachial artery and serum endothelial nitric oxide synthase (eNOS) levels. Metabolic parameters including fasting blood glucose, postprandial glucose, HbA1c, and lipid profile will also be evaluated.

Dental plaque is a biofilm that accumulates on tooth surfaces and is a primary factor in the development of gingival inflammation. While toothbrushing remains the main method for plaque control, the effectiveness of mechanical oral hygiene depends largely on individual compliance and technique. As a result, mouthwashes are commonly used as adjunctive chemical plaque control agents.
Chlorhexidine gluconate is an effective anti-plaque mouthwash, but its use is associated with undesirable effects that may limit patient acceptance. Resveratrol is a naturally derived compound with reported antimicrobial and anti-biofilm properties, suggesting its potential use as an alternative chemical plaque control agent.
This randomized, double-blind, placebo-controlled, three-period crossover clinical trial aims to evaluate the short-term anti-plaque effect of a resveratrol-based mouthwash by measuring supragingival plaque regrowth over a 24-hour period in periodontally healthy individuals. The effects of the resveratrol-based mouthwash will be compared with 0.12% chlorhexidine (CHX) and a placebo mouthwash under standardized conditions without mechanical oral hygiene

Epigenetic regulators polyphenols in neurodegenerative diseases: a promising intervention strategy

BACKGROUND:

Neurodegenerative diseases are complex disorders characterized by the progressive loss of neuronal structure and function, involving pathological mechanisms such as oxidative stress, chronic inflammation, protein misfolding, and impaired synaptic plasticity. Recent studies have revealed that epigenetic regulation plays a critical role in the onset and progression of these diseases, including mechanisms such as DNA methylation, histone modifications, and non-coding RNA regulation. Natural polyphenolic compounds, known for their safety and multi-target properties, have emerged as promising candidates for neuroprotection and therapeutic intervention.

METHODS:

This review summarizes recent advances in the neuroprotective effects of polyphenols in neurodegenerative diseases through epigenetic mechanisms. It focuses on their regulation of DNA methyltransferase activity, histone acetylation status, and non-coding RNA expression, as well as their influence on neurotrophic factors, inflammatory mediators, and synapse-related gene expression.

RESULTS:

Polyphenolic compounds such as curcumin, resveratrol, and epigallocatechin gallate (EGCG) have been shown to modulate epigenetic enzyme activity, alleviate neuroinflammation, improve mitochondrial function, and promote neuroregeneration, demonstrating multi-level neuroprotective effects. However, clinical translation is limited by low oral bioavailability, rapid metabolism, and insufficient brain-targeting capacity. Emerging strategies including nano-delivery systems, prodrug design, and omics technologies hold promise in overcoming these limitations and enhancing therapeutic efficacy.

CONCLUSIONS:

Polyphenols exert multi-target regulatory effects via epigenetic mechanisms in neurodegenerative diseases, showing great potential for intervention. In the future, integrating precision nutrition, personalized treatment, and multi-omics approaches may facilitate the development of novel epigenetic-based strategies for the prevention and treatment of neurodegenerative disorders.

Resveratrol inhibits renal ischemia and reperfusion injury in diabetes via reducing oxidative stress, inflammation, and apoptosis

OBJECTIVES:

The diabetic kidney is susceptible to renal ischemia/reperfusion (I/R) injury, which is associated with enhanced oxidative stress, inflammatory responses, and cell apoptosis. Resveratrol (RSV) plays a crucial role in protecting various organs against ischemia/reperfusion damage by exerting antioxidant, anti-inflammatory, and anti-apoptotic effects. However, the molecular mechanism of RSV protection against renal I/R injury in diabetic conditions remains unclear. The aim of the present study was to examine whether RSV attenuates renal I/R injury in diabetes and further clarify the underlying mechanisms.

METHODS:

Streptozotocin-induced diabetic rats and high glucose-cultured HK-2 cells were, respectively, treated with RSV before renal ischemia and hypoxia/reoxygenation induction. In vitro, cell viability, intracellular reactive oxygen species levels, and apoptosis were measured. In vivo, renal function, histology, oxidative stress level, inflammatory cytokines, and apoptosis were determined. Furthermore, the expression levels of B-cell lymphoma-2 (Bcl-2), Bcl-2-associated X protein (Bax), cleaved caspase-3, nuclear factor-erythroid 2-related factor 2 (Nrf2), heme oxygenase-1 (HO-1), toll-like receptor 4 (TLR4), and nuclear factor-κB (NF-κB) were detected.

RESULTS:

RSV improved renal function, reduced oxidative stress, inhibited inflammatory responses, and decreased apoptosis after renal ischemia/reperfusion in diabetes, accompanied by increased expression of Nrf2 and HO-1, and decreased expression of TLR4 and NF-κB. The beneficial effects of RSV were abolished by selective inhibition of Nrf2 with ML385.

CONCLUSION:

These findings indicate that RSV could attenuate renal I/R injury in diabetes, possibly by enhancing Nrf2/HO-1 signaling and suppressing the TLR4/NF-κB pathway.

Resveratrol and dexamethasone have cell-specific effects on the circadian clock but not on the rhythm of mitochondrial function in the fetal heart

Abstract:

Proper development of the heart during the fetal period is a prerequisite for health in later life. It was shown that maternal supplementation with stilbenoid resveratrol (RES) is beneficial for cardiovascular function. Synthetic glucocorticoid dexamethasone (DEX) is commonly used in the prenatal treatment of respiratory distress syndrome. RES affects the circadian clock in various tissues but its effect on the fetal heart has not been studied. We hypothetized that RES may affect the the circadian clock via modulation of the glucocorticoid signaling and/or mitochondrial function. Therefore, we tested the effects of different concentrations of RES and synthetic glucocorticoid dexamethasone (DEX) on the circadian clock, energy metabolism and mitochondrial function in fetal cardiomyocytes (CMCs) and cardiac fibroblasts (FBs). We found that RES affects the clock in both CMCs and FBs by increasing the stability of the clock protein PER2. In CMCs, the effect was mediated via the adenylyl cyclase signaling pathway. RES modulated DEX-induced effects on the circadian clock in CMCs and FBs. We were able to detect a circadian rhythm in mitochondrial function in fetal heart cells, which was confirmed by ATP and resazurin assays as well as visualization of the mitochondrial network and reactive oxygen species (ROS). Interestingly, we found that both drugs shifted the phase of fetal heart clock, but had no effect on the phase of mitochondrial rhythmicity, indicating a possible uncoupling of circadian and mitochondrial rhythms in fetal CMCs and FBs. Overall, our data revealed fetal heart-specific effects of RES on the circadian clock through the stabilization of PER2 protein and its ability to modulate DEX-induced effects on the clock.Graphical Abstract
