A retrospective analysis of medical records at Far Eastern Memorial Hospital from January 2008 to May 2020 about female patients with overactive bladder syndrome, who received Solifenacin or Mirabegron as the initial treatment. Factors affecting persistence of OAB medications will be analyzed.

Solifenacin promotes remyelination in cuprizone mouse model by inhibiting the Wnt/β-catenin signaling pathway

Demyelinating diseases are a type of neurological disorder characterized by the damage to the myelin sheath in the central nervous system. Promoting the proliferation and differentiation of oligodendrocyte precursor cells (OPCs) is crucial for treatment. Non-selective muscarinic receptor (MR) antagonists have been shown to improve remyelination in rodent models, although the mechanisms are still unclear. In this study, we treated cuprizone (CPZ)-induced demyelination mouse model with different concentrations of Solifenacin (Sol), a selective M3 receptor antagonist, to determine the optimal concentration for promoting remyelination. Behavioral tests and Luxol fast blue (LFB) staining were used to observe the extent of remyelination, while immunofluorescence was used to measure the expression levels of myelin-related proteins, including myelin basic protein (MBP) and platelet-derived growth factor receptor alpha (PDGFR-α). Western blot analysis was employed to analyze the expression levels of molecules associated with the Wnt/β-catenin signaling pathway. The results showed that Sol treatment significantly promoted myelin regeneration and OPCs differentiation in CPZ-induced demyelination mouse model. Additionally, Sol treatment inhibited the Wnt/β-catenin signaling pathway and reversed the effects of CPZ on OPCs differentiation. In conclusion, Sol may promote the differentiation of OPCs by inhibiting the Wnt/β-catenin signaling pathway, making it a potential therapeutic option for central nervous system demyelinating diseases.

Separation and simultaneous estimation of enantiomers and Diastereomers of muscarinic receptor antagonist Solifenacin using stability‐indicating Normal‐phase HPLC technique with chiral stationary phase amylose tris‐(3,5‐dimethylphenylcarbamate)

Abstract:

The R,S‐enantiomer impurity and diastereomer impurities (S,S‐isomer and R,R‐isomer) of the solifenacin (S,R‐enantiomer) were effectively separated and quantified simultaneously utilizing normal‐phase high‐performance liquid chromatography with a chiral stationary phase consisting of amylose tris (3,5‐dimethylphenylcarbamate) coated on silica‐gel (Chiralpak, AD‐H). The enantiomeric and stereo‐selective separation was achieved within a run time of 35 minutes using a mobile phase of ‘n‐hexane, ethanol, and diethylamine’ in an isocratic elution mode with a detection wavelength of 220 nm. The validation attributes assessed were accuracy (which showed excellent recoveries between 97.5% and 100.4%) and linearity (which was proven in the range of 0.081–1.275 μg.mL−1, with a linear regression of 0.999). The stress testing experiments proved that the developed methodology by the HPLC technique has stability‐indicating characteristics, as all closely eluting peak pairs were separated well with a resolution of 2.3 and without any interference. The proposed methodology was highly efficient in separating and simultaneously determining the chiral impurities (enantiomers and diastereomers) of the solifenacin in the release and stability sample analyses of drug substances and tablets in pharmaceutical formulations.

Fluorimetric study on a novel FDA-approved combination used for the treatment of overactive bladder syndrome in different matrices

For the first time, two spectrofluorimetric techniques were created for the concurrent determination of mirabegron (MBN) and solifenacin (SOL) in different matrices. This novel combination recently received FDA approval to treat overactive bladder syndrome and was marketed under the brand name Mirago^TMS₂₅. SOL has a native fluorescence and can be estimated directly in the presence of MBN without any interference at 290 nm after excitation at 230 nm. In contrast to SOL, MBN is a weak fluorescent substance that requires the use of a fluorogenic reagent (like NBD-Cl) to be measured simultaneously with SOL in their mixtures. So, we reacted MBN with NBD-Cl to yield a yellow-colored fluorophore that could be estimated at 546 nm after excitation at 471 nm without any preliminary separation or interference from SOL. All experimental conditions of the applied methods were tested and enhanced for the best results. The applied fluorimetric methods succeeded in determining both drugs in nanograms, making them applicable in biological fluids like human plasma. The linearity ranges of SOL and MBN were found to be 5-250 ng/mL and 50-600 ng/mL, respectively. The LOD values were 0.96 ng/mL and 5.09 ng/mL for SOL and MBN, in order. Excellent results were attained for both drugs during their analysis in spiked plasma samples employing the applied fluorimetric methods, confirming their suitability for use in real human plasma samples and paving the way for further bioequivalence studies on both drugs.
