Sharing the same photoexcitation process, fluorescence (FL) and photoelectrochemistry (PEC) accomplish optical-optical and optical-electrochemical switches respectively, and are both widely used in diverse fields. However, since FL and PEC are mutually exclusive in principle, it is difficult to obtain intense FL and PEC responses in a system, limiting FL-PEC dual-mode applications. In this study, Ag₂S quantum dots (QDs) and In(OH)₃ nanoblocks (NBs) are simultaneously synthesized by BSA-templated bio-mineralization, and the excited state electrons of Ag₂S/In(OH)₃@BSA nanocomposites (NCs) are further regulated by a proteolysis process. Owing to the coating of BSA, excited state electrons primarily return to ground state via radiation transition, resulting in a robust FL signal. When BSA is hydrolyzed by trypsin, the excited state electrons preferentially transfer to the electron acceptors, and the exposed Ag₂S/In(OH)₃@BSA NCs generate a strong PEC signal. Consequently, heparin, acting as an inhibitor of trypsin, can regulate the FL-PEC switch, allowing for the quantification of heparin by both the decrease in FL signals and the increase in PEC signals. Therefore, a dual-mode biosensor is established to detect heparin using FL and PEC signals, with linear ranges of 0.75-750 U mL^-1 and 3.75 × 10^-3 to 37.5 U mL^-1, and limits of detection of 0.68 U mL^-1 and 4.97 × 10^-4 U mL^-1, respectively. Additionally, the biosensor is used to monitor the rapid metabolism of heparin within 20 h in rats. Overall, this work demonstrates that manipulating excited state electrons of nanomaterials through a biological process offers an alternative perspective for designing FL-PEC dual-mode sensing systems.

Periovulatory anticoagulant therapy enhances embryo recovery rates in superovulated mares

Although protocols for superovulation have been described in horses, this technique has been discouraged due to the low embryo recovery rates in superovulated mares. The reason for these poor results is poorly understood, but the formation of a blood clot in the ovulation fossa following ovulations has been hypothesized. Therefore, this study aimed to assess the safety and effect of periovulatory anticoagulant therapy on embryo recovery of superovulated mares. In experiment 1, five mares were assigned to receive five anticoagulant treatments in a crossover design: intravenous injections of 150 (H1), 300 (H2), 400 (H3), 450 (H4), 600 (H5) IU/kg of unfractionated heparin (UFH, heparin sodium); and had blood samples sequentially collected for up to 48 h post-treatment to test Prothrombin (PT) and activated partial thromboplastin time (aPTT). In experiment 2, four mares were treated in a crossover design with intravenous injection of 450 IU/kg of UFH and 1 mg/kg of low molecular weight heparin (LMWH, enoxaparin) and had blood collected as previously for analysis of plasma anti-Xa activity. In experiment 3, eleven mares had four cycles randomly assigned to four groups. In the control group, mares did not receive any treatment. In contrast, in groups G1, G2, and G3, mares were superovulated with equine pituitary extract and treated 34 h after the induction of ovulation with a placebo (NaCl 0.9 %, G1), 450 IU/kg of UFH (G2), or 1 mg/kg of LMWH. Mares in all groups had ovulation induced with hCG plus histrelin acetate and were bred with fresh semen from one stallion. Embryo flushing was performed nine days post-ovulation. In experiment 1, only mares in groups H4 and H5 had increased aPTT and PT for up to 12 h, and in all groups, aPTT and PT values returned to baselines at 24 h post-treatment. In experiment 2, plasma anti-Xa activity was increased by both therapies for up to 12 h after treatment and was at baseline levels 24 h post-treatment. In experiment 3, periovulatory therapy with anticoagulants increased embryo recovery rates per cycle (G2, 250 %; G3, 260 %) compared to control-assigned cycles (60 %; P < 0.05), whereas G1-assigned cycles (160 %) had intermediate embryo recovery. In conclusion, periovulatory anticoagulant therapies may be an alternative to improve embryo recovery in superovulated mares.

In clinical settings, biofluid-contacting devices can suffer from biofouling, leading to thrombus formation and bacterial biofilm buildup, which impair device function and pose health risks. Traditional antifouling methods, including the use of hydrophilic polymers and heparin coatings, often suffer from instability and reduced bioactivity over time. Lubricant-infused surfaces (LIS) have emerged as a promising alternative due to their long-term stability and broad-spectrum repellency. However, current LIS technologies typically involve complex, multistep processes that restrict their application to surface layers, potentially compromising performance under mechanical stress. This study introduces a novel method for bulk modification of poly(vinyl alcohol) (PVA) films, creating flexible lubricant-infused PVA membranes with superior antifouling properties. These films are fabricated by cross-linking the PVA chains using n-propyltrichlorosilane (n-PTCS) and subsequent infusion with silicone oil as a lubricant. The modified PVA films significantly prevent bacterial adhesion and prolong blood and plasma clot formation. Additionally, these films exhibit enhanced mechanical properties, particularly in elasticity and flexibility compared to unmodified PVA films. The developed technique provides a straightforward method for creating flexible, super-repellent biointerfaces with the potential to prevent blood adhesion and bacterial biofilm formation, which are common complications associated with biofluid-contacting devices and medical implants.

A 2019 tacrolimus guidance document concludes that Bayesian estimation may be a better way to improve future tacrolimus TDM. Bone marrow transplant – Dose personalization improves patient outcomes for many drugs with a narrow therapeutic index and high inter-individuality variability, including busulfan. Operating revenue in the first nine months of 2024 was 564.6 million euros, a 5% decrease on the first nine months of 2023, mainly due to the performance of the contract development and manufacturing business ("CDMO"). This division generated (i) lower revenues from the manufacture of the COVID-19 vaccine in comparison to the first nine months of 2023, when ROVI had booked higher encone related to the production of the "pandemic" COVID-19 vaccine; and (ii) lower revenues related to the activities carried out to prepare the plant for production of the vaccine under the agreement with Moderna. However, CDMO sales increased by 16% to 134.4 million euros in the third quarter of 2024 compared to the third quarter of 2023, as a result of (i) higher revenue from existing customers (excluding Moderna); and (ii) higher revenue related to production of the COVID-19 vaccine due to a more concentrated seasonality in the third quarter. It should likewise be noted that the third quarter of 2024 is expected to be the strongest of the year in terms of CDMO sales performance.
