Delving into the Latest Updates on Ofloxacin with Synapse

Antibiotic residues in milk affect the quality and safety of dairy products, which are crucial to the health of consumers and the sustainable development of the dairy industry. This study utilizes fluorescence hyperspectral imaging (F-HSI) technology in combination with machine learning algorithms for the rapid, nondestructive detection of quinolone antibiotics (ciprofloxacin, ofloxacin) and chloramphenicol antibiotics in milk. The results demonstrated that the MSC-whole-band-SVM and MSC-RF-SVM combination classification models performed the best, with accuracies of 100% for the calibration set and 97.5610% for the prediction set. The original spectrum-SPA+2D-COS-PLSR algorithm model was the best quantitative model for ciprofloxacin residue samples, with an R2 P of 0.88167 and an RMSEP of 0.030637. The MSC-RF + 2D-COS algorithm model was the best quantitative model for ofloxacin residue samples, with an R2 P of 0.89832 and an RMSEP of 0.030531. The SNV-RF + 2D-COS algorithm model was the best quantitative model for chloramphenicol residue samples, with an R2 P of 0.8755 and an RMSEP of 0.036625. These results indicate that F-HSI combined with machine learning can effectively identify and predict the type and concentration of antibiotic residues in milk, and further evaluation under regulatory standards and practical conditions may enhance understanding of its applicability.

Fluoroquinolones (FQs) antibiotic residues threaten ecosystems and human health, necessitating rapid on-site monitoring tools. We synthesized a bimetallic Fe-Cu nanozyme using a Fe-MIL-100 self-sacrificing template via hydrothermal treatment and calcination. This nanozyme exhibited FQs-enhanced peroxidase-like activity, enabling a smartphone-based colorimetric platform for detecting enrofloxacin (ENR), ciprofloxacin (CIP), norfloxacin (NOR), and ofloxacin (OFL). The detection relies on antibiotic-enhanced catalytic oxidation of 3,3',5,5'-tetramethylbenzidine (TMB), producing color changes quantified by smartphone RGB (red-green-blue) analysis. The quantitative performance of the method was evaluated for four target FQs. It exhibited wide linear ranges with limits of quantification (LOQs): for ENR, 0.1-100 μg mL^-1 with an LOQ of 0.1 μg mL^-1; for CIP, 5.0-200 μg mL^-1 with an LOQ of 5.0 μg mL^-1; for NOR, 1.0-125.0 μg mL^-1 with an LOQ of 1.0 μg mL^-1; and for OFL, 1.0-200.0 μg mL^-1 with an LOQ of 1.0 μg mL^-1. Results correlated well with microplate reader data and demonstrated reliability in natural water and meat samples. Mechanistically, the nanozyme exhibits dramatically enhanced catalytic efficiency, with K_m value for H₂O₂ (0.566 mM), which is 6.54-fold lower than that of horseradish peroxidase (HRP). Furthermore, ENR acts as an efficient activity booster, reducing the K_m for H₂O₂ to 0.336 mM (11-fold lower than that of HRP) and increasing V_m values for H₂O₂ and TMB (3.95 × 10 ^-7 M s ^-1, 4.87 × 10 ^-7 M s ^-1, respectively) being 4.54-and 4.87-fold greater than those of HRP. This work elucidates the antibiotic-nanozyme synergistic mechanism and provides a portable, cost-effective sensing strategy for environmental and food safety applications.

High prevalence and co-existence of SHV and CTX-M genes in extensively drug-resistant (XDR) Gram-negative bacteria isolated from hospital settings in Faisalabad District, Pakistan

BACKGROUND: Hospital-acquired infections (HAIs) associated with increasing antimicrobial resistance (AMR) are becoming a global health challenge, causing AMR spread and a serious health threat. This study was designed to evaluate the phenotypic and genotypic status of AMR in Gram-negative HAI-associated bacteria, to gain a current insight into the local settings. METHODS AND RESULTS: Kirby-Bauer method and Double Disc Synergy Test were used for phenotypic assessment of AMR and extended-spectrum β-lactamase (ESBL) producers, respectively. Genes encoding antimicrobial resistance, ESBLs, and carbapenemases were identified by PCR followed by Sanger sequencing. Among 102 HAI samples, 62 were Gram-negative, identified as Escherichia coli (E. coli) (42.2%), Klebsiella pneumoniae (K. pneumoniae) (21.1%), and Pseudomonas aeruginosa (P. aeruginosa) (6.6%) by PCR. All K. pneumoniae and (95%) E. coli isolates exhibited multidrug resistance. K. pneumoniae isolates (95%) showed resistance against ceftazidime, cefotaxime, ceftriaxone, and aztreonam, and (89%) isolates were resistant to amoxicillin-clavulanic acid, ofloxacin, and cefepime (E. coli isolates (84%), (84%), (78%), (76%), (76%), and (73%) were resistant against cefotaxime, ceftriaxone, aztreonam, ceftazidime, doripenem, and colistin, respectively. Genotypically, the mcr-2 gene was detected in 5% K. pneumoniae isolates. Phenotypically, 63% isolates were identified as ESBL producers with high genetic prevalence of SHV-12 (78.9%) and SHV-1 (57.8%) in K. pneumoniae and CTX-M1 (60.5%) in E. coli. Notably, one K. pneumoniae isolate harbored all class-A ESBL genes, which is a novel finding. Co-existence of CTX-M1 + SHV-1 + TEM-1, SHV-1 + SHV-12 + TEM-1, and CTX-M1 + CTX-M15 + TEM-1 genes was dominant. CONCLUSION: The HAIs caused by extensively drug-resistant and ESBL-producing K. pneumoniae and E. coli were dominant, with less prevalence of moderately drug-resistant Pseudomonas isolates.

DCA officials seize illegal drugs worth over 15k from quack clinic

Hyderabad: Officials from the Drugs Control Administration (DCA) raided a quack's clinic in Karimnagar district and seized illegal drugs and steroids worth ₹15,011. During the raid, the officials detected 39 types of medicine, including 35 types of physician samples of antibiotics , analgesics and anti-ulcer drugs, which were being stored on the premises without a valid licence for the sale of drugs. The seized stock included antibiotics such as cefixime, ofloxacin, cloxacillin, amoxycillin and azithromycin. Officials warned that the indiscriminate sale of antibiotics by unqualified individuals could have serious consequences for public health, including the development of antimicrobial resistance (AMR). Officers took samples of the seized drugs for laboratory analysis. They added that a further investigation is underway and that legal action will be initiated against all those found responsible. "Stocking and selling drugs without a licence is a punishable offence under the Drugs and Cosmetics Act, which carries a penalty of up to 5 years' imprisonment," said a drug inspector.

Samples of drugs used in diabetes, hypertension, allergies, infections found substandard in quality tests

Jaipur: Samples of five drugs were found to be of substandard quality upon testing and analysis conducted by the Commissionerate, Food Safety and Drug Control (CFSDC). On Wednesday, the CFSDC issued directions to all the district drug controllers in the state to ensure that the stocks of these drugs are no longer consumed. They were instructed to provide appropriate safeguards to consumers by withdrawing the available stock and taking action as per the provisions of the Drugs & Cosmetics Act 1940. A senior CFSDC official said, samples of Levocetirizine Dihydrochloride Tablets (Wincet-L), manufactured by M/s YL Pharma of Baddi in Solan district, Himachal Pradesh, were found to be of substandard quality. The drug was sold as an antihistamine used to treat symptoms associated with various allergic conditions. A sample of Telmisartan and Amlodipine Tablets IP (Telpin-A), manufactured by India M/s Maskon Life Sciences Pvt. Ltd in Bhagwanpur of Roorkee, was found to be of substandard quality. The drug is used as a combination medication to treat high blood pressure (hypertension) in adults. The CFSDC tested a sample of Cefixime Oral Suspension manufactured by M/s Agron Remedies Pvt. Ltd of Sarverkhera, Uttarakhand, which failed the quality test. It is used as an antibiotic medication to treat a variety of bacterial infections, particularly in children and adults who have difficulty swallowing pills.>

CDSCO monthly review finds 56 drugs as substandard, flags 3 as spurious

New Delhi: As part of its monthly review undertaken to check and enforce the quality standards in the consumer drug market, the Central Drugs Standard Control Organization ( CDSCO ) has identified 56 drug samples as ‘Not of Standard Quality’ (NSQ) and 3 as ‘spurious.’ The drugs and formulations which have been classified as spurious picked by Bihar Drugs Control Authority include Pantoprazole Gastro-resistant and Domeperiodone Prolong Released Capsules IP ( PAN-D); Cefixime Tablets IP (TAXIM-O 200); and Rosuvastatin Tablets IP (Rosuvas 10). As per the CDSCO notification, each of these drugs were manufactured in 2023 and the central drugs regulator is still investigating the manufacturer of the above-mentioned spurious drugs . The list of drugs and formulations which have been identified as NSQ includes Trypsin & Chymotrypsin Tablets manufactured by Cachet Pharmaceuticals Private Limited; Diclofenac Sodium, Paracetamol & Chlorzoxazone Tablets by MODERN LABORATORIES; Liposomal Amphotericinn B Injection 50mg by UNITED BIOTECH (P) LIMITED; Ornidazole & Ofloxacin Tablets IP by Hindustan Antibiotics Ltd; Amlodipine and Perindopril Tablets by Intas Pharmaceuticals Ltd.; Natamycin Ophthalmic Suspension by Primus Pharmaceuticals, among 50 others. All samples notified under the NSQ list for the month of October (released in November) were manufactured between 2022 and 2024, and CDSCO has identified the manufacturers of each of the listed products. Meanwhile, during the same period the State Drugs Testing Laboratories across the country have identified 34 Drugs Samples as Not of Standard Quality (NSQ). According to CDSCO, a drug shall be deemed spurious if it is manufactured under a name belonging to another drug; or if it is an imitation of, or a substitute for, another drug; or resembles another drug in a manner likely to deceive or bears upon its label or container the name of another drug, among other provisions defined under Section 17-B of the Drugs and Cosmetics Act, 1940.
