This study investigates the time course of change in calcium in the blood after a pregnant subject takes an oral dose of calcium carbonate (commonly marketed as "Tums"). This information is important for trials investigating whether calcium can reduce postpartum hemorrhage, bleeding after delivery.

The Effect of Calcium Carbonate on Labor Induction: A Pilot Study

The investigators aim to evaluate the safety and efficacy of administering calcium carbonate to laboring participants undergoing labor inductions. The investigators hypothesize that calcium carbonate is a low-risk preventative measure to decrease oxytocin induction time and dosage, decrease the rate of labor dystocia, decrease the rate of cesarean deliveries, and demonstrate no differences in maternal or neonatal safety outcomes.

An Open Label, Single Center, Randomized, Two-way Crossover Parallel Group, Trial to Assess the Mechanism of Action of a Bi-layer Swallowable Tablet (Immediate Release and Sustained Release) Containing Calcium Carbonate Antacid in an Overnight and Day Application as Compared With a Standard, Non-layered (Immediate Release) Calcium Carbonate Swallow Tablet in Fed Healthy Male Participants.

Researchers are looking for a better way to treat people who have heartburn, indigestion, and problems due to excessive stomach acid. These are common problems which can affect daily life and disturb sleep during the night-time. Heartburn is the burning sensation or pain in the chest which occurs when stomach acid rises up in the food pipe (esophagus).
Calcium carbonate tablets are used to treat heartburn, indigestion, and related digestive problems. Calcium carbonate works by neutralizing the excess acid in the stomach. The study treatment is a new bi-layer calcium carbonate tablet that has two layers. One layer quickly releases calcium carbonate aimed to provide quick relief (called immediate release) while the other layer releases calcium carbonate slowly to make the relief last longer (called sustained release).
In this study, bi-layer calcium carbonate tablets will be given to healthy men for the first time. This study will provide information on how the new bi-layer tablet works inside the body.
The main purpose of this study is to learn about how the new bi-layer calcium carbonate tablet changes the average acidity levels (measured using pH) compared to the standard calcium carbonate tablet during the night-time.
For this, researchers will measure the acidity levels in the upper part of the stomach at regular intervals during the night-time.
The participants will be randomly (by chance) assigned to one of two treatment groups:
Participants in the first group will take the treatments at night.
Participants in the second group will take the treatments during the day.
All participants in both groups will take 2 bi-layer tablets and 2 standard tablets after a meal with a gap of 6 to 8 days between treatments. However, in each group, half the participants will receive the bi-layer tablets first while the other half will receive the standard tablets first.
Each participant will be in the study for around 52 days with up to 4 visits to the study site. This includes:
1. visit about 28 days before the treatment starts during which the doctors will confirm that the participant can take part in the study
2. visits for treatment with a gap of 6-8 days between each treatment, and
1 visit 7 to 14 days after the treatment ends during which the doctors will monitor the participants' health.
During the study, the doctors and their study team will:
check participants' overall health by performing tests such as blood and urine tests, and check heart health using an electrocardiogram (ECG)
take images of the stomach at different times after taking the treatment
measure acidity level (pH) using a device called pH probe that is inserted into the upper part of the stomach
ask the participants questions about how easy it is to take the study treatment
ask the participants what adverse events they are having.
An adverse event is any medical problem that a participant has during a study. Doctors keep track of all adverse events, irrespective of whether they think it is related to the study treatment, or not.
As this study is conducted in healthy men who will not gain any benefit from this treatment, access to the study treatment after the study is not planned.

Effect of monobutyrate on growth performance, intestinal development, and cecum microbiota in broilers

The prohibition of antibiotic growth promoters for animal production has created a demand to explore alternative feed additives to replace antibiotics.Monobutyrate has the potential to replace antibiotics.In this study, we investigated the effects of monobutyrate on broiler growth performance, intestinal development, and caecum microbiota.Total 240 1d-old Arbor Acres broilers were randomly divided into five groups.Each group had eight broilers, and the groups were replicated six times.The broilers were fed a basal diet (CK group), a basal diet containing 0.1% acetyl kitasamycin (Anti group), and basal diets containing 0.15‰, 0.30‰, and 0.45‰ monobutyrate (M1, M2 and M3 group) for 42 days.Compared with the CK group, F/G was significantly decreased in broilers in the M3 group (p <0.05).The relative length and weight of the duodenum were significantly higher in the M1, M2, and M3 groups than those in the CK group.Compared with the CK group, the relative ileum length was significantly increased in the M2 and M3 groups (p <0.05).Compared with those of the CK and Anti groups, Rikenellaceae, Alistipes, Sellimonas, and Ruminococcus_2 were enriched in the M3 group (p < 0.05).The results of this study indicated that monobutyrate promotes broiler growth performance, improves intestinal development, and regulates microbiota composition

AHP-TOPSIS-based analysis for the wettability of agro-industrial waste particulates Bhimal fiber-reinforced epoxy bio-composites in different oil water environments

The work studied the use of agro-industrial untreated waste for bio-composites as a sustainable material and determined its wettability in diverse environments.To prepare the bio-composites, Bhimal natural fiber was compositely reinforced with CaCO₃ composite platelets and Flyash along with TiO₂ at different concentrations (5%, 10%, and15% by weight) into an epoxy matrix.The samples were tested in petrol, diesel; kerosene, seawater Ganga water; rainwater, and distilled water at 168 h and the saturation was stabilized after measuring every 48 h.The absorption resistance, especially in oil-based environments was excellent in TiO₂ 15%-based bio-composites.Materials were ranked by the Analytic Hierarchy Process (AHP) and TOPSIS methods.The results show the potential of TiO₂ composites for low wettability applications without any chems. as well, which may encourage green materials from agro-industrial waste.

Effect of carbonated recycled aggregates on the mechanical properties and bonding performance of polyurethane mortar as a repair material

Abstract:

Recycled aggregate (RA) made from construction waste has limited application due to its high porosity, numerous cracks, and other disadvantages such as reduced concrete strength when using aggregate). In this paper, carbonated recycled aggregate (CRA) was obtained by carbonating recycled aggregate, and natural aggregate (NA) in polyurethane mortar (PUM) was replaced at different ratios. The physical changes and pore structure of RA before and after carbonation, the mechanical properties (compressive and flexural strength) of PUM, and the three-point bending bond performance were systematically investigated. The compactness of PUM was detected by ultrasonic velocity (UPV) test. The change mechanism of PUM microstructure was revealed by scanning electron microscopy (SEM). The results show that when the replacement rate is 50%, the compressive and flexural strength of carbonated PUM increased by 7.32% and 15.83%, respectively, and bond strength increased by 14.19%, all of which are greater than the increase at a replacement rate of 25%. The mechanical properties of P with RA as aggregate decreased, but the generated calcium carbonate after carbonation treatment can fill the surface pores and enhance the pore structure of CRA, and the mechanical properties of three-point bending bond performance of the test pieces with the same replacement ratio were improved compared with those replaced by RA.

Many MSME pharma firms making substandard drugs, CDSCO data shows

New Delhi: Many pharmaceutical companies in the micro, small and medium enterprise (MSME) sector are manufacturing substandard drugs , according to the data by India's drug regulator, the Central Drugs Standard Control Organisation ( CDSCO ). According to the data for April, most of these companies are making drugs that failed safety standards and have been found to be sub- standard quality. As part of the continuous regulatory surveillance , drug samples are picked from sales and distribution points and analysed at various government run laboratories. According to the drug regulatory authority, in April about 60 samples failed to meet the quality standards and have been declared as Not of Standard Quality (NSQ) Drugs . Samples which failed tests are carboxymethylcellulose sodium eye drops, an eye lubricant, also known as artificial tears, bupivacaine hydrochloride injection, a local anaesthetic, calcium carbonate and vitamin D3 tablets, among others. Interestingly, they have been manufactured at lesser known pharma companies like Gidsha Pharmaceuticals in Gujarat, Martin and Brown Biosciences in Solan, Himachal Pradesh, Sai Parenterals in Hyderabad, Nestor Pharmaceuticals, Faridabad, among others. These companies could not be contacted immediately. "This has been the trend for some time now. Most of these MSME are found to be manufacturing drugs which are not of standard quality. That is the reason the health ministry came out with a notification which outlines standards for the pharma companies," said a person with knowledge of the matter. Earlier in 2023, over 65 per cent of MSME enterprises were found to be manufacturing drugs that were not of standard quality (NSQ) during risk-based inspections (RBI) of pharma companies. "Of the total MSME units inspected, 30 per cent were issued stop production orders (SPO), while samples of 68% MSME companies have failed. This is alarming," the person said. The CDSCO and state drug inspectors have been conducting these inspections as part of a massive nationwide crackdown on spurious and substandard medicines. This followed a controversy over alleged Made-in-India cough syrups being responsible for the deaths of children in The Gambia. The CDSCO has found that these companies have issues related to infrastructure, quality management systems and product manufacturing process knowledge. "There has been a strong correlation between NSQ and the condition of manufacturing set up. Monthly NSQ data shows repeat failures for a company with the same product with multiple companies month on month," another person said. India's drug regulator had earlier pointed to an urgent need to review the current GMP regulations and Quality Management Systems being followed by pharmaceutical companies during the RBI. Six common medications, including antibiotics and allergy drugs, fail quality tests

Jaipur: The health department released a list of six drugs on Monday that failed quality tests and are being sold in the market. These include commonly used antibiotics such as amoxicillin and clavulanate potassium, anti-allergic medicines like levocetirizine hydrochloride and montelukast, the gastro-resistant drug pantoprazole, and cefpodoxime, which is an antibiotic drug. These drugs failed the quality test as they did not contain any of the promised medicinal content. The drugs were manufactured in Bengaluru, Punjab, Sikkim, Mohali, and Himachal Pradesh. A sample of the medicine Pregabalin, which is in a class of medications called anticonvulsants, failed the quality test as it was found to have nil content of pregabalin. The drug was manufactured by a company in Mohali. A health department official said that directions have been issued to drug control officers in districts to remain alert and take necessary action. They have been directed to collect samples of other medicines manufactured by the companies whose samples failed the quality test. The samples of amoxicillin and clavulanate potassium manufactured by a Bengaluru-based firm failed as they contained none of the promised drugs. Earlier, a report was issued by the health department in which samples of drugs in combination with nimesulide and paracetamol (pain relief), pheniramine maleate injection (to treat symptoms of allergic conditions), a combination of calcium carbonate and vitamin D3 suspension (dietary supplement for people who have bone problems or low calcium and vitamin D levels), a combination of telmisartan, amlodipine, and hydrochlorothiazide (to treat high blood pressure), a combination of telmisartan and amlodipine (for high blood pressure), a combination of levocetirizine dihydrochloride and montelukast (relieves sneezing and runny nose due to allergies), heparin sodium injection (anticoagulant), and an injection of sulphamethoxazole and trimethoprim (treats infections caused by bacteria) failed quality tests. These drugs were manufactured in Baddi, Sirmour and Solan (Himachal Pradesh), Vadodara (Gujarat), Roorkee (Uttarakhand), and Digiana (Jammu).

After 2023 Snub, Amneal Clinches Approval for Extended-Release Parkinson's Disease Med Crexont

After a major setback in 2023, Amax Pharmaceuticals finally ushered in a brilliant victory in the United States, its signature innovative delayed release Parkinson's disease treatment drug - Crexont, successfully obtained the authoritative approval of the United States Food and Drug Administration (FDA). This unique oral calcium carbonate/levodopa (CD/LD) extended-release formulation opens a new chapter in the treatment of patients with Parkinson's disease with its superior efficacy and convenience, not only significantly extending their "well" period, but also reducing the number of burdensome daily doses to three times, greatly improving patients' quality of life. The FDA accepted Crexont's application after a thorough review and analysis of the detailed Phase III clinical trial data submitted by Amax. In this critical clinical trial of RISE-PD, Crexont stood out for its exceptional performance, providing patients with an additional 30 precious minutes of "good" time per day compared to traditional ready-to-release CD/LD. In addition, its average daily frequency of taking only three times, compared with five times for traditional drugs, undoubtedly brings unprecedented convenience to patients. The successful approval of Crexont is undoubtedly a significant milestone in the treatment of Parkinson's disease. In the face of the large population of patients in the United States - about 1 million people with Parkinson's disease, and about 90,000 new cases each year, the introduction of Crexont has undoubtedly brought unprecedented hope for patients. The co-ceos of Amax Pharmaceuticals, Chirag Patel and Chintu Patel, could not contain their excitement, saying that it is their greatest honor to bring this epoch-making new treatment to Parkinson's disease patients in the United States. At the same time, the company is also confidently planning to bring this innovative treatment to the international market. However, behind the success is often accompanied by unremitting efforts and challenges. Amax had previously been rejected by the FDA because of insufficient safety data on Crexont. This setback did not break them, but inspired them to be more determined and more rigorous. Now, with the successful approval of Crexont, the drug is expected to generate annual sales of up to $300 million to $500 million, further solidifying its leading position in the treatment of Parkinson's disease. It's worth noting that Amax is not alone. Abbott and Newrod Pharmaceuticals, among others, have faced similar regulatory challenges on the road to developing new levodopa preparations. These experiences show that despite the difficulties ahead, pharmaceutical companies are still working hard to find better treatments for Parkinson's disease. Amax's success has undoubtedly set an example for the entire industry, and has brought more treatment options and light of hope to patients with Parkinson's disease.
