Persistent daytime symptoms of sleepiness in individuals with obstructive sleep apnea (OSA) who are using Continuous Positive Airway Pressure (CPAP) are associated with adverse long term medical and functional outcomes. Supplementary exposure to bright light has beneficial effects on sleep quality and daytime vigilance in healthy individuals and it has been increasingly applied in a variety of sleep and neuropsychiatric conditions. This study will explore the role of Bright Light Therapy (BLT), a well-established non-pharmacological intervention for circadian disturbances, for the treatment of residual daytime symptoms of OSA which do not respond to CPAP. BLT will be delivered via therapy glasses in a cross-over design, where each participant will be exposed to active treatment and sham treatment (4 weeks in each arm) in a randomized order. The hypothesis is that participants will demonstrate improvements in the variables of interest during the four-week active treatment portion of the eight-week crossover study, compared to the four-week sham treatment portion.

Determination of Sedative and Anesthetic Drug Residues in Aquatic Food Products Using Solid Phase Extraction (SPE) and Liquid Chromatography–Tandem Mass Spectrometry (LC–MS/MS)

The residues of sedative and anesthetic drugs in aquatic products are an important factor affecting food safety.In order to screen the residues of sedative and anesthesia drugs in aquatic products, a rapid and sensitive method was established using solid phase extraction (SPE) and high-performance liquid chromatog.-tandem mass spectrometry (HPLC-MS/MS).The samples were extracted by 80% aqueous acetonitrile by an alumina column and separated by a C18 column.The mass spectra were determined in pos. ion mode.The standard curves were matrix matched and quantified by the external standard method.The results showed that the linear ranges of the drugs in aquatic products were from 0.5 to 100 μg kg-1.The correlation coefficient (R2) was greater than 0.990 and the detection limits were between 0.001 and 0.322 μg kg-1.The average recovery rate and relative standard deviation were between 78.64% and 112.15% and from 0.41%-8.26%.Overall, the developed method showed promising results for the simultaneous qual. and quant. determination of the drug residues in aquatic products.

Synthesis, antitumoral activity, and in silico studies on Smoothened receptor of new 2,6,9-trisubstituted purine derivatives

In this work, a series of 30 new 2,6,9-trisubstituted purine derivatives were synthesized and evaluated in silico as potential ligands of the Smoothened (SMO) receptor, as well as their ability to inhibit growth in Hedgehog (Hh)-dependent and Hh-independent cancer cell lines.The synthesis involved a convergent strategy, conventional methods and microwave irradiationInitial antitumor evaluation was performed by testing cell growth inhibition in seven cancer cell lines and one non-neoplastic cell line (HEK-293) at 50 μM.IC₅₀ values were determined for compounds showing < 50 % cell viability.Compounds 7l and 9j showed promising results with high cytotoxicity in three Hh-dependent cell lines and low cytotoxicity in HEK-293 cells.Compound 7l was more potent and selective than gemcitabine in BxPC-3, AsPc-1 and MIA-PaCa-2 cells and more than 5-fluorouracil in HT-29 cells, while 9j was more potent and selective than 5-fluorouracil in HCT116 and HT-29 cells.Mol. docking studies in SMO allowed the recognition of two binding sites related to ligand size and purine substitution patterns, while 7l bound to the top pocket (TMD-1), 9j bound to a deeper pocket (TMD-2).This study provides new evidence supporting the purine ring as a privileged scaffold for the development of new antitumor drugs targeting the SMO receptor.

Abstract:

Various series of 4,6‐disubstituted‐2‐thiopyridine derivatives were synthesized and evaluated as potential ecto‐5’‐nucleotidase (CD73) inhibitors. Altogether, about ninety compounds were prepared using a general synthetic pathway involving one or two steps (eventually one‐pot) procedures. Variation of the nature of the substituents in positions 4 and 6 (methyl, trifluoromethyl or phenyl) of the thiopurine ring, as well as on the thiol function, was examined and led to marked differences both in term of reactivity and ability to interfere with the putative target protein. Using a functional assay on immune cells, few compounds belonging to series 4 were shown to be able to antagonize the inhibition of the T‐cell proliferation at both 100 μM and 10 μM (completely for 4 ab and partially for 4 ai), that is as potent as AOPCP which entirely reversed the inhibitory impact of exogenous ATP on T cell proliferation until 62.5 μM. In addition, we have shown that both compounds (4 ab and 4 ai) were also capable of moderately inhibiting the hA2A receptor with Ki in the μmolar range in HEK‐293 cells. Thus, with the aim to reduce the molecular size and the lipophilicity of our initial scaffold, we finally observed by serendipity a modification of the potential target of our compounds.

Alliance Pharma agrees to £362m acquisition by DBAY

UK-based Alliance Pharma has agreed to a final increased cash offer from asset management company DBAY Advisors, valuing the former at approximately £362m ($467.5m). The offer, made through a newly formed company called Aegres Bidco Limited, will see DBAY acquire all shares in Alliance, except those it already owns through its affiliates, for 64.75p per share. This represents a 3.6% increase from DBAY’s initial offer of 62.5p per share in January 2025 and a 46% premium on Alliance’s share price as of 9 January 2025, the day before the offer period began. Alliance Pharma specialises in the marketing and distribution of consumer healthcare products and prescription medicines. Its portfolio includes Kelo-Cote, a scar treatment; and Anbesol (benzocaine), an oral pain relief product. DBAY initially approached Alliance last year, following a series of challenges faced by the company, including the delayed publication of its annual results and the resignation of its CEO Peter Butterfield. Having gradually increased its stake in the company over two years, DBAY now holds a 27.9% share of Alliance. In its initial takeover offer, DBAY stated that it believed Alliance had “strong future prospects” but required significant operational changes and increased investment to maximise its growth potential. The asset management company argued that such changes would be more effectively implemented away from the pressures of the public market. In January 2025, Alliance’s board of directors recommended shareholders accept the initial offer of 62.5p per share. However, after receiving feedback from shareholders, the scheduled general meeting last month was adjourned to allow for further discussions. According to a 10 March filing on the London Stock Exchange, DBAY will fund the acquisition through a combination of equity investments from its affiliated funds and external financing, with a small portion of shares in Midco, the parent company of Bidco, being offered to certain lenders. Shareholders are expected to vote on the proposal at a rescheduled meeting on 13 March 2025. If approved, the deal is anticipated to be completed in the first half of 2025, resulting in Alliance Pharma becoming a privately owned company under DBAY’s control. In September 2024, Alliance reported underlying revenue of £84m for the six months to 30 June 2024, reflecting an increase from £81.4m in the same period the previous year. Profit before tax also rose year-on-year, reaching £12.7m compared to £10.3m in 2023.

Insight Pharmaceuticals Issues Voluntary Nationwide Recall of Americaine® 20% Benzocaine Topical Anesthetic Spray Due to the Presence of Benzene

TARRYTOWN, N.Y., Dec. 22, 2023 (GLOBE NEWSWIRE) -- Insight Pharmaceuticals, a Prestige Consumer Healthcare Inc. company (“Insight”), is voluntarily recalling one lot of Americaine® 20% Benzocaine Topical Anesthetic Spray to the consumer level. While benzene is not an ingredient in any Americaine® 20% Benzocaine Topical Anesthetic Spray products, a recent review by our manufacturer and their third-party lab found that a sample from one lot of the product showed a low level of benzene came from the propellant that sprays the product out of the can. Importantly, no other lots of Americaine® 20% Benzocaine Topical Anesthetic Spray (either before or after this batch code) are in the scope of this recall and may continue to be used by consumers safely and as intended. Risk Statement: Benzene is classified as a human carcinogen. Exposure to benzene can occur by inhalation, orally, and through the skin and it potentially can result in cancers including leukemia and blood cancer of the bone marrow and blood disorders which can be life threatening. Insight is recalling these products out of an abundance of caution. To date, the Company has not received any serious adverse events related to this recall. Benzene is ubiquitous in the environment. Humans around the world have daily exposures to it indoors and outdoors from multiple sources. Americaine® 20% Benzocaine Topical Anesthetic Spray is packaged in white aerosol cans with Teal caps with Lot codes located on the bottom of the can. Samples of the recalled lot below have been found to contain trace levels of Benzene related to the propellant that sprays the product out of the can.
